The mtDNA rnl-U7 region has been examined for the presence of introns in selected species of the genus Ceratocystis. Comparative sequence analysis identified group I and group II introns encoding single and double motif LAGLIDADG open reading frames (ORFs) at the following positions L1671, L1787, and L1923. In addition downstream of the rnl-U7 region group I introns were detected at positions L1971 and L2231, and a group II intron at L2059. A GIY-YIG type ORF was located within one mL1923 LAGLIDADG type ORF and a degenerated GIY-YIG ORF fused to a nad2 gene fragment was found in association with the mL1971 group I intron. The diversity of composite elements that appear to be sporadically distributed among closely related species of Ceratocystis illustrates the potential for homing endonucleases and their associated introns to invade new sites. Phylogenetic analysis showed that single motif LADGLIDADG ORFs related to the mL1923 ORFs have invaded the L1787 group II intron and the L1671 group I intron. Phylogenetic analysis of intron encoded single and double motif LAGLIDADG ORFs also showed that these ORFs transferred four times from group I into group II B1 type introns.  相似文献

The use of glyphosate modifies the environment which stresses the living microorganisms. The aim of the present study was to determine the real impact of glyphosate on potential pathogens and beneficial members of poultry microbiota in vitro. The presented results evidence that the highly pathogenic bacteria as Salmonella Entritidis, Salmonella Gallinarum, Salmonella Typhimurium, Clostridium perfringens and Clostridium botulinum are highly resistant to glyphosate. However, most of beneficial bacteria as Enterococcus faecalis, Enterococcus faecium, Bacillus badius, Bifidobacterium adolescentis and Lactobacillus spp. were found to be moderate to highly susceptible. Also Campylobacter spp. were found to be susceptible to glyphosate. A reduction of beneficial bacteria in the gastrointestinal tract microbiota by ingestion of glyphosate could disturb the normal gut bacterial community. Also, the toxicity of glyphosate to the most prevalent Enterococcus spp. could be a significant predisposing factor that is associated with the increase in C. botulinum-mediated diseases by suppressing the antagonistic effect of these bacteria on clostridia.  相似文献

We have previously reported that angiotensin receptor blockade reduces reperfusion hemorrhage in a suture occlusion model of stroke, despite increasing matrix metalloproteinase (MMP-9) activity. We hypothesized that candesartan will also decrease hemorrhage associated with delayed (6 h) tissue plasminogen activator (tPA) administration after embolic stroke, widening the therapeutic time window of tPA. Adult male Wistar rats were subjected to embolic middle cerebral artery occlusion (eMCAO) and treated with either candesartan (1 mg/kg) alone early at 3 h, delayed tPA (10 mg/kg) alone at 6 h, the combination of candesartan and tPA, or vehicle control. Rats were sacrificed at 24 and 48 h post-eMCAO and brains perfused for evaluation of neurological deficits, cerebral hemorrhage in terms of hemoglobin content, occurrence rate of hemorrhage, infarct size, tissue MMP activity and protein expression. The combination therapy of candesartan and tPA after eMCAO reduced the brain hemorrhage, and improved neurological outcome compared with rats treated with tPA alone. Further, candesartan in combination with tPA increased activity of MMP-9 but decreased MMP-3, nuclear factor kappa-B and tumor necrosis factor-α expression and enhanced activation of endothelial nitric oxide synthase. An activation of MMP-9 alone is insufficient to cause increased hemorrhage in embolic stroke. Combination therapy with acute candesartan plus tPA may be beneficial in ameliorating tPA-induced hemorrhage after embolic stroke.  相似文献

The probiotic E. coli Nissle 1917 (EcN) has been reported to have various health benefits; however, very little is known about their underlying mechanisms. In this regard, the present study aimed to elucidate the effect of the bacterium on mucin production by intestinal epithelial cells. Incubation of HT-29 cells with EcN lead to a contact time-dependent rise in mRNA levels of the MUC2, MUC3, MUC5AC, and MUC5A. The expression was markedly higher with MUC5AC gene. In most cases, MUC genes expression was more pronounced in polarized cells compared to non-polarized ones. In contrast to MUC3, the basal stimulation of polarized cells brought about markedly higher levels of other tested mucins. Similar but milder results were observed when living EcN was replaced by inactivated bacteria. With exception of MUC3, the conditioned media showed no significant effect on the mRNA level of the tested mucins. The above-mentioned mRNA results were confirmed on protein level using enzyme-linked lectin assay (ELLA) and enzyme-linked immunosorbant assay (ELISA). In contrast to other treatments, basal stimulation of polarized cells showed a growth phase-dependent MUC induction with more prominent effect by stationary-phase bacteria. In contrast to MUC 2 and MUC3, the induction of MUC5AC and MUC5B showed a bacterial count-dependent pattern. In conclusion, EcN was found to stimulate MUC gene expression in HT-29 intestinal cells. This stimulation was more distinct with polarized cells. Such observation may partially interpret some health benefits of the probiotic bacterium including antagonizing pathogen adhesion and protection of the intestinal mucosa.  相似文献

Insulin, in nature, has a stimulatory effect on microorganisms. These effects include the acceleration of sugar metabolism, triacylglycerol anabolism, growth rate, and formation of oils. We also observed that insulin may cause indirect activation of triacylglycerol lipase by forcing the cell to permanently require an energy source. Thus, cells can consume all of their accumulated internal fuel sources such as lipids, proteins, and carbohydrates. After studying the effects of using two types of insulin (Humulin 70/30, and human insulin expressed in yeast) at different concentrations on microalgae (Chlorella sp.), we found that with certain concentrations of insulin (1:3.3 ml unit Humulin 70/30 per ml; 1:2.6 ml unit yeast insulin per ml), there was an increase in algal growth rate and decrease in cell size. We therefore studied the effect of insulin under conditions of lipase inhibition by Triton WR 1339 (Tyloxapol), which was used at different concentrations with and without insulin. We found strong regression in the growth rate with increasing Triton concentrations. However, we also observed that the cell size under the effect of Triton and Triton-insulin was larger than the cell size under the effect of insulin alone, and also larger than for control cells. Also, the oil content of the Triton-insulin cells was higher than those of the control cells or the cells under the effect of insulin alone.  相似文献

Summary The adherence of zoospores of Lagenidium giganteum to the cuticle of mosquito larvae is the initial step in the infection process. Subsequently, a germ tube penetrates the integument, inducing a rapid melanization of the injured cuticle and epidermis. After entering the hemocoel the developing hyphae are occasionally encapsulated locally. This process is slow (6 to 12 h postincubation) and most frequently cell-free, although it can be mediated by circulating hemocytes. Sporadic hemocyte mediation of the humoral encapsulation process in larval stages of Culicidae adds a previously unreported dimension to this unusual type of defense reaction. The defense reactions of larvae of Aedes aegypti were ineffective against observed infection by Lagenidium giganteum.  相似文献

Data of 56 normal and 9 abnormal estrous cycles were collected from 9 Egyptian buffaloes (Bublus bublis) to describe the follicular growth wave pattern. Heat was checked twice daily while, ovaries were scanned daily to monitor the patterns of follicular waves. Day of ovulation was determined when the largest follicle was replaced by corpus haemorrhgicum (CH). Number of waves/cycle, day of emergence of the follicular wave, characteristics of the dominant follicle and corpus luteum (CL) growth features were monitored. Buffaloes displayed mainly two types of follicular waves; two (46.4%) and three (53.6%). In cycles of three wave pattern, time of emergence of the 1st wave post-heat was longer (P < 0.05) and number of recruited follicles/wave were larger (P < 0.05) compared to the corresponding values of the two wave pattern. Number of recruited follicles in early follicular waves (1st or the 2nd) had larger number (P < 0.05) compared to the subsequent ones. Follicles that reached ovulation in both types of estrous cycle had shorter life-span (P < 0.05) than the previous ones. Life-span of CH, growing and regressed CL were 3.6 ± 0.6, 11.2 ± 0.8 and 4.4 ± 0.5 days, respectively with no difference in both types of follicular wave. Three types of ovarian disorders were observed. Follicular waves and CL growth features showed unique pattern for each individual. These results demonstrate that buffaloes display two main types of follicular waves with dominance of three wave type.  相似文献

The Burgers equation is obtained to study the characteristics of nonlinear propagation of ionacoustic shock, singular kink, and periodic waves in weakly relativistic plasmas containing relativistic thermal ions, nonextensive distributed electrons, Boltzmann distributed positrons, and kinematic viscosity of ions using the well-known reductive perturbation technique. This equation is solved by employing the (G'/G)-expansion method taking unperturbed positron-to-electron concentration ratio, electron-to-positron temperature ratio, strength of electrons nonextensivity, ion kinematic viscosity, and weakly relativistic streaming factor. The influences of plasma parameters on nonlinear propagation of ion-acoustic shock, periodic, and singular kink waves are displayed graphically and the relevant physical explanations are described. It is found that these parameters extensively modify the shock structures excitation. The obtained results may be useful in understanding the features of small but finite amplitude localized relativistic ion-acoustic shock waves in an unmagnetized plasma system for some astrophysical compact objects and space plasmas.  相似文献
