The current investigation was conducted to elucidate the potential modulatory functions of both enzymatic and non-enzymatic scavenging elements of three Iranian basil (Ocimum basilicum L.) cultivars in response to different water-deficit stress treatments [i.e., control (W1: 100 % FC), mild (W2: 75 % FC), moderate (W3: 50 % FC), and severe (W4: 25 % FC)]. In general, the growth parameters, viz., plant height, number of lateral branches, number of flowers in the inflorescence per plant, and dry and fresh weights of leaves and inflorescence followed by yield were considerably affected by water-deficit stress levels (p ≤ 0.05), though some fluctuations were observed among three cultivars. Under severe water-deficit stress (W4), total chlorophyll content overall increased, while a pronounced reduction in the carotenoid content was observed by boosting of water-deficit stress intensities. Apart from some quantitative variations, ROS-scavenging enzymes, such as SOD, CAT, APX, GPX, and PPO, exhibited different behaviors versus different levels of water-deficit stress in the basil cultivars, concluding that their modulation could be a cultivar-dependent mechanism and stress-dependent mechanism. Among different metabolites detected in the essential oil of basil cultivars, both methyl chavicol and squalene were superior in the cultivars 2 and 3, while in cultivar 1, linalool and squalene were the predominant constituents, under water deprivation conditions. Taking all the features studied here into consideration, presumably, cultivar 1 is qualified enough to nominate as the most tolerant basil cultivar, could be accordingly utilized as a promising source/material for breeding programs of basil under drought stress, and possibly other abiotic stresses.  相似文献

Background

Transposable Elements (TEs) are key components that shape the organization and evolution of genomes. Fungi have developed defense mechanisms against TE invasion such as RIP (Repeat-Induced Point mutation), MIP (Methylation Induced Premeiotically) and Quelling (RNA interference). RIP inactivates repeated sequences by promoting Cytosine to Thymine mutations, whereas MIP only methylates TEs at C residues. Both mechanisms require specific cytosine DNA Methyltransferases (RID1/Masc1) of the Dnmt1 superfamily.

Results

We annotated TE sequences from 10 fungal genomes with different TE content (1-70%). We then used these TE sequences to carry out a genome-wide analysis of C to T mutations biases. Genomes from either Ascomycota or Basidiomycota that were massively invaded by TEs (Blumeria, Melampsora, Puccinia) were characterized by a low frequency of C to T mutation bias (10-20%), whereas other genomes displayed intermediate to high frequencies (25-75%). We identified several dinucleotide signatures at these C to T mutation sites (CpA, CpT, and CpG). Phylogenomic analysis of fungal Dnmt1 MTases revealed a previously unreported association between these dinucleotide signatures and the presence/absence of sub-classes of Dnmt1.

Conclusions

We identified fungal genomes containing large numbers of TEs with many C to T mutations associated with species-specific dinucleotide signatures. This bias suggests that a basic defense mechanism against TE invasion similar to RIP is widespread in fungi, although the efficiency and specificity of this mechanism differs between species. Our analysis revealed that dinucleotide signatures are associated with the presence/absence of specific Dnmt1 subfamilies. In particular, an RID1-dependent RIP mechanism was found only in Ascomycota.

A fundamental advancement in the evolution of complexity is division of labour. This implies a partition of tasks among cells, either spatially through cellular differentiation, or temporally via a circadian rhythm. Cyanobacteria often employ either spatial differentiation or a circadian rhythm in order to separate the chemically incompatible processes of nitrogen fixation and photosynthesis. We present a theoretical framework to assess the advantages in terms of biomass production and population size for three species types: terminally differentiated (heterocystous), circadian, and an idealized species in which nitrogen and carbon fixation occur without biochemical constraints. On the basis of real solar irradiance data at different latitudes, we simulate population dynamics in isolation and in competition for light over a period of 40 years. Our results show that in isolation and regardless of latitude, the biomass of heterocystous cyanobacteria that optimally invest resources is comparable to that of the idealized unconstrained species. Hence, spatial division of labour overcomes biochemical constraints and enhances biomass production. In the circadian case, the strict temporal task separation modelled here hinders high biomass production in comparison with the heterocystous species. However, circadian species are found to be successful in competition for light whenever their resource investment prevents a waste of fixed nitrogen more effectively than do heterocystous species. In addition, we show the existence of a trade-off between population size and biomass accumulation, whereby each species can optimally invest resources to be proficient in biomass production or population growth, but not necessarily both. Finally, the model produces chaotic dynamics for population size, which is relevant to the study of cyanobacterial blooms.  相似文献

BACKGROUND: A number of reports indicate that circulating immune complexes (CIC) and activation of the complement system contribute to the pathogenesis of Type I allergy. The aim of this study was to investigate the status of CIC in 113 patients with rhinitis in Lebanon and determine complement components C3 and C4 serum levels in the CIC-positive patients. Serum specific IgE antibodies were previously detected and reported in 74 of the 113 patients. METHODS: CIC were detected by polyethylene glycol precipitation and serum C3 and C4 levels quantified by radial immunodiffusion. RESULTS: CIC was positive in 20 of the specific IgE-positive and 13 of the specific IgE-negative patients. C3 and C4 levels were within the normal range in all the 33 CIC-positive patients. CONCLUSIONS: The antibody class that constitutes the complexes does not seem to be IgG or IgM. Moreover, complement activation does not seem to be involved in the allergic reaction since both C3 and C4 levels were normal in all patients. The role of these complexes, if any, in the pathogenesis of rhinitis is yet to be determined.  相似文献

Chemical functionalization of a zigzag carbon nanotube (CNT) with 1, 3-cyclohexadiene (CHD), previously reported by experimentalists, has been investigated in the present study using density functional theory in terms of energetic, geometric, and electronic properties. Then, the thermodynamic and kinetic feasibility of H2 dissociation on the pristine and functionalized CNTs have been compared. The dissociation energy of the H2 molecule on the pristine and functionalized CNT has been calculated to be about ?1.00 and ?1.55 eV, while the barrier energy is found to be about 3.70 and 3.51 eV, respectively. Therefore, H2 dissociation is thermodynamically more favorable on the CNT-CHD system than on the pristine tube, while the favorability of the dissociation on the pristine tube is higher in term of kinetics.  相似文献

The roles of gibberellic acid (GA3) and ethylenediaminetetraacetic acid (EDTA) in phytoremediation of cadmium (Cd)-contaminated soil by Parthenium hysterophorus plant was investigated. GA3 (10?9, 10?7, and 10?5M) was applied as a foliar spray. EDTA was added to soil in a single dose (160 mg/kg soil) and split doses (40 mg/kg soil, four split doses). GA3 and EDTA were used separately and in various combinations. P. hysterophorus was selected due to its fast growth and unpalatable nature to herbivores to reduce the entrance of metal into the food chain. The Cd phytoextraction potential of the P. hysterophorus plant was evaluated for the first time. Cd significantly reduced plant growth and dry biomass (DBM). GA3 alone increased the plant growth and biomass in Cd-contaminated soil, whereas EDTA reduced it. GA3 in combination with EDTA significantly increased the growth and biomass. The highest significant DBM was found in treatment T3 (10?5M GA3). All treatments of GA3 or EDTA significantly enhanced the plant Cd uptake and accumulation compared with control (C1). The highest significant root and stem Cd concentrations were found in the combination treatment T11 (GA3 10?5M + EDTA split doses), whereas in leaves it was found in the EDTA treatments. Cd concentration in plant parts increased in the order of stem < leaves < roots. The combination treatment T9 (GA3 10?7M + EDTA split doses) showed the significantly highest total Cd accumulation (8 times greater than control C1, i.e., only Cd used). The GA3 treatments accumulated more than 50% of the total Cd in the roots, whereas the EDTA treatments showed more than 50% in the leaves. Root dry biomass showed a positive and significant correlation with Cd accumulation. GA3 is environment friendly as compared with EDTA. Therefore, further investigation of GA3 is recommended for phytoremediation research for the remediation of metal-contaminated soil.  相似文献
