Calcium silicate (CaSiO3, CS) ceramic composites reinforced with graphene nanoplatelets (GNP) were prepared using hot isostatic pressing (HIP) at 1150°C. Quantitative microstructural analysis suggests that GNP play a role in grain size and is responsible for the improved densification. Raman spectroscopy and scanning electron microscopy showed that GNP survived the harsh processing conditions of the selected HIP processing parameters. The uniform distribution of 1 wt.% GNP in the CS matrix, high densification and fine CS grain size help to improve the fracture toughness by ∼130%, hardness by ∼30% and brittleness index by ∼40% as compared to the CS matrix without GNP. The toughening mechanisms, such as crack bridging, pull-out, branching and deflection induced by GNP are observed and discussed. The GNP/CS composites exhibit good apatite-forming ability in the simulated body fluid (SBF). Our results indicate that the addition of GNP decreased pH value in SBF. Effect of addition of GNP on early adhesion and proliferation of human osteoblast cells (hFOB) was measured in vitro. The GNP/CS composites showed good biocompatibility and promoted cell viability and cell proliferation. MicroRNAs (miRNAs) constitute a potent layer of gene regulation by guiding RISC to target sites located on mRNAs and, consequently, by modulating their translational repression. Changes in miRNA expression have been shown to be involved in the development of all major complex diseases. Furthermore, recent findings showed that miRNAs can be secreted to the extracellular environment and enter the bloodstream and other body fluids where they can circulate with high stability. The function of such circulating miRNAs remains largely elusive, but systematic high throughput approaches, such as miRNA profiling arrays, have lead to the identification of miRNA signatures in several pathological conditions, including neurodegenerative disorders and several types of cancers. In this context, the identification of miRNA expression profile in the cerebrospinal fluid, as reported in our recent study, makes miRNAs attractive candidates for biomarker analysis.There are several tools available for profiling microRNAs, such as microarrays, quantitative real-time PCR (qPCR), and deep sequencing. Here, we describe a sensitive method to profile microRNAs in cerebrospinal fluids by quantitative real-time PCR. We used the Exiqon microRNA ready-to-use PCR human panels I and II V2.R, which allows detection of 742 unique human microRNAs. We performed the arrays in triplicate runs and we processed and analyzed data using the GenEx Professional 5 software.Using this protocol, we have successfully profiled microRNAs in various types of cell lines and primary cells, CSF, plasma, and formalin-fixed paraffin-embedded tissues. 相似文献

Maintaining the equilibrium between saturated and unsaturated fatty acids within membrane phospholipids (PLs) is crucial to sustain the optimal membrane biophysical properties, compatible with selective organelle-based processes. Lipointoxication is a pathological condition under which saturated PLs tend to accumulate within the cell at the expense of unsaturated species, with major impacts on organelle function. Here, we show that human bronchial epithelial cells extracted from lungs of patients with Obstructive Pulmonary Diseases (OPDs), i. e. Cystic Fibrosis (CF) individuals and Smokers, display a characteristic lipointoxication signature, with excessive amounts of saturated PLs. Reconstitution of this signature in cellulo and in silico revealed that such an imbalance results in altered membrane properties and in a dramatic disorganization of the intracellular network of bronchial epithelial cells, in a process which can account for several OPD traits. Such features include Endoplasmic Reticulum-stress, constitutive IL8 secretion, bronchoconstriction and, ultimately, epithelial cell death by apoptosis. We also demonstrate that a recently-identified lipid-like molecule, which has been shown to behave as a “membrane-reshaper”, counters all the lipointoxication hallmarks tested. Altogether, these insights highlight the modulation of membrane properties as a potential new strategy to heal and prevent highly detrimental symptoms associated with OPDs. 相似文献

Plants must continually balance the influx of CO2 for photosynthesis against the loss of water vapor through stomatal pores in their leaves. This balance can be achieved by controlling the aperture of the stomatal pores in response to several environmental stimuli. Elevation in atmospheric [CO2] induces stomatal closure and further impacts leaf temperatures, plant growth and water-use efficiency, and global crop productivity. Here, we review recent advances in understanding CO2-perception mechanisms and CO2-mediated signal transduction in the regulation of stomatal movements, and we explore how these mechanisms are integrated with other signaling pathways in guard cells. 相似文献

Daily feeding rhythms in Atlantic salmon II: size-related variation in feeding patterns of post-smolts under constant environmental conditions 总被引:2,自引:0,他引:2
Previous studies of feeding rhythms in post-smolt Atlantic salmon Salmo salar have produced variable results. Therefore, the present study was performed to clarify the source of these differences and to examine size-related variation in daily feeding patterns. A small group of hatchery reared post-smolts was kept in an indoor seawater tank under conditions of constant photoperiod and light intensity, and negligible diurnal variation in water temperature. Feeding responses were monitored by video camera throughout the day. The resultant feeding pattern for the population mirrored an earlier study, with the highest levels of feeding activity in the early morning and late evening. Differences between this and other studies may be due to variation in climate, season or life-history stage. Daily feeding patterns were size-dependent. The largest fish fed at a relatively constant level through the day, medium-sized fish fed in a pattern similar to that described above for the population and small fish fed most at first and last light. These differences were due probably to the effect of competitive interactions upon fish of different sizes. 相似文献

The red list has become a ubiquitous tool in the conservation of species. We analyzed contemporary trends in the threat levels of European orchids, in total 166 species characterized in 27 national red lists, in relation to their reproductive biology and growth form, distribution area, and land cover where they occur. We found that species in central Europe are more threatened than those in the northern, southern, or Atlantic parts of Europe, while species were least threatened in southern Europe. Nectarless and tuberous species are significantly more threatened than nectariferous and rhizomatous taxa. Land cover (ratios of artificial land cover, area of pastures and grasslands, forests and inland wetlands) also significantly impacted the threat level. A bigger share of artificial land cover increases threat, and a bigger share of pasture and grassland lowers it. Unexpectedly, a bigger share of inland wetland area in a country increased threat level, which we believe may be due to the threatened nature of wetlands themselves relative to other natural land cover types. Finally, species occurring in multiple countries are on average less threatened. We believe that large‐scale analysis of current IUCN national red lists as based on their specific categories and criteria may particularly inform the development of coordinated regional or larger‐scale management strategies.
