Positive interactions between plants play a fundamental role in environments with extreme conditions, such as coastal sand dunes, where the establishment of many species is limited by high temperature, and low availability of water and nutrients in the soil. The ability to store water and enrich the soil with nutrients characterizes bromeliads as potential nurse plants for other species. This study aimed to analyze the potential of the bromeliad Vriesea friburgensis to act as a nurse plant, and to compare this interaction in coastal dunes in herbaceous versus scrub vegetation. The study was performed on Santa Catarina Island in Brazil. Juveniles of other species associated with V. friburgensis were collected within and under bromeliad tanks and on adjacent plots beside each bromeliad. The abundance, species richness and height of juveniles were recorded. We found that abundance and richness of Vf‐associated juveniles were lower than those Vf‐not associated, in herbaceous vegetation (negative interaction). However, the height of some shrubs species, like Eupatorium casarettoi and Tibouchina urvilleana, was greater when Vf‐associated (positive interaction). In scrub vegetation, the abundance and richness of juveniles were not significantly affected by V. friburgensis presence (neutral interaction). However, the height of some shrubs and trees species, like Clusia criuva, Eugenia catharinae, Myrsine parvifolia and Ocotea pulchella, was greater when Vf‐associated. Species composition associated with V. friburgensis differed from the assemblage in adjacent plots, indicating that some species interact more with nurse plants, whereas others fail to develop when associated with this bromeliad; the difference between groups in species composition was less evident in scrub vegetation areas. Clusia criuva was considered an indicator species for Vf‐associated juveniles in both vegetation types. Despite the fact that these tank bromeliads do not function as nurse plants for some species, there are others that seem to be favoured by this interaction.  相似文献

On cats with pretrigeminal brainstem transection, we studied the properties of visually sensitive neurons of the extrastriate associative cortical area 21b. The dimensions and spatial distribution of the receptive fields (RF) of the neurons within the vision field were determined. It was found that large-sized RF prevailed within the area 21b (10 to 200 deg2, 61%; greater than 200 deg2, 22%), whereas small-sized RF (1 to 10 deg2) constituted 17% of all the studied RF. Stationary visual stimuli evoked onoff, off, and on responses in 43, 30, and 27% neurons of the area 21b, respectively. In the cases where moving stimuli were presented, 35% of the neurons demonstrated directional sensitivity; the rest of the neurons (65%) were directionally insensitive. We also found a group of neurons that were capable of differentiating not only the direction of the stimulus movement along the RF but also the dimension, shape, and orientation of a complicated moving stimulus. Taking into account the data obtained, we discuss the functional role of the neurons, which demonstrated a specific (specialized with respect to a set of the parameters of visual stimulus, and not to a single parameter) response in central processing of the sensory information.  相似文献

The ether-à-go-go potassium channels hEag1 and hEag2 are highly homologous. Even though both possess identical voltage-sensing domain S4, the channels act differently in response to voltage. Therefore we asked whether transmembrane domains other than the voltage sensor could contribute to the voltage-dependent behaviour of these potassium channels. For this chimaeras were created, in which each single transmembrane domain of hEag1 was replaced by the corresponding segment of hEag2. The voltage-dependent properties of the chimaeras were analysed after expression in Xenopus laevis oocytes using the two-electrode voltage-clamp method. By this we found, that only the mutations in transmembrane domains S5 and S6 are able to change the voltage sensitivity of hEag1 by shifting the half-activation potential (V 50) to values intermediate between the two wild types. Moreover, the presence of Mg2+ has strong effects on the voltage sensitivity of hEag2 shifting V 50 by more than 50 mV to more positive values. Interestingly, despite the identical binding site Mg2+ showed only little effects on hEag1 or the chimaeras. Altogether, our data suggest that not only transmembrane spanning regions, but also non-membrane spanning regions are responsible for differences in the behaviour of the hEag1 and hEag2 potassium channels. Bioluminescence resonance energy transfer (BRET) is increasingly being used to monitor protein-protein interactions and cellular events in cells. However, the ability to monitor multiple events simultaneously is limited by the spectral properties of the existing BRET partners. Taking advantage of newly developed Renilla luciferases and blue-shifted fluorescent proteins (FPs), we explored the possibility of creating novel BRET configurations using a single luciferase substrate and distinct FPs. Three new (to our knowledge) BRET assays leading to distinct color bioluminescence emission were generated and validated. The spectral properties of two of the FPs used (enhanced blue (EB) FP2 and mAmetrine) and the selection of appropriate detection filters permitted the concomitant detection of two independent BRET signals, without cross-interference, in the same cells after addition of a unique substrate for Renilla luciferase-II, coelentrazine-400a. Using individual BRET-based biosensors to monitor the interaction between G-protein-coupled receptors and G-protein subunits or activation of different G-proteins along with the production of a second messenger, we established the proof of principle that two new BRET configurations can be multiplexed to simultaneously monitor two dependent or independent cellular events. The development of this new multiplexed BRET configuration opens the way for concomitant monitoring of various independent biological processes in living cells.  相似文献

Anaerobic metabolism and oxygen carrying-capacity of white shrimp (Penaeus vannamei) exposed to short term (three days) and long term (two weeks) moderate hypoxia (2-2.6 mg/L) was investigated. Glucose and lactate levels in hemolymph increased under both hypoxic conditions, indicating an activation of anaerobic pathways during the two-weeks exposure period. In muscle, no differences of glucose and lactate levels were observed between the control group and the exposed groups. In animals exposed to hypoxia for two weeks, hemocyanin and copper in hemolymph were higher than in animals under normoxic conditions or exposed for three days. These results indicate that an increase in oxygen carrying-capacity in shrimp is evident only after a sustained condition of hypoxia. Copper levels in the hepatopancreas decreased in both hypoxic conditions, suggesting a mobilization of copper stores for hemocyanin synthesis. These results indicate that penaeid shrimp can tolerate moderate hypoxic conditions by physiological adaptations, such as anaerobic metabolism and increased oxygen carrying-capacity. These adaptations require an adequate dietary supply of proteins and copper for hemocyanin synthesis and of carbohydrates for anaerobic metabolism.  相似文献

Muscarinic activation of tracheal smooth muscle (TSM) involves a M3AChR/heterotrimeric-G protein/NPR-GC coupling mechanism. G protein activators Mastoparan (MAS) and Mastoparan-7 stimulated 4- and 10-fold the NPR-GC respectively, being insensitive to PTX and antibodies against Gαi/o subfamily. Muscarinic and MAS stimulation of NPR-GC was blocked by antibodies against C-terminal of Gαq16, whose expression was confirmed by RT-PCR. However, synthetic peptides from C-terminal of Gαq15/16 stimulated the NPR-GC. Coupling of αq16 to M3AChR is supported by MAS decreased [3H]QNB binding, being abolished after M3AChR-4-DAMP-alkylation. Anti-i3M3AChR antibodies blocked the muscarinic activation of NPR-GC, and synthetic peptide from i3M3AChR (M3P) was more potent than MAS increasing GTPγ [35S] and decreasing the [3H]QNB activities. Coupling between NPR-GC and Gαq16 was evaluated by using trypsin-solubilized-fraction from TSM membranes, which displayed a MAS-sensitive-NPR-GC activity, being immunoprecipitated with anti-Gαq16, also showing an immunoreactive heterotrimeric-G-β -subunit. These data support the existence of a novel transducing cascade, involving Gαq16β γ coupling M3AChR to NPR-GC.  相似文献

Apicomplexans are the causative agents of numerous important infectious diseases including malaria and toxoplasmosis. Most of them harbour a chloroplast-like organelle called the apicoplast that is essential for the parasites’ metabolism and survival. While most apicoplast proteins are nuclear encoded, the organelle also maintains its own genome, a 35 kb circle. In this study we used Toxoplasma gondii to identify and characterise essential proteins involved in apicoplast genome replication and to understand how apicoplast genome segregation unfolds over time. We demonstrated that the DNA replication enzymes Prex, DNA gyrase and DNA single stranded binding protein localise to the apicoplast. We show in knockdown experiments that apicoplast DNA Gyrase A and B, and Prex are required for apicoplast genome replication and growth of the parasite. Analysis of apicoplast genome replication by structured illumination microscopy in T. gondii tachyzoites showed that apicoplast nucleoid division and segregation initiate at the beginning of S phase and conclude during mitosis. Thus, the replication and division of the apicoplast nucleoid is highly coordinated with nuclear genome replication and mitosis. Our observations highlight essential components of apicoplast genome maintenance and shed light on the timing of this process in the context of the overall parasite cell cycle.  相似文献
