Miniature end-plate potentials were used in studying severalaspects of the neuromuscular systems in the cockroach femur.The similar sizes and time courses of miniatures associatedwith fast and slow type excitatory axons suggest that they employthe same transmitter. There is other evidence also indicatingthat the essential difference between these two excitatory systemsis in the number of packets of transmitter released per nerveimpulse rather than different transmitter substances. From theshapes of miniatures it was suspected that typical muscle fibersmight have a branching structure. This was confirmed by histologicalexamination, intracellular stimulation, and intracellular dyeinjection. That inhibitory transmission is quantal is indicatedby hyperpolarizing miniatures which occur at random time intervals.Inhibitory transmission can be made to fail and recover in astepwise manner by manipulating the Ca/Mg ratio. In studiesof toxins which affect transmitter release at vertebrate motorend-plates, botulinal toxin was found to be without effect ateither excitatory or inhibitory junctions in cockroach muscle.However, black widow spider venom acted as it does in vertebrates,promoting massive release of transmitters and then permanentinactivation of the junctions.  相似文献

SUMMARY: The heat resistance of a number of strains of Streptococcus lactis and Str. faecalis at 60° in Ringer's solution and separated milk has been determined and the Decimal Reduction Times (D.R.T.) calculated.
Heating in separated milk at 60° enabled Str. lactis to be sharply differentiated in most cases from Str. faecalis. The latter gave D.R.T. values usually much above 3 min. while the former gave values usually under 1 min. and certainly less than 3 min. Single tests might give misleading results with certain strains.
The log. survivor-time graphs of both species were not always straight lines over all their length. A preliminary period of slow death was frequently observed.  相似文献

ABSTRACT. The abundances of benthic naked amoebae in the sediments of the Clyde Sea Area, Scotland, were studied throughout 1991. Four sites differing in sediment grain size and organic carbon content were chosen for study. Amoebae were enumerated by enrichment cultivation methods and found to be numerically important, attaining densities of up to 14,883 amoebae/cm3 on one occasion. They were most abundant, and temporally stable, in finer sediments where they averaged 2,224 cells/cm3 and lower and more variable in the sandy sediments, averaging 874 cells/cm3 throughout the year. In general, amoebae were most abundant in the surface sediment layers. Around 70 different morphotypes were recorded, and 61% of all amoebae counted were less than 10 μm in length. This is the first detailed quantitative study of benthic marine gymanamoebae and shows that naked amoebae and flagellates are numerically the dominant interstitial fauna in sediments of this area. Moreover, the gymnamoebae comprised the largest proportion of total protozoan biomass (excluding foraminiferans) and clearly need to be considered in future models of benthic carbon flow.  相似文献

Autumn-sown crops of broad beans (Vicia faba L.) in England often contain plants with some leaves characteristically distorted and with a chlorotic mosaic. From some of these plants true broad-bean mosaic virus was isolated in 1959 and 1960 but not in 1965 and 1966. From other plants a similar but distinct virus, which caused staining of the seeds and we call broad-bean stain virus, was isolated in 1960, 1965 and 1966. The two viruses were readily distinguished in serological tests, and in some test plants. Both were seed-borne, and spread in crops, but were not transmitted by several animal species tested as vectors. Both viruses have isometric particles about 25 mμ in diameter. Some of these particles contain about 35% ribonucleic acid, some about 26% and some of those of broad-bean stain virus contain none; these three types of particles had sedimentation coefficients of about 120–130 S, 100 S and 60 S respectively. The ribonucleic acid of each virus had molar base content of G 23%, A 26%, C 18% and U 32%. These two viruses are members of the cowpea mosaic group of plant viruses; broad-bean strain virus was serologically related to cowpea mosaic, F I, red-clover mottle, and squash mosaic viruses. The particles of all these viruses and of true broad-bean mosaic virus were similar in appearance, sedimentation behaviour, and nucleic acid content and composition. 1. Sedimentary remains of aquatic plants, both vegetative (turions, leaves, spines) and reproductive (fruits, seeds, pollen), may provide a record of temporal changes in the submerged vegetation of lakes. An independent assessment of the degree to which these remains reflect past floristic change is, however, rarely possible. 2. By exploiting an extensive series of historical plant records for a small shallow lake we compare plant macrofossil (three cores) and pollen (one core) profiles with the documented sequence of submerged vegetation change since c. 1750 AD. The data set is based on 146 site visits with 658 observations including 42 taxa classified as aquatic, spanning 250 years. 3. Approximately 40% of the historically recorded aquatic taxa were represented by macro‐remains. In general macrofossils underestimated past species diversity, with pondweeds (three of eight historically recorded Potamogeton species were found) particularly poorly represented. Nonetheless, several taxa not reported from historical surveys (e.g. Myriophyllum alterniflorum and Characeae) were present in the sediment record. 4. The pollen record revealed taxa which left no macro‐remains (e.g. Littorella uniflora), and the macrofossil record provided improved taxonomic resolution for some taxa (e.g. Potamogeton) and a more reliable record of persistence, appearance and loss of others (e.g. Myriophyllum spp. and Nymphaeaceae). 5. Detrended correspondence analysis indicated that changes in the community composition evidenced by the palaeolimnological and historical records were synchronous and of a similar magnitude. Both records pointed to a major change at around 1800, with the historical record suggesting a more abrupt change than the sedimentary data. There was good agreement on a subsequent change c. 1930. 6. The palaeolimnological data did not provide a complete inventory of historically recorded species. Nevertheless, these results suggest that combined macrofossil and pollen records provide a reliable indication of temporal change in the dominant components of the submerged and floating‐leaved aquatic vegetation of shallow lakes. As such palaeolimnology may provide a useful tool for establishing community dynamics and successions of plants over decadal to centennial timescales.  相似文献

1. In a region of south‐eastern England, we investigated the hierarchical genetic structure of populations of two stream‐dwelling caddisflies (Trichoptera: Polycentropodidae) with contrasting distributions: Plectrocnemia conspersa inhabits numerous small, patchily distributed seeps and streams, while the confamilial Polycentropus flavomaculatus is found in fewer but larger streams and rivers. We also contrasted the genetic structure of P. conspersa in the lowland south‐east with that in an upland region in the north west. 2. Microsatellite genotypes were obtained from samples of both species taken from a ‘core area’ and at sites 15, 40 and 100 km from this core (two regions for P. conspersa, totalling 45 sites and 1405 larvae; one region for P. flavomaculatus, totalling 10 sites and 269 larvae). 3. The genetic structure of P. conspersa differed in the two regions. In the upland north‐west, significant genetic differentiation was observed at a spatial scale of around 40 km from the core, while there was no structure in the lowland south‐east up to around 100 km. Areas of high altitude did not appear directly to reduce gene flow, whereas other potential landscape barriers, including particular geological formations, large urban areas and the sea had a pronounced effect. 4. Weak genetic differentiation in P. conspersa across large distances, particularly in the lowland south‐east, suggests that it disperses strongly, facilitating gene flow within and between catchments. Conversely, for P. flavomaculatus we found strong genetic differentiation between almost all sites, suggesting that dispersal is much more limited. 5. Greater dispersal in the patchily distributed P. conspersa than in P. flavomaculatus, which occupies larger and presumably more persistent habitats, could be a general feature of other similarly distributed aquatic insects. While higher relief is potentially a partial barrier to dispersal, P. conspersamust have effective gene flow through such apparently inhospitable terrain, perhaps attributable to dispersal between neighbouring small and ephemeral populations. Indeed, its exploitation of headwaters and seeps requires the ability to disperse between such sites. Apparently it cannot, however, overcome more continuous barriers, consisting of large tracts of landscape with few habitable larval sites. Such landscapes, including those created by humans, may have a stronger effect on population connectivity and colonization in the longer term.  相似文献

A mechanistic model was developed, to simulate the main facets of photoinhibition in phytoplankton. Photoinhibition is modelled as a time dependent decrease in the initial slope of a photosynthesis versus irradiance curve, related to D1 (photosystem II reaction centre protein) damage and non-photochemical quenching. The photoinhibition model was incorporated into an existing ammonium-nitrate nutrition interaction model capable of simulating photoacclimation and aspects of nitrogen uptake and utilization. Hence the current model can simulate the effects of irradiance on photosynthesis from sub-saturating to inhibitory photon flux densities, during growth on different nitrogen sources and under nutrient stress. Model output conforms well to experimental data, allowing the extent of photoinhibition to be predicted under a range of nutrient and light regimes. The ability of the model to recreate the afternoon depression of photosynthesis and the enhancement of photosynthesis during fluctuating light suggests that these two processes are related to photoinhibition. The model may be used to predict changes in biomass and/or carbon fixation under a wide range of oceanographic situations, and it may also help to explain the progression to dominance of certain algal species, and bloom formation under defined irradiance and nutrient conditions.  相似文献

Seven polymorphic microsatellite markers suitable for population genetic studies and genetic mapping were developed for Leptosphaeria maculans, a fungal pathogen of canola (Brassica napus). Polymorphism was evaluated using 14 isolates from diverse geographical locations. Each locus had either two or three alleles. Cross‐species amplification was observed for almost all loci in L. biglobosa ‘brassicae’ and L. maculans ‘lepidii’.  相似文献

1. Changes in nutrients and climate have occurred over approximately the same timescales in many European lake catchments. Here, we attempt to interpret the sedimentary diatom record of a large shallow lake, Loch Leven, in relation to these pressures using information gained from analysis of long‐term data sets of water quality, climate and planktonic diatoms. 2. The core data indicate the enrichment of Loch Leven starting in c. 1800–1850, most likely from agricultural practices in the catchment, with a more marked phase since c. 1940–1950 caused by increased phosphorus inputs from sewage treatment works, land drainage and a woollen mill. 3. While the recent diatom plankton remains are dominated by taxa associated with nutrient‐rich conditions, an increase in Aulacoseira subarctica relative to Stephanodiscus taxa since the mid‐1980s suggests that reductions in external catchment sources of nutrients (since 1985) may have resulted in partial recovery. This observation accords well with the long‐term monitoring series of water chemistry and phytoplankton. 4. On a decadal‐centennial scale, the eutrophication signal in the sediment record outweighs any evidence of climate as a control on the diatom community. However, at an inter‐annual scale, while the diatom data exhibit high variability, there are several changes in species composition in the recent fossil record that may be attributed to climatic controls. 5. The study highlights the value of a palaeolimnological approach, particularly when coupled with long‐term data sets, for developing our understanding of environmental change at a range of temporal scales. The diatom record in the sediment can be used effectively to track recovery from eutrophication, but requires greater understanding of contemporary ecology to fully interpret climate impacts. 6. The study illustrates the complexity of ecosystem response to synchronous changes in nutrients and climate, and the difficulty of disentangling the effects of these multiple, interacting pressures.  相似文献
