Alternaria alternata is a common fungal parasite on fruits and other plants and produces a number of mycotoxins, including alternariol (3,7,9-trihydroxy-1-methyl-6H-dibenzo [b,d]pyran-6-one), alternariol monomethyl ether (3,7-dihydroxy-9-methoxy-1-methyl-6H-dibenzo[b,d]pyran-6-one), and the mutagen altertoxin I {[1S-(1α,12aβ,12bα)] 1,2,11,12,12a, 12b-hexahydro-1,4,9,12a-tetrahydroxy-3,10-perylenedione}. Alternariol and alternariol monomethyl ether have previously been detected in some samples of fruit beverages. Stability studies of these toxins as well as altertoxin I added to fruit juices and wine (10–100 ng/mL) were carried out. To include altertoxin I in the analysis, cleanup with a polymer-based Varian Abselut solid phase extraction column was used, as recoveries from C-18 columns were low. The stabilities of alternariol and alternariol monomethyl ether in a low acid apple juice containing no declared vitamin C were compared with those in the same juice containing added vitamin C (60 mg/175 ml); there were no apparent losses at room temperature over 20 days or at 80°C after 20 min. in either juice. Altertoxin I was moderately stable in pH 3 buffer (75% remaining after a two week period). Furthermore, altertoxin I was stable or moderately stable in three brands of apple juice tested over 1–27 day periods and in a sample of red grape juice over 7 days. It is concluded that altertoxin I is sufficiently stable to be found in fruit juices and should be included in methods for alternariol and alternariol monomethyl ether.  相似文献

Aphides ( Capitophorus fragariae ) were fed for periods of up to 24 hr. on strawberry plants infected with mild crinkle, severe crinkle or yellow-edge and then transferred to plants of the wild strawberry, Fragaria vesca , or of the cultivated strawberry, variety Royal Sovereign. On F. vesca the symptoms produced were chlorotic speckling, distortion and dwarfing of the leaves, varying in intensity', and on Royal Sovereign scattered, inconspicuous, diffuse, chlorotic spots.
The symptoms from all three sources of infection were similar and were indistinguishable from those of mild crinkle of Harris & King. The virus thus selectively transmitted is tentatively concluded to be the mild crinkle virus.
The virus was transmitted after feeding periods of 1 hr. or more and did not generally persist in the vector for more than 3 hr.  相似文献

ATPase activity has been localized at an ultrastructural levelin the absorptive region of the scutella of germinating barley(Hordeum vulgare L.). The enzyme is localized on the plasmalemmaof the epithelial cells. Using the Gomori reaction the depositionof reaction product on the plasmalemma, which is dependent uponthe presence of supplied ATP, was precluded or reduced by theinhibitors orthovanadate, mercuric chloride and DCCD, whilstß-glycerophosphate would not act as an alternativesubstrate. The mitochondria demonstrated phosphatase activitywith both ATP and ß-glycerophosphate as substrate.The results are discussed in relation to the active uptake ofmetabolites by the scutellum during germination and the structuralmodification of the plasmalemma of the epithelial cells to formplasmatubules. This is the first study to demonstrate significant differences between two natural Trinidadian guppy populations in susceptibility to a pathogenic monogenean parasite, Gyrodactylus turnbulli . Following experimental infection with an isogenic laboratory culture of G. turnbulli , fish from the Upper Aripo (UA) lost parasites more slowly and carried up to three times as many parasites at peak infection than did those from the Lower Aripo (LA). The UA population appeared to be more susceptible than the LA fish, even though fish of both populations were naïve to this particular laboratory strain of G. turnbulli and had not encountered any gyrodactylid infection for at least 65 days. The parasite infection reduced the feeding response and feeding activity of UA and LA fish to a similar extent. Our findings suggest that this ectoparasite may have an important impact on the evolutionary biology of guppies (and possibly of other teleosts), particularly as the prevalence of Gyrodactylus infection in natural populations may be as high as 50%.
