The genus Avena contains five different chloroplast genomes, I-V. A physical map of chloroplast (ct) DNA of Avena sativa (type I chloroplast genome) was constructed using three restriction endonucleases, PstI, SalI and SmaI. This genome is ca. 135.5 kbp in size, and contains two inverted repeats of ca. 22.5 kbp each, separated by a large (ca. 79.0 kbp) and small (ca. 12.5 kbp) single copy region. The rbcL gene which codes for the large subunit of ribulose 1,5-bisphosphate carboxylase, was located in the map. Restriction fragment patterns of all five chloroplast genomes were compared, and among them five fragment size and five restriction site mutations were disclosed. Four site mutations were found in two or more chloroplast genomes, the other site and five fragment size mutations were specific to one or another of the chloroplast genomes. A dendrogram showing phylogenetic relationships among the five chloroplast genomes, based on the distribution of the common and specific mutations among them, indicates that chloroplast genome divergence characterized by three restriction site mutations occurred first between two diploid groups, each carrying A and C genome (nuclear), respectively, followed by further speciation in each group.  相似文献

Absorption and magnetic circular dichroism (MCD) spectra are reported for chlorophyll (Chl) a and Chl b dissolved in nematic liquid crystal solvents. The spectra were measured with the dye molecules oriented uniaxially along the direction of. the magnetic field and measuring light beam. It is significant that under such conditions the MCD spectra recorded in the wavelength region of the Q and Soret bands of the chlorophyll are essentially unchanged with respect to rotation of the sample cell around this axis, even though there is almost complete orientation of the chlorophyll molecules by the liquid crystals. The MCD spectra of Chl a and b in the nematic liquid crystal solvents used in this study are surprisingly similar to the spectra obtained under isotropic conditions. These results illustrate an important technique with which to examine the optical spectra of dyes oriented in liquid crystal matrices in which the anisotropic effects can be reduced the negligible proportions by the application of a strong magnetic field parallel to the direction of the measuring light beam. The first deconvolution calculations are reported that describe the deconvolution of pairs of absorption and MCD spectra, in the Q and B band regions, for both Chl a and b. The spectral analysis to obtain quantitative estimates of transition energies was accomplished by carrying out detailed deconvolution calculations in which the both the absorption and MCD spectral envelopes were fitted with the same number of components; each pair of components had the same hand centres and bandwidth values. This procedure resulted in an assignment of each of the main transitions in the absorption spectra of both Chl a and b. Chl a is clearly monomeric, with Qy, Qx, By and Bx located at 671, 582, 439 and 431 nm, respectively. Analysis of the spectral data for Chl b located Qy, By and Bx, at 662, 476 and 464 nm, respectively.  相似文献

A band-type alternating pattern of acidic and alkaline regions formed along the Characean cell wall is discussed theoretically. The model system is constructed from linear diffusion equations for the concentration of H+ outside the internode and in the protoplasm. The plasmalemma is taken as a boundary transporting H+ under energy supply by light. The sizes of the protoplasm and extracellular water phase are taken into account explicitly in the present model system to reproduce qualitatively the characteristics observed in various types of experiments. Theoretical analysis shows that the band pattern belongs to dissipative structures emerging far from equilibrium, and is stabilized through the electric current loops produced by locally activated electrogenic H+ pumps and spatially separated passive H+ influx (or OH- efflux) across the membrane. Both the numerical calculation and the theoretical analysis using a generalized time-dependent Ginzburg-Landau equation reveal the following points: (i) the intemodal cell with a larger vacuole in a smaller size of the extracellular water phase tends to exhibit a clearer band pattern; (ii) the increase in viscosity of the external aqueous medium makes the bands appear more easily and, furthermore, distinctly; (iii) the change in size of the extracellular water phase significantly affects the kinetics of the pattern- formation process. These results are interpreted reasonably by taking account of the electric current circulating between the acidic and alkaline regions.  相似文献

用L-赖氨酸产生株纯齿棒状杆菌PI-3-2(Hsc-,AEC+)在8L自控发酵小罐上,用恒稀释率指数递增方式连续补加葡萄糖液进行L-赖氨酸分批发酵的研究。结果表明,一次投糖分批发酵时,较高糖浓度使比产酸速率Qp值下降,不能有效地提高产酸水平。采用连续补糖方式可以改变菌体竞争底物的能力或改善代谢途径,增大耗底物分数a2或真正产酸率yp,;从而增加表观产酸率Yp值,提高葡萄糖转化率。此方式的发酵属Caden动力学分类第I型,在发酵的中后期控制H等条件,可增加比产酸速率Qp值,提高发酵水平。PI-3-2菌株的产酸水平可由47.Mg/ml提高到64.2mg/ml(总糖浓度18.18%时),避离可达73.3mg/m1(总糖浓度22.73mg/ml时)。  相似文献

The activity of superoxide dismutase was investigated in needles of spruce trees. To obtain maximum activity, needles were homogenized in the presence of Triton X-100 and polyvinylpyrrolidone. Superoxide dismutase activity was measured in dialyzed extracts with a modified epinephrine assay (HP Misra, I Fridovich [1972] J Biol Chem 247: 3170-3175) at pH 10.2. The extracts contained 70 to 120 units of superoxide dismutase per milligram protein. One unit of superoxide dismutase was completely inhibited in the presence of 20 micromolar NaCN. On native polyacrylamide gels three electromorphs were visualized after staining for activity. All three species were sensitive to CN and H2O2 and were therefore assumed to be Cu/Zn-superoxide dismutases. Superoxide dismutase activity was dependent on the age of the needles and declined by approximately 25% within 3 to 4 years.  相似文献

Consideration must be exercised in determination of buffers and solutions used when carrying out enzyme-linked immunosorbent assays (ELISAs). A commercial monoclonal antibody kit for abscisic acid (Idetek, Inc.) gives significant false-positives with tricarboxylic acid cycle intermediates. The organic acids or contaminants interfered with ELISA assays for ABA as indicated by deviations in the slopes of standard curves of ABA in the organic acids. The interference, in the case of α-ketoglutarate, was caused by a contaminant. Of the organic buffers tested—Tris, Tricine, and Hepes—only Hepes showed false-positive ABA. In addition, we present data indicating the presence of ABA in commercial mannitol and provide a simple procedure for removal of the ABA.  相似文献

Effects of red and blue light at irradiances from 1.6 to 28.3 micromolar per square meter per second on chloroplast pigments, light-harvesting pigment-proteins associated with photosystem II, and the corresponding mRNA were evaluated in maize (Zea mays L.) plants (OP Golden Bantum) grown for 14 days under 14 hours light/10 hours dark cycles. Accumulation of pigments, pigment-proteins, and mRNA was less in blue than in red light of equal irradiance. The difference between blue and red light, however, varied as a function of irradiance level, and the pattern of this variation suggests irradiance-controlled activation/deactivation (switching) of blue-light receptor. The maximum reduction in blue light of mRNA and proteins associated with light-harvesting complex occurs at lower irradiance levels than the maximum reduction of chlorophylls a and b.  相似文献

Photosynthetic characteristics of four high-CO2-requiring mutants of Chlamydomonas reinhardtii were compared to those of wild type before and after a 24-hour exposure to limiting CO2 concentrations. The four mutants represent two loci involved in the CO2-concentrating system of this unicellular alga. All mutants had a lower photosynthetic affinity for inorganic carbon than did the wild type when grown at an elevated CO2 concentration, indicating that the genetic lesion in each is expressed even at elevated CO2 concentrations. Wild type and all four mutants exhibited adaptive responses to limiting CO2 characteristic of the induction of the CO2-concentrating system, resulting in an increased affinity for inorganic carbon only in wild type. Although other components of the CO2-concentrating system were induced in these mutants, the defective component in each was sufficient to prevent any increase in the affinity for inorganic carbon. It was concluded that the genes corresponding to the ca-1 and pmp-1 loci exhibit at least partially constitutive expression and that all components of the CO2-concentrating system may be required to significantly affect the photosynthetic affinity for inorganic carbon.  相似文献

Tall fescue (Festuca arundinacea Schreb.) leaf blades elongated 33% faster at continuous low than at continuous high irradiance (60 versus 300 micromoles per second per square meter photosynthetic photon flux density) when temperature of the leaf elongation zone was held constant at 21°C. Increased rate of elongation was associated with a near proportional increase in length of the elongation zone (+38%). In contrast, growth in width and thickness was decreased at low irradiance, resulting in only a 12% increase in leaf area production and 5% less total growth-associated water deposition than at high irradiance. At low irradiance dry matter (DM) import into the elongation zone was 28% less, and 55% less DM was used per unit leaf area produced. DM use in synthesis of structural components (i.e. DM less water-soluble carbohydrates) was only 13% less at low irradiance, whereas water-soluble carbohydrates (WSC) deposition was 43% less. The lower rate of WSC deposition at low irradiance was associated with a higher net rate of monosaccharide deposition (+39%), whereas net deposition rates for sucrose (−27%) and fructan (−56%) were less than at high irradiance. Still, at low irradiance, net fructan accumulation accounted for 64% of WSC deposition, i.e. 25% of DM import, demonstrating the high sink strength of the leaf elongation zone.  相似文献
