Species extinctions and declines are occurring globally and commonly have cascading effects on ecosystems. In Australia, mammal extinctions have been extensive, particularly in arid areas, where precipitation drives ecosystems. Many ecologically extinct mammals feed on soil‐dwelling insects. However, how this top‐down pressure affected their prey and how this contrasts with the bottom‐up impacts of fluctuating precipitation remains unclear. We constructed a long‐term exclusion experiment in a multi‐species mammal reintroduction zone in semi‐arid Australia to test how top‐down (reintroduced mammals) and bottom‐up (precipitation) factors affect root‐feeding chafer beetles (Coleoptera: Melolonthinae). We used emergence traps in ten replicate 20 × 20 m plots of control, exclusion and procedural control treatments to trap chafers biannually from 2009 to 2015. Annual precipitation during this period varied from 173 to 481 mm. Mammal exclusion did not affect chafers, indicating that top‐down regulation was not important. Instead, chafer abundance, species density and biomass increased with precipitation. Chafer body size and assemblage composition were best predicted by sampling year, suggesting that random drift determined species abundances. Increased resource availability therefore favoured all species similarly. We thus found no evidence that mammal predation alters chafer populations and conclude that they may be driven primarily by bottom‐up processes. Further research should determine if the cascading effects of species loss are less important for herbivores generally than for higher level trophic groups and the role of ecosystem stability in mediating these patterns.  相似文献

Summary The terminal parts of radially directed neurite bundles growing out from chick embryo spinal cord in vitro have been examined by phase and electron-microscopy,A type of ending is described in which the terminal parts of the neurites are associated with a glial cell. The latter sends a single major process proximally towards the explant. Distally it is attached to the substrate, and the neurite ends are related to its dorsal (nonsubstrate) aspect. Appearances suggesting a mechanism of adhesion of neurites to each other and to the gial cell are described.Growth vesicles were found in both neurites and glia.It is suggested that movements of terminal glial cells may affect the pattern of outgrowth of their attached neurite bundles.We are grateful to the Medical Research Council for financial assistance, to Mr. A. The neotenous salamander Ambystoma mexicanum reaches sexual maturity without completing metamorphosis. Females must, therefore, synthesize vitellogenin, the precursor of the egg-yolk proteins. We show that livers of female axolotls synthesize and secrete a phosphoprotein which migrates with Xenopus vitellogenin on SDS-gels and is precipitated by antibody prepared against Xenopus vitellogenin. The livers of male axolotls do not normally synthesize this protein but can be induced to do so by treatment in vivo with estradiol. A receptor with a high affinity for estradiol (Kd = 0.3 × 10?9M) was found in the nuclei prepared from livers of male and female axolotls. It sediments at 3.7 S at 0°C in sucrose gradients containing 0.5 M KCl. Each nucleus contains about 1300 binding sites for estradiol, 13 times the number found in normal male Xenopus nuclei, but as axolotl nuclei are about 12 times larger, the concentrations of binding sites are similar. In contrast to Xenopus, there is no detectable increase in the number of nuclear binding sites following estrogen treatment. We conclude that the controls affecting both the appearance of vitellogenin inducibility and the induction of vitellogenin synthesis differ between the two species A. mexicanum and Xenopus laevis.  相似文献

In order to assess the clinical importance of R factors 524 “coliform” infections were studied in a general hospital. Of these, 95 were caused by Pseudomonas aeruginosa. Among the remaining 429, 43% were caused by drug-resistant enterobacteria; resistance was frequently multiple and determined by transmissible R factors. Choice of therapy was thus limited but in every case at least one antibiotic, effective in vitro, was still available.  相似文献

Homozygous mutation of the Csf1r locus (Csf1rko) in mice, rats and humans leads to multiple postnatal developmental abnormalities. To enable analysis of the mechanisms underlying the phenotypic impacts of Csf1r mutation, we bred a rat Csf1rko allele to the inbred dark agouti (DA) genetic background and to a Csf1r-mApple reporter transgene. The Csf1rko led to almost complete loss of embryonic macrophages and ablation of most adult tissue macrophage populations. We extended previous analysis of the Csf1rko phenotype to early postnatal development to reveal impacts on musculoskeletal development and proliferation and morphogenesis in multiple organs. Expression profiling of 3-week old wild-type (WT) and Csf1rko livers identified 2760 differentially expressed genes associated with the loss of macrophages, severe hypoplasia, delayed hepatocyte maturation, disrupted lipid metabolism and the IGF1/IGF binding protein system. Older Csf1rko rats developed severe hepatic steatosis. Consistent with the developmental delay in the liver Csf1rko rats had greatly-reduced circulating IGF1. Transfer of WT bone marrow (BM) cells at weaning without conditioning repopulated resident macrophages in all organs, including microglia in the brain, and reversed the mutant phenotypes enabling long term survival and fertility. WT BM transfer restored osteoclasts, eliminated osteopetrosis, restored bone marrow cellularity and architecture and reversed granulocytosis and B cell deficiency. Csf1rko rats had an elevated circulating CSF1 concentration which was rapidly reduced to WT levels following BM transfer. However, CD43hi non-classical monocytes, absent in the Csf1rko, were not rescued and bone marrow progenitors remained unresponsive to CSF1. The results demonstrate that the Csf1rko phenotype is autonomous to BM-derived cells and indicate that BM contains a progenitor of tissue macrophages distinct from hematopoietic stem cells. The model provides a unique system in which to define the pathways of development of resident tissue macrophages and their local and systemic roles in growth and organ maturation.  相似文献

Over the last 40 years, new sunflower downy mildew isolates (Plasmopara halstedii) have overcome major gene resistances in sunflower, requiring the identification of additional and possibly more durable broad‐spectrum resistances. Here, 354 RXLR effectors defined in silico from our new genomic data were classified in a network of 40 connected components sharing conserved protein domains. Among 205 RXLR effector genes encoding conserved proteins in 17 P. halstedii pathotypes of varying virulence, we selected 30 effectors that were expressed during plant infection as potentially essential genes to target broad‐spectrum resistance in sunflower. The transient expression of the 30 core effectors in sunflower and in Nicotiana benthamiana leaves revealed a wide diversity of targeted subcellular compartments, including organelles not so far shown to be targeted by oomycete effectors such as chloroplasts and processing bodies. More than half of the 30 core effectors were able to suppress pattern‐triggered immunity in N. benthamiana, and five of these induced hypersensitive responses (HR) in sunflower broad‐spectrum resistant lines. HR triggered by PhRXLRC01 co‐segregated with Pl22 resistance in F3 populations and both traits localized in 1.7 Mb on chromosome 13 of the sunflower genome. Pl22 resistance was physically mapped on the sunflower genome recently sequenced, unlike all the other downy mildew resistances published so far. PhRXLRC01 and Pl22 are proposed as an avirulence/resistance gene couple not previously described in sunflower. Core effector recognition is a successful strategy to accelerate broad‐spectrum resistance gene identification in complex crop genomes such as sunflower.  相似文献

Bees are model organisms for the study of learning and memory, yet nearly all such research to date has used a single reward, nectar. Many bees collect both nectar (carbohydrates) and pollen (protein) on a single foraging bout, sometimes from different plant species. We tested whether individual bumblebees could learn colour associations with nectar and pollen rewards simultaneously in a foraging scenario where one floral type offered only nectar and the other only pollen. We found that bees readily learned multiple reward–colour associations, and when presented with novel floral targets generalized to colours similar to those trained for each reward type. These results expand the ecological significance of work on bee learning and raise new questions regarding the cognitive ecology of pollination.  相似文献

Our understanding of the role of freshwaters in the global carbon cycle is being revised, but there is still a lack of data, especially for the cycling of methane, in rivers and streams. Unravelling the role of methanotrophy is key to determining the fate of methane in rivers. Here we focus on the carbon conversion efficiency (CCE) of methanotrophy, that is, how much organic carbon is produced per mole of CH4 oxidised, and how this is influenced by variation in methanotroph communities. First, we show that the CCE of riverbed methanotrophs is consistently high (~50%) across a wide range of methane concentrations (~10–7000 nM) and despite a 10-fold span in the rate of methane oxidation. Then, we show that this high conversion efficiency is largely conserved (50%± confidence interval 44–56%) across pronounced variation in the key functional gene (70 operational taxonomic units (OTUs)), particulate methane monooxygenase (pmoA), and marked shifts in the abundance of Type I and Type II methanotrophs in eight replicate chalk streams. These data may suggest a degree of functional redundancy within the variable methanotroph community inhabiting these streams and that some of the variation in pmoA may reflect a suite of enzymes of different methane affinities which enables such a large range of methane concentrations to be oxidised. The latter, coupled to their high CCE, enables the methanotrophs to sustain net production throughout the year, regardless of the marked temporal and spatial changes that occur in methane.  相似文献
