Summary A gene encoding superoxide dismutase (EC 1.15.1.1., SOD) was isolated from a plasmid library of chromosomal DNA from Listeria ivanovii by functional complementation of an SOD-negative Escherichia coli host. The nucleotide sequence of the cloned gene was determined and contained an open reading frame which codes for a protein of 202 amino acid residues (calculated molecular weight 22 755 Da including the amino-terminal methionine residue). Comparison of the deduced amino acid sequence of L. ivanovii SOD with previously reported SOD amino acid sequences revealed considerable homologies with Fe- and Mn-dependent SODs. Enzymatic analyses using cell lysates and the purified recombinant enzyme indicated that this SOD is manganese-dependent. The recombinant SOD accounted for up to 30% of the total soluble protein in recombinant E. coli and protected sodA sodB mutants against the toxic effects of paraquat. Subunits of the recombinant Listeria SOD and of both E. coli SODS formed enzymatically active hybrids in vivo.Some of our preliminary observations have been published as a conference report of SOD V (Jerusalem, 1989) in Free Rad Res Commun (1991) 12–13:371  相似文献

A method is described for obtaining transgenic plants with a high level of expression of the introduced gene. Tobacco protoplasts were transformed with an expression construct containing a translational fusion between mature -amylase from Bacillus licheniformis and the signal peptide of the tobacco PR-S protein. A total number of 5200 transformed protoplasts was cultured to microcalli and screened for -amylase expression by incubation on media containing starch followed by staining with iodine. The calli were divided into four classes, based on the resulting halo sizes on the plates. The halo sizes were found to correlated with the expression levels in transgenic plants regenerated from the calli. The expression levels varied between 0 and 0.5% of soluble leaf protein in the regenerated transgenic plants. Wider implications of this method are discussed.  相似文献

Macro and Micro cation content, concentration of water soluble cations and anions, as well as nitrogen content in beech leaves from a variety of forest stands in the eastern part of Austria have been determined. Special attention has been focused on the highly air polluted Vienna Woods. A broad natural variability in total content of minerals, as well as in nitrogen content could be observed. In general, cation nourishment has been found to be adequate on all stands. The pattern of water soluble i.e. physiologically active cations, especially with regard to divalent cations, varies broadly among beeches from different provenances: Mg/Mn ratios differ within three orders of magnitude between dolomite and highly acidic, nutrient poor stands over silicate rock. The term mineral deficiency limit should, therefore, be used very carefully with respect to single nutrients. Organic anions (quinate, malate, citrate) clearly dominate over inorganic anions. The relative amount of the latter increased from NO3, Cl, inorganic phosphate to SO4. A concentration of SO4 is markedly in leaves within the direct reach of SO2 sources, its level, however, remaining well below the organic anions. Slight evidence of an antagonistic behavior between SO4 and organic acids could be found. Total nitrogen as well as the ratio insoluble:soluble nitrogen were tendencially higher in Vienna Wood beeches, indicating an additional use of the airborne nitrogen. The high flexibility of beech with respect to mineral content and ionic balance is discussed with regard to the ecological distribution of beech and the susceptibility to air pollution.  相似文献

Unilateral pulmonary artery obstruction (PAO) for 24-48 h, followed by reperfusion, results in pulmonary edema and lung inflammation. We hypothesized that lung injury actually occurred during the period of PAO but, because of low microvascular pressures during the period of occlusion, was not detected until perfusion was reestablished. To test this hypothesis, we studied 14 rabbits divided into three groups: group I rabbits underwent sham occlusion of the left pulmonary artery for 24 h; group II rabbits underwent PAO but were not reperfused; and group III rabbits were subjected to PAO and then reperfused for 4 h. The fluid filtration coefficient measured during a zone 3 no-flow hydrostatic stress (pulmonary arterial pressure = pulmonary venous pressure, both greater than alveolar pressure) in group I lungs was less than that of lungs in either group II or III [0.52 +/- 0.02 (SE) ml.min-1.cmH2O.100 g wet wt-1 vs. 0.94 +/- 0.11 and 0.86 +/- 0.13 for groups II and III, respectively, P less than 0.05]. The wet-to-dry weight ratio of the left lung measured after the zone 3 stress was applied for 20 min was 6.90 +/- 0.09 in group I rabbits and 9.21 +/- 0.75 and 11.75 +/- 0.44 in groups II and III, respectively (P less than 0.05). Radiolabeled microspheres demonstrated that flow to the left lung was diminished after the period of PAO (38 +/- 4, 9 +/- 5, and 2 +/- 1% of cardiac output in groups I, II, and III, respectively; P less than 0.05 for group I vs. groups II and III).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Selection on locomotor performance was determined for a series of marked and recaptured individuals from a population of garter snakes (Thamnophis sirtalis fitchi) in Northern California. We measured snake length and mass, burst speed, endurance on a treadmill, and the distance crawled around a stationary circular track. Size-corrected values (residuals) of mass and locomotor performance were generated from the scaling equations of S–V length (SVL). Randomization tests and regressions were used to determine the probability that a trait was a significant predictor of survivorship, and a nonparametric, cubic spline estimate of the fitness function was used to facilitate detection of the patterns of selection. From 275 (“cohort”) snakes measured and tested within 8 days of birth in 1985, 79 were recaptured in the spring–summer of 1986 and subsequent years. Birth SVL was the only significant (randomization P = 0.022) predictor of neonatal survival from 1985 to 1986 with directional selection favoring larger individuals. In addition to the lab-born cohort, 382 field-born snakes from all ages in the population were captured, tested, and released during spring–summer 1986. Similar to the 1985 cohort, the survivorship of 37 of 86 neonates from 1986 to 1987 showed no significant relationship with any residual value using any statistical test. Survivorship from 1986 to 1987 for 127 of 250 yearlings (including 32 lab-born cohort snakes) analyzed with the randomization test showed that greater values of both speed (P = 0.007) and distance residual (P = 0.008) significantly favored survival, whereas intermediate values of mass residual (P = 0.006) were significantly more likely to survive. Univariate regressions predicting the survival of yearlings from 1986 to 1987 gave similar results to the randomization test, but in a multiple regression with yearling burst speed residual, distance capacity residual, and a quadratic term of mass residual, distance capacity residual was the least important predictor variable. For the survivorship of 37 of the 113 older snakes, greater burst speed residual significantly favored survival (randomization P = 0.001).  相似文献

Cyclic adenosine monophosphate arrests proliferating T lymphocytes in the G1 phase of the cell cycle. Here we demonstrate that exogenous and endogenous elevations in cyclic AMP concentration result in diminished mitogen stimulation, cell cycle arrest, and decreased ribonucleotide reductase messenger RNA concentrations in peripheral blood mononuclear cells. At lower concentrations (less than 1mM) of dibutyryl cyclic AMP that do not generate cell cycle arrest there is inhibition of ribonucleotide reductase activity without decreased messenger RNA concentration for the M2 subunit of ribonucleotide reductase. However, at higher concentrations of dibutyryl cyclic AMP there is G1 cell cycle arrest and reduced M2 specific messenger RNA concentration. Thus, cyclic AMP inhibition of lymphocyte activation may occur by different mechanisms that are dose dependent.  相似文献

Time-resolved fluorescence anisotropy of the sterol analogue, cholestatrienol, and 13C nuclear magnetic resonance (NMR) spin lattice relaxation time (T1c) measurements of [13C4] labeled cholesterol were exploited to determine the correlation times characterizing the major modes of motion of cholesterol in unsonicated phospholipid multilamellar liposomes. Two modes of motion were found to be important: (a) rotational diffusion and (b) time dependence of the orientation of the director for axial diffusion, or "wobble." From the time-resolved fluorescence anisotropy decays of cholestatrienol in egg phosphatidylcholine (PC) bilayers, a value for tau perpendicular, the correlation time for wobble, of 0.9 x 10(-9) s and a value for S perpendicular, the order parameter characterizing the same motion, of 0.45 s were calculated. Both tau perpendicular and S perpendicular were relatively insensitive to temperature and cholesterol content of the membranes. The T1c measurements of [13C4] labeled cholesterol did not provide a quantitative determination of tau parallel, the correlation time for axial diffusion. T1c from the lipid hydrocarbon chains suggested a value for tau perpendicular similar to that for cholesterol. Steady-state anisotropy measurements and time-resolved anisotropy measurements of cholestatrienol were used to probe sterol behavior in a variety of pure and mixed lipid multilamellar liposomes. Both the lipid headgroups and the lipid hydrocarbons chains contributed to the determination of the sterol environment in the membrane, as revealed by these fluorescence measurements. In particular, effects of the phosphatidylethanolamine (PE) headgroup and of multiple unsaturation in the lipid hydrocarbon chains were observed. However, while the steady-state anisotropy was sensitive to these factors, the time-resolved fluorescence analysis indicated that tau perpendicular was not strongly affected by the lipid composition of the membrane. S perpendicular may be increased by the presence of PE. Both steady-state anisotropy measurements and time-resolved anisotropy measurements of cholestatrienol were used to probe sterol behavior in three biological membranes: bovine rod outer segment (ROS) disk membranes, human erythrocyte plasma membranes, and light rabbit muscle sarcoplasmic reticulum membranes. In the ROS disk membranes the value for S perpendicular was marginally higher than in the PC membranes, perhaps reflecting the influence of PE. The dramatic difference noted was in the value for tau perpendicular. In both the ROS disk membranes and the erythrocyte membranes, tau perpendicular was one-third to one-fifth of tau perpendicular in the phospholipid bilayers. This result may reveal an influence of membrane proteins on sterol behavior.  相似文献

Dopaminergic D2 receptors are widely regarded as typical inhibitory receptors, as they both inhibit adenylyl cyclase and decrease the cytosolic free Ca2+ concentration ([Ca2+]i) by activating K+ channels. A D2 receptor has recently been cloned (Bunzow, J. R., Van Tol, H. H. M., Grandy, D. K., Albert, P., Salon, J., Christie, M. D., Machida, C. A., Neve, K. A., and Civelli, O. (1988) Nature 336, 783-787) and expressed in two different cell lines, pituitary GH4C1 cells and Ltk- fibroblasts, where it has been shown to induce inhibition of adenylyl cyclase. We have investigated the additional effector systems coupled to this receptor. The responses observed in the two cells lines, which express similar levels of receptors (0.5-1 x 10(5)/cell), were surprisingly different. In GH4C1 cells D2 receptors failed to affect phosphoinositide hydrolysis and induced a decrease of [Ca2+]i. This latter effect appears to be mediated by hyperpolarization, most likely due to the activation of K+ channels. In striking contrast, in Ltk- fibroblasts the D2 receptor induced a rapid stimulation of inositol(1,4,5)-trisphosphate (+73% at 15 s) followed by the other inositol phosphates, and an immediate increase of [Ca2+]i due to both Ca2+ mobilization from internal stores and influx from the extracellular medium. In both GH4C1 and Ltk- cells, the D2 receptor response was mediated by G protein(s) sensitive to pertussis toxin. The increases of inositol trisphosphate and [Ca2+]i observed in Ltk- cells required dopamine concentrations only slightly higher than those inhibiting adenylyl cyclase (EG50 = 25, 29, and 11 nM, respectively) and were comparable in magnitude to the responses induced by the endogenous stimulatory receptor agonists, thrombin and ATP. The results demonstrate that in certain cells D2 receptors are efficiently coupled to the stimulation of phosphoinositide hydrolysis. The nature of receptor responses appears therefore to depend on the specific properties not only of the receptor molecule but also of the cell type in which it is expressed.  相似文献

Histochemical determinations for storage of carbohydrates in rhizomes, roots, and young shoots of Typha latifolia L. (Typhaceae) were conducted during the overwintering period from November to April. Early winter analysis showed that rhizomes and roots contained large amounts of starch (45.03% and 22.80% dry weight, respectively). The major storage tissue was parenchyma of the rhizome central core. From winter into spring a gradual decrease in storage starch in the rhizome and root occurred concurrently with starch accumulation near zones of rapid development in young shoots (buds), but the rhizome retained much starch (27.40% dry weight) into the start of its 2nd yr.  相似文献
