CD8 T cells are involved in pathogen clearance and infection-induced pathology in respiratory syncytial virus (RSV) infection. Studying bulk responses masks the contribution of individual CD8 T cell subsets to protective immunity and immunopathology. In particular, the roles of subdominant responses that are potentially beneficial to the host are rarely appreciated when the focus is on magnitude instead of quality of response. Here, by evaluating CD8 T cell responses in CB6F1 hybrid mice, in which multiple epitopes are recognized, we found that a numerically subdominant CD8 T cell response against DbM187 epitope of the virus matrix protein expressed high avidity TCR and enhanced signaling pathways associated with CD8 T cell effector functions. Each DbM187 T effector cell lysed more infected targets on a per cell basis than the numerically dominant KdM282 T cells, and controlled virus replication more efficiently with less pulmonary inflammation and illness than the previously well-characterized KdM282 T cell response. Our data suggest that the clinical outcome of viral infections is determined by the integrated functional properties of a variety of responding CD8 T cells, and that the highest magnitude response may not necessarily be the best in terms of benefit to the host. Understanding how to induce highly efficient and functional T cells would inform strategies for designing vaccines intended to provide T cell-mediated immunity. 相似文献

To determine whether or not the secretory granules of insulin-secreting cells contained glycoproteins, isolated rat pancreatic islets were incubated for 2 and 4 hr in a medium containing L-[3H]-fucose. Anthropogenic habitat disturbance is fundamentally altering patterns of disease transmission and immunity across the vertebrate tree of life. Most studies linking anthropogenic habitat change and disease focus on habitat loss and fragmentation, but these processes often lead to a third process that is equally important: habitat split. Defined as spatial separation between the multiple classes of natural habitat that many vertebrate species require to complete their life cycles, habitat split has been linked to population declines in vertebrates, e.g. amphibians breeding in lowland aquatic habitats and overwintering in fragments of upland terrestrial vegetation. Here, we link habitat split to enhanced disease risk in amphibians (i) by reviewing the biotic and abiotic forces shaping elements of immunity and (ii) through a spatially oriented field study focused on tropical frogs. We propose a framework to investigate mechanisms by which habitat split influences disease risk in amphibians, focusing on three broad host factors linked to immunity: (i) composition of symbiotic microbial communities, (ii) immunogenetic variation, and (iii) stress hormone levels. Our review highlights the potential for habitat split to contribute to host-associated microbiome dysbiosis, reductions in immunogenetic repertoire, and chronic stress, that often facilitate pathogenic infections and disease in amphibians and other classes of vertebrates. We highlight that targeted habitat-restoration strategies aiming to connect multiple classes of natural habitats (e.g. terrestrial–freshwater, terrestrial–marine, marine–freshwater) could enhance priming of the vertebrate immune system through repeated low-load exposure to enzootic pathogens and reduced stress-induced immunosuppression. 相似文献

The role of nitric oxide (NO) mediated vasorelaxation in splanchnic blood flow during heat stress was studied in rats before and after heat acclimation (1 month, at 34°C). Superior mesenteric artery and portal vein blood flows were measured on-line in the conscious rats during heat stress at 40°C and 42°C before and after LNNA administration. NO mediated vasorelaxation in these vessels was studies in vitro, in response to pilocarpine, Ca ionophore and Na nitroprusside stimulation. The data suggest that NO is an integral part of the thermoregulatory splanchnic vasomotor response, and that heat acclimation enhances its regulatory importance. 相似文献

Because of a number of major advances in the past one to two decades, there is little doubt that the inherent cellular and membrane properties of neurons in an oscillating network play an important role in shaping the output of that network. There are a number of such examples in vertebrate and invertebrate systems. In this review, we present some of the newer methods that have been used in the identification of membrane properties and detail some cellular studies performed in both vertebrate (locomotion and sleep/waking rhythms) and invertebrate network systems (escape swimming in Tritonia diomedia and pyloric rhythm in Panulirus interruptus). Studies examining the cellular or membrane properties of respiratory neurons have been scarce until recently. The importance of these properties in dictating respiratory rhythm generation and output in the mature and immature animal is not yet known; however, we put this issue in perspective by building a parallel between mammalian respiration and other vertebrate networks that have been better investigated and characterized. 相似文献

Competition for limited resources is considered a key factor controlling invasion success. Resource availability can be viewed in either the long or short‐term. Long‐term availability depends on the baseline nutrient availability in the ecosystem and how those conditions shape the ecological community. Short‐term resource availability fluctuates with disturbances that alter nutrient availability and/or the density and composition of the ecological community. We investigated how species’ traits interact with short and long‐term resource availability to determine the outcome of invasions. We manipulated long‐term baseline resource availability, disturbance intensity, disturbance frequency, and propagule pressure in a fully factorial design using protist microcosms. Our results show that short and long‐term resource availability and the direct mortality from disturbance interact with the traits of resident community members and traits of invaders to determine community invasibility. While competitively dominant invaders with slow growth rates may suffer rather than benefit from short‐term resource fluctuations, quickly growing but competitively inferior invaders can benefit from both the resource fluctuations and the heterogeneity in community composition created by disturbance. Our findings empirically synthesize two explanations for invasion success, namely short‐term resource fluctuations and long‐term resource availability, and highlight the importance of considering traits of invaders and residents, such as growth rate and competitive ability, in the context of productivity and disturbance gradients. This species’ traits approach could resolve idiosyncratic results from natural systems undergoing disturbance and invasion that do not follow patterns predicted by traditional invasion frameworks. 相似文献
