We studied the dragonfly fauna along a 15 km stretch of thefloodplain of the regulated, first-order river Tisza, Hungary. Data on capturedand observed adults, larvae and exuvia were recorded. Observations were madefrom May to October 1998 and 1999 on 34 species, which is 52% of theHungarian fauna. Classification on the basis of faunistic similarity revealedthat habitat-level differences, associated with various degrees of physicalanthropogenic impact on bank vegetation, can exceed variation among the waterbodies of different types (backwater, pond, marsh, canal, river). The richestsites were dominated by structurally diverse macrophyte vegetation, while flowing waters (river and canal) were poorest in species. Odonata were found tobe reliable indicators of small-scale habitat patterns, reflecting vegetationdifferences even within single water bodies where the sampling spots werearranged just a few hundred metres apart. Along a gradient of utilisationintensity, the species number of Odonata assemblages and the summed relativeabundance of the five rarest species of the study area decreased linearly withincreasing fragmentation of the marginal vegetation. Sports fishermanactivities, implying disruption of the littoral marsh zone by establishment ofclearings and excessive trampling of the banks, can also be monitored bydragonfly faunistic investigations. Our results demonstrate that conservation ofthese varied floodplain water bodies requires the control of sports fishingactivity, suggesting that (i) to maintain the representative odonate fauna ofthe water bodies, some non-fragmented shores must be provided; and (ii) permanentfishing stands should not exceed 8 m mean width and should beseparated by at least 12 m of intact riparian sections.  相似文献

In this paper we describe the identification of a gene, MsDWF1 coding for a putative gibberellin 3-beta-hydroxylase (GA3ox), whose natural mutation is conditioning a dwarf growth phenotype in Medicago sativa. The dwarf phenotype could not be complemented with grafting, which indicates that the bioactive gibberellin compound necessary for shoot elongation is immobile. On the contrary, exogenously added gibberellic acid restored normal growth. The genetic position of the Msdwf1 gene was mapped to linkage group 2 (LG2) and the physical location was delimited by map-based cloning using Medicago truncatula genomic resources. Based on the similar appearance and behavior of the dwarf Medicago sativa plants to the pea stem length mutant (le) as well as the synthenic map position of the two genes it was postulated that MsDWF1 and pea Le are orthologs. The comparison of wild type and mutant allele sequences of MsGA3ox revealed an amino acid change in a conserved position in the mutant allele, which most probably impaired the function of the enzyme. Our results indicate that the dwarf phenotype was the consequence of this mutation.  相似文献

Aggregates of living cells (i.e. model tissue fragments) under appropriate conditions fuse like liquid drops. According to Steinberg's differential adhesion hypothesis (DAH), this may be understood by assuming that cells are motile and tissues made of such cells possess an effective surface tension. Here we show that based on these properties three-dimensional cellular structures of prescribed shape can be constructed by a novel method: cell aggregate printing. Spherical aggregates of similar size made of cells with known adhesive properties were prepared. Aggregates were embedded into biocompatible gels. When the cellular and gel properties, as well as the symmetry of the initial configuration were appropriately adjusted the contiguous aggregates fused into ring-like organ structures. To elucidate the driving force and optimal conditions for this pattern formation, Monte Carlo simulations based on a DAH motivated model were performed. The simulations reproduced the experimentally observed cellular arrangements and revealed that the control parameter of pattern evolution is the gel-tissue interfacial tension, an experimentally accessible parameter.  相似文献

A complete set of mono-, di- and triphosphorylated peptides comprising amino acids 10-27, the Mdm2 and p300 binding site(s) of p53, with and without a fluorescein label at the N-terminus, was synthesized by step-by-step solid phase synthesis. Fluorescence polarization analysis revealed that phosphorylation at Thr18 decreased binding to recombinant Mdm2 protein compared with the unphosphorylated and the two other single phosphorylated analogues. Unlabelled multiply phosphorylated peptides corresponding to this amino-terminal transactivation domain proved to be powerful tools in analysing the phosphate specificity of existing anti-p53 monoclonal and polyclonal antibodies using direct ELISA. The tetramerization domain of human p53 protein was modelled with a 53 residue-long unlabelled unphosphorylated and Ser315-phosphorylated peptide pair. CD analysis showed similar alpha-helical structures for both peptides and no major difference in the secondary structure could be observed upon phosphorylation. Size-exclusion HPLC indicated that these synthetic oligomerization domain mimics underwent a pH-dependent tetramerization process, but the presence of a phosphate group at Ser315 did not modify the oligomeric state of the 308-360 p53 fragments. Nevertheless, the fluorescein-labelled Ser315 phosphorylated peptide bound to the downstream signalling ligand DNA topoisomerase I protein with slightly higher affinity than did the unphosphorylated analogue.  相似文献

Summary Converging lines of evidence suggest that theN-methyl-d-aspartic acid (NMDA) antagonist properties of conantokin G (ConG) are mediated through a novel polyamine-associated site. Moreover, structural modification of the heptadecapeptide yields peptides that can either mimic the NMDA antagonist properties of the parent peptide or produce polyamine-like actions at NMDA receptors. We synthesized a panel of ConG fragments and evaluated their effects using a neurochemical assay that predicts pharmacological actions at NMDA receptors. While the C-terminal tetrapeptide elicited a polyamine-like activation of [3H]MK-801 binding with a potency comparable to spermine, the N-terminal pentapeptide produced a marginal inhibition of spermine-enhanced [3H]MK-801 binding. These observations suggest that the parent peptide interacts with two distinct sites on NMDA receptors. In contrast, amino acid replacements in the middle region of ConG resulted in analogues that were of comparable or greater potency than the parent peptide. The Ala7, Tyr10 derivative is of particular interest since it is a potent inhibitor (IC50≈80 nM) of spermine-enhanced [3H]MK-801 binding, and may thus serve as a precursor for studies designed to125I-label putative ConG binding sites. Our observations are also consistent with the hypothesis that the termini of ConG are essential for an interaction with NMDA receptors, while the middle region of this peptide serves as a spacer unit. This hypothesis is consonant with spectroscopic evidence that ConG possesses a central rigid helical backbone with flexible N- and C-terminal regions. Nonetheless, ConG variants in which the termini were connected with conformationally stabilized α-or 310-helical spacers grew progressively less potent as NMDA antagonists as the structural stability of these peptides increased. Thus, the middle region of ConG appears to possess functions other than providing conformational stability. These newly synthesized ConG derivatives may serve as a basis for the design of novel peptide or peptidomimetic agents.  相似文献

1. The volume fraction of autophagic vacuoles in liver parenchymal and exocrine pancreatic cells was smallest and the serum insulin level highest in the 24 hr prestarved mouse immediately after 3 hr feeding period. 2. The size of the autophagic vacuole and lysosome (dense body) compartments increased in both types of cells during 2-72 hr fasting parallel with decreasing serum insulin levels. 3. The protein content of the cells decreased and the DNA-based activity of acid phosphatase showed little change throughout fasting. The activity of cathepsin D increased during days 2 and 3 of food deprivation. 4. Vinblastine (50 mg/kg body wt) applied for the last 2 hr of different periods (2, 12, 24, 48 and 72 hr) of fasting decreased serum insulin level and increased the fractional cytoplasmic volume of autophagic vacuoles and dense bodies. This increase was smaller when the drug was applied shortly after feeding and much larger after prolonged fasting. The increase was more pronounced in the pancreatic than in the liver cells. 5. Our data show that the effect of vinblastine on the size of the autophagic-lysosomal compartment depends on the feeding status of the animals.  相似文献

The aromatase (estrogen synthetase) enzyme catalyzes the conversion of androgens to estrogens in peripheral tissues, as well as in the brain. Our study aimed at comparing the brain distribution of aromatase-immunoreactive neurons in male and female, normal and gonadectomized rats. Light microscopic immunostaining was employed using a purified polyclonal antiserum raised against human placental aromatase. Two anatomically separate aromatase-immunoreactive neuronal systems were detected in the rat brain: A “limbic telencephalic” aromatase system was composed by a large population of labeled neurons in the lateral septal area, and by a continuous “ring” of neurons of the laterodorsal division of the bed nucleus of stria terminalis, central amygdaloid nucleus, stria terminalis, and the substantia inominata-ventral pallidum-fundus striati region. The other, “hypothalamic” aromatase system consisted of neurons scattered in a dorsolateral hypothalamic area including the paraventricular, lateral and dorsomedial hypothalamic nuclei, the subincertal nucleus as well as the zona incerta. In addition, a few axon-like processes (unresponsive to gonadectomy) were present in the preoptic-anterior hypothalamic complex, the ventral striatum, and midline thalamic regions. No sexual dimorphism was observed in the distribution or intensity of aromatase-immunostaining. However, 3 days, 2, 3, 8, 16, or 32 weeks after gonadectomy, aromatase-immunoreactive neurons disappeared from the hypothalamus, whereas they were still present in the limbic areas of both sexes. The results indicate the existence of two distinct estrogen-producing neuron systems in the rat brain: (1) a “limbic ring” of aromatase-labeled neurons of the lateral septum-bed nucleus-amygdala complex unresponsive to gonadectomy; and (2) a sex hormone-sensitive “hypothalamic” aromatase neuron system.  相似文献
