The uptake and metabolism of sugars by suspension-cultured Catharanthusroseus cells were investigated. Substantially all the sucrosein the culture medium was hydrolyzed to glucose and fructosebefore being taken up by the cells. The activity of invertasebound to cell walls, determined in situ, was high at the earlystage of culture. Glucose was more easily taken up by the cellsthan was fructose. Tracer experiments using [U-14C]glucose and[U-14C]fructose indicated that glucose is a better precursorfor respiration than fructose, while fructose is preferentiallyutilized for the synthesis of sucrose, especially in the earlyphase of cell growth. Possible metabolic routes of sugar insuspension-cultured Catharanthus roseus cells are discussedin the context of these results. There are only a few reports on the relative biological effectiveness (RBE) of thermal neutrons and 10B(n,α)7Li reactions either in vitro or in vivo. The data in this paper summarize almost all previously published in vitro data. Because only a few reactors are available for biomedical purposes, it is difficult to make a comparison of data from experiments using the same kind of radiation, and also to make a comparison of data from experiments using the different kinds of radiations. However, it is indispensable for boron neutron capture therapy to make a radiobiological analysis. Polyploidization, or genome duplication, has played a critical role in the diversification of animals, fungi and plants. Little is known about the population structure and multiple origins of polyploid species because of the difficulty in identifying multiple homeologous nuclear genes. The allotetraploid species Arabidopsis kamchatica is closely related to the model species Arabidopsis thaliana and is distributed in a broader climatic niche than its parental species. Here, we performed direct sequencing of homeologous pairs of the low-copy nuclear genes WER and CHS by designing homeolog-specific primers, and obtained also chloroplast and ribosomal internal transcribed spacer sequences. Phylogenetic analysis showed that 50 individuals covering the distribution range including North America are allopolyploids derived from Arabidopsis lyrata and Arabidopsis halleri . Three major clusters within A. kamchatica were detected using Bayesian clustering. One cluster has widespread distribution. The other two are restricted to the southern part of the distribution range including Japan, where the parent A. lyrata is not currently distributed. This suggests that the mountains in Central Honshu and surrounding areas in Japan served as refugia during glacial–interglacial cycles and retained this diversity. We also found that multiple haplotypes of nuclear and chloroplast sequences of A. kamchatica are identical to those of their parental species. This indicates that multiple diploid individuals contributed to the origin of A. kamchatica . The haplotypes of low-copy nuclear genes in Japan suggest independent polyploidization events rather than introgression. Recent human activities have spread numerous plant species across the globe, yet it is unclear to what degree historical human activities influenced plant dispersal. In western North America, Camassia quamash was one of the most important food plants for indigenous peoples, who transported its propagules either intentionally or accidentally. We investigated how human and natural dispersal might have contributed to the current pattern of spatial genetic structure in C. quamash by performing phylogeographical surveys at two geographical scales. We sequenced two noncoding regions of chloroplast deoxyribonucleic acid (DNA) in 226 individuals from 53 populations of C. quamash as well as 126 individuals from 21 populations of the non-food plant Zigadenus venenosus . Contrary to the expectation of anthropogenic transport, C. quamash populations did not exhibit weaker genetic structure than Z. venenosus populations. We also failed to find convincing evidence for signatures of transport. Instead, our data showed strong effects of past glaciation and geographical barriers of the mountains in the Cascade Range, Olympic Peninsula and Vancouver Island. West of the Cascades, the species appears to have largely migrated northward from a southern refugium after deglaciation, whereas few populations having a highly divergent haplotype might have survived in southwestern Washington. Our data suggest that despite substantial ethnobotanical evidence for anthropogenic transport, the current pattern of genetic structure of C. quamash does not show any detectable signatures of transport by indigenous peoples and is better understood as the result of natural dispersal processes. 相似文献

Changes in the nucleoli of maturing oocytes and the eggs of Cynops pyrrhogaster were studied with light and electron microscopy. Extrachromosomal nucleoli moved toward the center of the germinal vesicle in response to the maturation stimulus, released presumed ribosomal ribonucleoprotein, and further moved toward the center of the nucleus to form an aggregate with chromosomes which behaved in a similar manner. A few. ball-shaped nucleolar masses were formed from this aggregate, leaving the chromosomes and probably the extrachromosomal nucleolar organizer. The chromosomes then proceeded to the first meiotic metaphase. The nucleolar masses were surrounded by a layer of mitochondria and became smaller with formation of pinched-off fragments, which were also surrounded by mitochondria, during the time the egg was moving down the oviduct. Abstract The pollination biology of Hosta sieboldiana and H. sieboldii is investigated comparatively in Central Japan. Both species have homogamous, one-day flowers pollinated by bumblebees. The abdomens of the bees touch the stigma on the extended style when they land on the anthers inside the herkogamous flower, and autogamy is effectively prevented. However, the flowers are fairly self-compatible, and geitonogamy may occur rather frequently because two or more flowers on a scape very often bloom at the same time and many ramets are contiguous. The pollen/ovule ratios suggest that these species are facultative outbreeders. The flower of H. sieboldii seems completely suited to bumblebee pollination. In H. sieboldiana the stigma of the flower, whose style strongly protrudes, is not always touched by bumblebees, but frequent visitation of bumblebees results in pollination of almost all the flowers. Both species have similar pollination systems but seem reproductively isolated by blooming times and habitats. The comparative morphology and anatomy of the leaves of the rheophytic Rhododendron ripense and the closely related inland species Rhododendron macrosepalum were examined. The leaf of R. ripense is thinner than that of R. macrosepalum, with leaf length to width ratios (leaf index) of 2.92 and 1.91, respectively. Moreover, the leaf of R. ripense consists of fewer cells than the leaf of R. macrosepalum, suggesting stenophyllization of R. ripense caused by the decreased number of cells. In addition, leaf thickness and the number of stomata per leaf of R. ripense were significantly greater than those of R. macrosepalum, but the density of the short glandular pilose hairs on the leaf of R. ripense was lower. The observed morphological differences between the two species may be explained by certain aspects of the riparian environment, such as high irradiation and frequent flooding after heavy rainfall, to which R. ripense is exposed. 相似文献
