Comparisons of congeneric species have provided unique insights into invasion ecology. Most often, non‐native species are compared to native ones to look for traits predicting invasion success. In this study, we examine a different facet of congeneric comparisons in which both species are non‐native. Ecological variability among non‐native congeners might 1) lead to the inhibition or facilitation of either species’ ability to colonize and spread, 2) result in larger cumulative impacts due to synergies between species, and 3) depend on the physical context of the invaded habitat. To explore these possibilities, we studied the distribution and abundance of two non‐native beach grasses: European beach grass Ammophila arenaria and American beach grass Ammophila breviligulata, their interaction with one another, and their biotic and physical impacts on dune ecosystems of the Pacific coast of North America. We found that over a two‐decade period, A. breviligulata has increased its dominance over A. arenaria on dunes where it was originally planted in 1935 and has actively spread to new sites formerly dominated by A. arenaria. Our results also show that dune plant species richness was lower at A. breviligulata sites, although there was an increase in the native beach grass Elymus mollis. More significantly, we found that the two grass species are associated with significantly different foredune shapes that are likely controlled by a combination of variability in sand supply along the coast and subtle differences in the congeners’ morphology and growth form. These differences have significant implications for the coastal protection services of dunes to humans and the conservation of native species. They provide a cautionary tale on the impacts of introducing novel species based purely on analogy with closely related species.  相似文献

Tumor necrosis factor alpha (TNFalpha) is a potent pleiotropic cytokine produced by many cells in response to inflammatory stress. The molecular mechanisms responsible for the multiple biological activities of TNFalpha are due to its ability to activate multiple signal transduction pathways, including nuclear factor kappaB (NFkappaB), which plays critical roles in cell proliferation and survival. TNFalpha displays both apoptotic and antiapoptotic properties, depending on the nature of the stimulus and the activation status of certain signaling pathways. Here we show that TNFalpha can lead to the induction of NFkappaB signaling with a concomitant increase in spermidine/spermine N(1)-acetyltransferase (SSAT) expression in A549 and H157 non-small cell lung cancer cells. Induction of SSAT, a stress-inducible gene that encodes a rate-limiting polyamine catabolic enzyme, leads to lower intracellular polyamine contents and has been associated with decreased cell growth and increased apoptosis. Stable overexpression of a mutant, dominant negative IkappaBalpha protein led to the suppression of SSAT induction by TNFalpha in these cells, thereby substantiating a role of NFkappaB in the induction of SSAT by TNFalpha. SSAT promoter deletion constructs led to the identification of three potential NFkappaB response elements in the SSAT gene. Electromobility shift assays, chromatin immunoprecipitation experiments and mutational studies confirmed that two of the three NFkappaB response elements play an important role in the regulation of SSAT in response to TNFalpha. The results of these studies indicate that a common mediator of inflammation can lead to the induction of SSAT expression by activating the NFkappaB signaling pathway in non-small cell lung cancer cells.  相似文献

Phosphatidylcholine (PC, lecithin) has long been considered a solely eukaryotic membrane lipid. Only a minority of all bacteria is able to synthesize PC. The plant‐transforming bacterium Agrobacterium tumefaciens encodes two potential PC forming enzymes, a phospholipid N‐methyltransferase (PmtA) and a PC synthase (Pcs). We show that PC biosynthesis and tumour formation on Kalanchoë plants was impaired in the double mutant. The virulence defect was due to a complete lack of the type IV secretion machinery in the Agrobacterium PC mutant. Our results strongly suggest that PC in bacterial membranes is an important determinant for the establishment of host–microbe interactions.  相似文献

Abstract: We present the first record of the cardiid genus Monodacna from the Pliocene of Anatolia, Turkey. Monodacna imrei sp. nov. was found in the Pliocene Killik Formation from the western margin of the Baklan Basin, in very marginal brackish to freshwater lacustrine deposits. The new record extends the stratigraphic range of the modern Ponto‐Caspian genus back into the Pliocene and adds to earlier evidence that modern Ponto‐Caspian taxa originated in the Pliocene of south‐western Turkey.  相似文献

Quaternary climatic fluctuations have left contrasting historical footprints on the neutral genetic diversity patterns of existing populations of different tree species. We should expect the demography, and consequently the neutral genetic structure, of taxa less tolerant to particular climatic extremes to be more sensitive to long‐term climate fluctuations. We explore this hypothesis here by sampling all six pine species found in the Iberian Peninsula (2464 individuals, 105 populations), using a common set of chloroplast microsatellite markers, and by looking at the association between neutral genetic diversity and species‐specific climatic requirements. We found large variation in neutral genetic diversity and structure among Iberian pines, with cold‐enduring mountain species (Pinus uncinata, P. sylvestris and P. nigra) showing substantially greater diversity than thermophilous taxa (P. pinea and P. halepensis). Within species, we observed a significant positive correlation between population genetic diversity and summer precipitation for some of the mountain pines. The observed pattern is consistent with the hypotheses that: (i) more thermophilous species have been subjected to stronger demographic fluctuations in the past, as a consequence of their maladaptation to recurrent glacial cold stages; and (ii) altitudinal migrations have allowed the maintenance of large effective population sizes and genetic variation in cold‐tolerant species, especially in more humid regions. In the light of these results and hypotheses, we discuss some potential genetic consequences of impending climate change.  相似文献

ABSTRACT. The marine scuticociliate Paratetrahymena parawassi n. sp. is described on the basis of morphology, especially infraciliature, and the sequence of its small subunit (SSU) rRNA gene to become the second known member of its genus. Paratetrahymena and other ciliates in the order Loxocephalida possess a mixture of morphological and morphogenetic features characteristic of the subclasses Hymenostomatia and Scuticociliatia. Accordingly, we used SSU rRNA sequences to analyze the phylogeny of Paratetrahymena and three other loxocephalid genera. Paratetrahymena and Cardiostomatella vermiformis formed a moderately well‐supported clade that diverged at a deep level from all other scuticociliates, supporting separation of loxocephalids from other scuticociliates as a suprafamilial taxon. Sathrophilus holtae was a sister taxon to Paratetrahymena and Cardiostomatella in a poorly supported, unresolved relationship; nevertheless, association of all three genera into a single clade was supported by an approximately unbiased (AU) test. Any association of these genera singly or as a group with the Hymenostomatia was rejected decisively by AU tests and by a complete absence in the loxocephalids of the unique nucleotide identities that distinguish hymenostomes. Therefore, the morphological and morphogenetic similarities of loxocephalids to hymenostomes may be plesiomorphies, and the conflicting mix of scuticociliate and hymenostome characteristics seen in loxocephalids may result from differing rates of character evolution. Dexiotrichides pangi and Urocentrum, which is currently classified as a peniculid, formed a small clade that associated with hymenostomes and peritrichs. Monophyly of the Loxocephalida with Dexiotrichides and/or Urocentrum included was not rejected by AU; however, inclusion of Urocentrum in the Peniculia was rejected by AU tests. A hypothesis is offered to explain the lack of resolution of loxocephalid ciliates and Urocentrum in phylogenetic trees, namely that their phylogenetic positions are influenced by a combination of heterogeneous data and long‐branch attraction caused by poor representation of taxa in analyses. The well‐known genus Cyclidium, a member of the order Pleuronematida, was revealed to be polyphyletic as a byproduct of our analyses of loxocephalids. In particular, Cyclidium porcatum appears to fall outside the clade containing typical members of the subclass Scuticociliatia and thus invites investigation as a possible member of the order Loxocephalida.  相似文献

To further investigate the use of DNA repair-enhancing agents for skin cancer prevention, we treated Cdk4R24C/R24C/NrasQ61K mice topically with the T4 endonuclease V DNA repair enzyme (known as Dimericine) immediately prior to neonatal ultraviolet radiation (UVR) exposure, which has a powerful effect in exacerbating melanoma development in the mouse model. Dimericine has been shown to reduce the incidence of basal-cell and squamous cell carcinoma. Unexpectedly, we saw no difference in penetrance or age of onset of melanoma after neonatal UVR between Dimericine-treated and control animals, although the drug reduced DNA damage and cellular proliferation in the skin. Interestingly, epidermal melanocytes removed cyclobutane pyrimidine dimers (CPDs) more efficiently than surrounding keratinocytes. Our study indicates that neonatal UVR-initiated melanomas may be driven by mechanisms other than solely that of a large CPD load and/or their inefficient repair. This is further suggestive of different mechanisms by which UVR may enhance the transformation of keratinocytes and melanocytes.  相似文献

Differences exist in body composition assessed by dual‐energy X‐ray absorptiometers (DXAs) between devices produced by different manufacturers and different models from the same manufacturer. Cross‐calibration is needed to allow body composition results to be compared in multicenter trials or when scanners are replaced. The aim was to determine reproducibility and extent of agreement between two fan‐beam DXA scanners (QDR4500W, Discovery Wi) for body composition of regional sites. The sample was: 39 women 50.6 ± 9.6 years old with BMI 26.8 ± 5.5 kg/m2, body fat 33 ± 7%. Four whole body scans (two on each device) were performed over 3 weeks. Major variables were fat mass, nonosseous lean mass, and bone mineral content (BMC) for the truncal and appendicular regions. Extent of agreement was assessed using Bland and Altman plots. Both devices demonstrated good precision with mean test–retest differences close to zero for fat mass, nonosseous lean mass, and BMC of the truncal and appendicular regions. Evaluation of interdevice agreement revealed significant differences for truncal and appendicular BMC, nonosseous lean mass, and fat mass. The greatest interdevice difference was for truncal fat mass (0.69 ± 0.60 kg). Differences in truncal and appendicular fat mass increased in magnitude at higher mean values. Furthermore, differences in truncal and appendicular fat mass were strongly related to BMI (R = ?0.61, R = ?0.55, respectively). In conclusion, in vivo cross‐calibration is important to ensure comparability of regional body composition data between scanners, especially for truncal fat mass and for subjects with higher BMI.  相似文献
