Recent studies demonstrated that a combination of the gut microbiome has the vital effect on the efficacy of anticancer immune therapies. Regulatory effects of microbiota have been shown in different types of cancer therapies such as chemotherapy and immunotherapy. Immune-checkpoint-blocked therapies are the recent efficient cancer immunotherapy strategies. The target of immune-checkpoint blocking is cytotoxic T lymphocyte protein-4 (CTLA-4) or blockade of programmed death-1 (PD-1) protein and its ligand programmed death ligand 1 (PD-L1) that they have been considered as cancer immunotherapy in recent years. In the latest studies, it have been demonstrated that several gut bacteria such as Akkermansia muciniphila, Bifidobacterium spp., Faecalibacterium spp., and Bacteroides fragilis have the regulatory effects on PD-1, PD-L1, and CTLA-4 blocked anticancer therapy outcome.  相似文献

Abstract Inside-out and right-side out vesicles were isolated from the intracytoplasmic membrane system of the photosynthetic bacterium Rhodopseudomonas viridis and treated with proteinase K. Afterwards the pigment-binding proteins of the photosynthetic apparatus were extracted from the membrane, purified and the N- and C-terminal amino acyl sequences determined.
Forty-eight amino acids were found to be removed from the N-terminal domain of the M-subunit and twenty-eight amino acids split off the L-subunit of reaction center when inside-out vesicles were digested with proteinase K.
Six amino acids of the N-terminal region of the beta polypeptide of the light-harvesting complex B1020 were removed when inside-out vesicles were treated with proteinase K. The N-terminal domains of alpha and gamma polypeptides of the antenna complex B1020 were not cleaved by proteinase K either in right-side out or in inside-out vesicles. It is concluded that the N-terminal domains of M-, L- and β-subunits are exposed and accessible to proteinase K on the cytoplasmic surface of the membrane. This is in agreement with results obtained with other photosynthetic bacteria. The orientation of the other light-harvesting polypeptides is discussed.  相似文献

The prevalence of type 2 diabetes mellitus (T2DM) is increasing rapidly with its associated morbidity and mortality. Many pathophysiological pathways such as oxidative stress, inflammatory responses, adipokines, obesity-induced insulin resistance, improper insulin signaling, and beta cell apoptosis are associated with the development of T2DM. There is increasing evidence of the role of mitochondrial dysfunction in the onset of T2DM, particularly in relation to the development of diabetic complications. Here, the role of mitochondrial dysfunction in T2DM is reviewed together with its modulation by antidiabetic therapeutic agents, an effect that may be independent of their hypoglycemic effect.  相似文献

Ovarian cancer (OC) is the fifth leading cause of cancer-related death among women. The high mortality rate is due to lack of early symptoms, late diagnosis, limited treatment options, and also emerging of drug resistance. Todays, molecular markers have become promising in tumor-targeted therapy. Several molecular markers have been known in OC immunotherapy. Identification of the specific molecular markers with prognostic significance is interested. CD24 is a small sialoglycoprotein which is localized in lipid rafts through its glycosylphosphatidylinositol (GPI) anchor. It has been reported that CD24 is overexpressed in many cancers including OC. Also, CD24 is identified as a cancer stem cell marker in OC. The CD24 expression is associated with the development, invasion, and metastasis of cancer cells. The exact role of CD24 in cancer cells is not clearly understood. Recently, CD24 has been identified as an independent prognostic marker of survival in patients with OC. In this study, we reviewed the molecular targets in OC immune-targeted therapy and also presented an overview of the new molecular marker CD24 and its association with the OC by reviewing the recent literature.  相似文献

Functional connectivity (FC) and graph measures provide powerful means to analyze complex networks. The current study determines the inter-subject-variability using the coefficient of variation (CoV) and long-term test-retest-reliability (TRT) using the intra-class correlation coefficient (ICC) in 44 healthy subjects with 35 having a follow-up at years 1 and 2. FC was estimated from 256-channel-EEG by the phase-lag-index (PLI) and weighted PLI (wPLI) during an eyes-closed resting state condition. PLI quantifies the asymmetry of the distribution of instantaneous phase differences of two time-series and signifies, whether a consistent non-zero phase lag exists. WPLI extends the PLI by additionally accounting for the magnitude of the phase difference. Signal-space global and regional PLI/wPLI and weighted first-order graph measures, i.e. normalized clustering coefficient (gamma), normalized average path length (lambda), and the small-world-index (SWI) were calculated for theta-, alpha1-, alpha2- and beta-frequency bands. Inter-subject variability of global PLI was low to moderate over frequency bands (0.12<CoV<0.28), higher for wPLI (0.25<CoV<0.55) and very low for gamma, lambda and SWI (CoV<0.048). TRT was good to excellent for global PLI/wPLI (0.68<ICC<0.80), regional PLI/wPLI (0.58<ICC<0.77), and fair to good for graph measures (0.32<ICC<0.73) except wPLI-based lambda in alpha1 (ICC = 0.12). Inter-electrode distance correlated very weakly with inter-electrode PLI (−0.06<rho<0) and weakly with inter-electrode wPLI (−0.22<rho<−0.18). Global PLI/wPLI and topographic connectivity patterns differed between frequency bands, and all individual networks showed a small-world-configuration. PLI/wPLI based network characterization derived from high-resolution EEG has apparently good reliability, which is one important requirement for longitudinal studies exploring the effects of chronic brain diseases over several years.  相似文献
