THE reconstitution in vitro of flagellar filaments from their component flagellin monomers in Salmonella has shown that the filaments have structural polarity and grow at an end distal to the cell body1; flagella in vivo also grow from their tips2,3. This suggests that even when flagella are attached to living cells, filaments may be reconstituted from exogenous flagellin monomers at the tips in appropriate conditions. In spite of some negative results4, we have been encouraged5–10 to re-examine the question.  相似文献

To elucidate the factors responsible for the evolution of host specialisation, prey capture performance in hatchlings of two sibling ladybird species, Harmonia yedoensis and H. axyridis, feeding on four aphid species was examined. Harmonia yedoensis is a specialist predator that preys only on pine aphids in the field, whereas H. axyridis is a generalist predator with a broad prey range. 2. In H. yedoensis, sibling cannibalism in each clutch was intense and predation against pine aphid as well as other aphid species was moderately successful. In contrast, the predation success rate of H. axyridis against pine aphid was quite low. 3. Moreover, it was experimentally shown that increased maternal investment enhanced prey capture performance against pine aphid in H. yedoensis but not in H. axyridis hatchlings, despite their increased body size due to maternal investment. 4. In addition, morphological and behavioural analysis showed that hatchlings of H. yedoensis had longer legs and a larger head capsule size and could walk faster than H. axyridis. 5. In summary, the interactive effects between a large amount of maternal investment and morphological specialisation of the first instars may enable H. yedoensis to capture the pine aphid efficiently, a highly elusive prey for ladybird hatchlings. The ability of H. yedoensis to utilise the three other aphid species in addition to the pine aphid suggests that a trade‐off in prey capture performance is not the main factor in the host specialisation of H. yedoensis.  相似文献

We tested 14 pairs of microsatellite primers that had been developed for the genus Polistes to assess their applicability for determining genetic structure in a colony of the genus Parapolybia. At least one of the 14 Polistes primer sets was useful for Parapolybia indica, and the locus identified by this primer set was highly polymorphic. Using this primer set, we estimated the average relatedness among female nestmates of Parapolybia indica to be 0.82 ± 0.05 (mean ± SE), indicating that in this species single mating is normal, although some cases of multiple mating was found.  相似文献

A novel inhibitor of topoisomerase II designated as 2070-DTI was isolated from the culture filtrate of Streptomyces sp. strain No. 2070. The structure was determined to be that of the known soyasaponin I on the basis of spectroscopic methods (NMR and MS). 2070-DTI strongly inhibited the decatenation activity of human placenta topoisomerase II in a noncompetitive manner, and weakly inhibited or was inert towards the relaxation activities of various topoisomerase I's and DNA-related enzymes. 2070-DTI is an inhibitor belonging to the cleavable complex-nonforming type without DNA intercalation.  相似文献

Roots in the soil are illuminated by far‐red (FR) light passed through plant tissues in the daytime, and are in complete darkness at night. To evaluate whether gene expression of roots is affected by a dark‐FR light cycle, gene expression profiles were analysed for dark‐adapted versus light‐grown plants and for FR light‐illuminated versus dark‐adapted plants using the RIKEN Arabidopsis full‐length cDNA microarray (containing approximately 7000 independent, full‐length cDNA groups). Among candidate dark‐ and FR‐regulated genes, several were further analysed. Eleven dark‐inducible and five dark‐repressed genes were characterized. Almost all the dark‐inducible and –repressed genes were oppositely regulated by FR light illumination. The functions of dark‐ and FR‐responsive genes and the significance of FR light‐regulated gene expression in roots under ground are discussed.  相似文献

A murine macrophage-like cell line,J774,acquried,in response to LPS,an ability to kill tumor necrosisfactor(TNF)-insensitive target P815 mastocytoma cells,whereas another cell line,P388D1,did not.LPS-triggered signaling mechanisms between the two celllines were compared with an aim to inquire about thepossible nature of the above-mentioned difference.Theresults showed that two cell lines respond to LPS-treatment by parallel activation of both phospholipasesC and A_2(PLC and PLA_2)to approximately the sameextent.The maximum response of both enzymes of J774cells was noted within 10 min of the treatment,whereas that of P388D1 cells required more than 20min.The other properties of LPS-responsive enzymesstudied were similar between two cell lines,ineludingActivation of PLC and PLA_2 and PKC in macrophages by LPSCa~(2 )augmentation of enzyme activation,participationof guanine nucleotide binding (G) proteins in theinitial activation processes,and inhibition of enzymeactivation by the prior treatment of cells with choleraorpartussis toxins etc.Moreover,LPS-triggered activationof PLC and PLA2 was found to be followed by theincrease of PKC activities in both cell lines.In spite ofthese similarities,J774 cells possessed both basic andacidic forms of PKC activities,while P 388D1 cells ownedonly PKC of basic form.Nevertheless,the question whyJ774 cells,but not P388D1 cells,can acquire thetumoricidal actiyity,aganist P815 cells following LPS-treatment remains to be answered.  相似文献

Salix hukaoana is an endangered riparian tree that is found and known only in several isolated sites in eastern Japan, and that has been suffering from the loss and fragmentation of its habitat. To investigate the genetic variation and structures of this willow in these sites, eight polymorphic microsatellites with three to eight alleles were characterized. Cross‐species amplification was successful for many of the loci using the congeners Salix sachalinensis and Chosenia arbutifolia. This set of microsatellites will be useful for the study of the spatial genetic structures of S. hukaoana and other coexisting willows suffering from habitat fragmentation in riparian forests.  相似文献
