Reliable determination of time since death in human skeletons or single bones often is limited by methodically difficulties. Determination of the specific activity ratio of natural radionuclides, in particular of 232Th (Thorium), 228Th and 228Ra (Radium) seems to be a new appropriate method to calculate the post mortem interval. These radionuclides are incorporated by any human being, mainly from food. So with an individual's death the uptake of radionuclides ends. But the decay of 232Th produces 228Ra and 228Th due to its decay series, whereas 228Th is continuously built up in the human's bones. Thus, it can be concluded that in all deceased humans at different times after death different activity ratios of 228Th to 228Ra will develop in bone. According to this fact it should be possible to calculate time since death of an individual by first analysing the specific activities of 228Th and 228Ra in bones of deceased and then determining the 228Th/228Ra activity ratio, which can be assigned to a certain post-mortem interval.  相似文献

Particulate organic matter (POM) and dissolved organic carbon (DOC) release by six dominant hermatypic coral genera (Acropora, Fungia, Goniastrea, Millepora, Pocillopora and Stylophora) were measured under undisturbed conditions by laboratory incubations during four seasonal expeditions to the Northern Red Sea. In addition, the influence of environmental factors (water temperature, light availability and ambient inorganic nutrient concentrations) was evaluated. Particulate organic carbon (POC) and particulate nitrogen (PN) release were always detectable and genus-specific, with Stylophora releasing most POM (6.5 mg POC and 0.5 mg PN m−2 coral surface area h−1) during all seasons. The fire coral Millepora released significantly less POM (0.3 mg POC and 0.04 mg PN m−2 coral surface area h−1) than all investigated anthozoan genera. The average POC:PN ratio of POM released by all coral genera was 12 ± 1, indicating high carbon/low nitrogen content of coral-derived organic matter. POM release showed little seasonal variation, but average values of POC and PN release rates correlated with water temperature, light availability and ambient nitrate concentrations. DOC net release and elevated DOC:POC ratios were detectable for Acropora, Goniastrea and Millepora, revealing maximum values for Acropora (30.7 mg DOC m−2 coral surface area h−1), whilst predominant DOC uptake was observed for Pocillopora, Fungia and Stylophora. Depth-mediated light availability influenced DOC fluxes of Acropora and Fungia, while fluctuations in water temperature and ambient inorganic nutrient concentrations showed no correlation. These comprehensive data provide an important basis for the understanding of coral reef organic matter dynamics and relevant environmental factors.  相似文献

This paper presents seasonal in situ monitoring data on benthic coverage and coralalgae interactions in high-latitude fringing reefs of the Northern Red Sea over a period of 19 months. More than 30% of all hermatypic corals were involved in interaction with benthic reef algae during winter compared to 17% during summer, but significant correlation between the occurrence of coralalgae interactions and monitored environmental factors such as temperature and inorganic nutrient availability was not detected. Between 5 and 10-m water depth, the macroalgae Caulerpa serrulata, Peyssonnelia capensis and filamentous turf algae represented almost 100% of the benthic algae involved in interaction with corals. Turf algae were most frequently (between 77 and 90% of all interactions) involved in interactions with hermatypic corals and caused most tissue damage to them. Maximum coral tissue loss of 0.75% day−1 was observed for Acropora-turf algae interaction during fall, while an equilibrium between both groups of organisms appeared during summer. Slow-growing massive corals were more resistant against negative algal influence than fast-growing branching corals. Branching corals of the genus Acropora partly exhibited a newly observed phenotypic plasticity mechanism, by development of a bulge towards the competing organism, when in interaction with algae. These findings may contribute to understand the dynamics of phase shifts in coral reefs by providing seasonally resolved in situ monitoring data on the abundance and the competitive dynamic of coralalgae interactions.  相似文献

Identification of armored heterotrophic dinoflagellates relies, in part, on plate tabulations obtained by SEM. Currently, two methods are used to visualize plate morphology and develop plate tabulations: swelling the sutures between the cellulose plates of intact organisms or stripping off the outer membranes with ethanol to expose the underlying cellulose plates. Both approaches are problematic with lightly armored dinoflagellates because sutures do not consistently swell to enable visualization, and the outer membranes are not consistently stripped. Further, generic and species differences necessitate frequent modification of these protocols to obtain reliable results. We describe an improved membrane stripping technique using the detergent Triton X‐100. Our method provides a more consistent standardized approach to removing the outer membranes of lightly armored dinoflagellates, including Pfiesteria shumwayae Glasgow & Burkholder, a taxon that has, until now, proven very difficult to strip with currently published methods. This method allows visualization of the sulcus, a region previously difficult to observe, and will greatly facilitate taxonomic studies of the lightly armored forms.  相似文献

The triglyceride-synthesizing enzyme acyl CoA:diacylglycerol acyltransferase 1 (DGAT1) plays a critical role in hepatitis C virus (HCV) infection by recruiting the HCV capsid protein core onto the surface of cellular lipid droplets (LDs). Here we find a new interaction between the non-structural protein NS5A and DGAT1 and show that the trafficking of NS5A to LDs depends on DGAT1 activity. DGAT1 forms a complex with NS5A and core and facilitates the interaction between both viral proteins. A catalytically inactive mutant of DGAT1 (H426A) blocks the localization of NS5A, but not core, to LDs in a dominant-negative manner and impairs the release of infectious viral particles, underscoring the importance of DGAT1-mediated translocation of NS5A to LDs in viral particle production. We propose a model whereby DGAT1 serves as a cellular hub for HCV core and NS5A proteins, guiding both onto the surface of the same subset of LDs, those generated by DGAT1. These results highlight the critical role of DGAT1 as a host factor for HCV infection and as a potential drug target for antiviral therapy.  相似文献

Staphylococcus aureus community‐acquired (CA) MRSA strains are highly virulent and can cause infections in otherwise healthy individuals. The most important mechanism of the host for clearing S. aureus is phagocytosis by neutrophils and subsequent killing of the pathogen. Especially CA‐MRSA strains are very efficient in circumventing this neutrophil killing. Interestingly, only a relative small number of virulence factors have been associated with CA‐MRSA, one of which are the phenol soluble modulins (PSMs). We have recently shown that the PSMs are functionally inhibited by serum lipoproteins, indicating that PSMs may exert their cytolytic function primarily in the intracellular environment. To further investigate the intracellular role of the PSMs we measured the effect of the α‐type and β‐type PSMs on neutrophil killing after phagocytosis. Using fluorescently labelled S. aureus, we measured bacterial survival after phagocytosis in a plate reader, which was employed next to flow cytometry and time‐lapse microscopy. Phagocytosis of the CA‐MRSA strain MW2 by human neutrophils resulted in rapid host cell death. Using mutant strains of MW2, we demonstrated that in the presence of serum, the intracellular expression of only the psmα operon is both necessary and sufficient for both increasedneutrophil cell death and increased survival of S. aureus. Our results identify PSMα peptides as prominent contributors to killing of neutrophils after phagocytosis, a finding with major implications for our understanding of S. aureus pathogenesis and strategies for S. aureus vaccine development.  相似文献

Background

Alzheimer disease (AD) is the most common form of dementia but the identification of reliable, early and non-invasive biomarkers remains a major challenge. We present a novel miRNA-based signature for detecting AD from blood samples.

Results

We apply next-generation sequencing to miRNAs from blood samples of 48 AD patients and 22 unaffected controls, yielding a total of 140 unique mature miRNAs with significantly changed expression levels. Of these, 82 have higher and 58 have lower abundance in AD patient samples. We selected a panel of 12 miRNAs for an RT-qPCR analysis on a larger cohort of 202 samples, comprising not only AD patients and healthy controls but also patients with other CNS illnesses. These included mild cognitive impairment, which is assumed to represent a transitional period before the development of AD, as well as multiple sclerosis, Parkinson disease, major depression, bipolar disorder and schizophrenia. miRNA target enrichment analysis of the selected 12 miRNAs indicates an involvement of miRNAs in nervous system development, neuron projection, neuron projection development and neuron projection morphogenesis. Using this 12-miRNA signature, we differentiate between AD and controls with an accuracy of 93%, a specificity of 95% and a sensitivity of 92%. The differentiation of AD from other neurological diseases is possible with accuracies between 74% and 78%. The differentiation of the other CNS disorders from controls yields even higher accuracies.

Conclusions

The data indicate that deregulated miRNAs in blood might be used as biomarkers in the diagnosis of AD or other neurological diseases.  相似文献

The oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) is a serious pest of fruit crops in Asia, several Pacific Islands and sometimes the western United States, particularly California. Sterile insect technique programs have been developed for management of several tephritid fruit fly pests. These programs are based on continuous production of adult fruit flies. The high expense of mass-rearing oriental fruit flies drives research to improve the cost effectiveness of rearing programs. One recent improvement for mass rearing oriental fruit flies involves adding wheat germ oil (WGO) to the larval culture medium, which improved several parameters of biological performance. The performance enhancing influence of WGO is due to the presence of polyunsaturated fatty acids (PUFAs), some of which are nutritionally essential for many insect species. We considered the issue of whether WGO supplementation of the larval culture medium influences the fatty acid make up of adult tissues. We report that WGO supplementation led to substantial increases in adult tissue C18 PUFAs. Unlike the outcomes of unrelated nutritional studies on moths, the PUFA components of WGO did not improve adult fruit fly performance. Taken with recent publications reporting that WGO in larval diets influences gene expression, we conclude that dietary WGO improves biological performance of adults through changes in tissue C18 PUFAs and gene expression.  相似文献
