Comparisons between the 60C γ-ray survival curves of diploid strains of the yeast Saccharomyces cerevisiae that are homozygous for two non-allelic radition-sensitive mutations and the corresponding single-mutant diploids suggest that there are two main types of repair of ionizing radiation damage in this organism. The first, which is defined by the rad52 epistasis groups, depends on the activities of the RAD50 through RAD57 genes and is responsible for repairing the larger amount of lethal damage. Previous work [22] shows that this type of repair is essentially error-free. The second, defined by the rad6 epistasis group, depends on the activities of the RAD6, RAD9, RAD18, REV1 and REV3 genes and repairs a smaller, though still substantial, amount of lethal damage. It is also responsible for induced mutagenesis [22,23]. Data for survival and mutation induction after irradiation in air and partial anoxia show that oxygen-dependent damage can be repaired by either of these two pathways. They also show similar oxygen-enhancement ratios for survival and mutagenesis.  相似文献

In a study of 2,149 emergency admissions because of haematemesis or melaena during a 15-year period, the sex ratio, age distribution, and main diagnostic groups showed no major change. Various factors affected the prognosis, such as the age of the patient, the underlying diagnosis, a low blood pressure on arrival at hospital, gross anaemia on arrival there, and the pattern of bleeding after admission. The fatality rate remained virtually constant throughout the period studied in spite of changes in diagnostic methods and management. There was a changing pattern in the type of operation performed in the treatment of bleeding peptic ulcer. Vagotomy combined with a drainage procedure and with a direct surgical attack on the bleeding point became more widely used at the expense of Polya or Billroth I partial gastrectomy and gave the best results. It is at first paradoxical that improved surgical results should not be reflected in a general improvement in the fatality rate, but this finding can be explained by the smaller proportion of patients treated by emergency surgery in the later years of the period studied. It is concluded that emergency surgery should be performed more frequently and that vagotomy plus drainage is the operation of choice in the peptic ulcer group.  相似文献

The stability of the outer-membrane proteins and antigens of a strain of Bacteroides intermedius (VPI 8944 group genotype II) grown in continuous culture at varying pH and growth rates (D = 0.025-0.2 h-1, pH 6.0-7.3) has been measured. The membranes showed nine major proteins (greater than 67-19.55 kilodaltons) and six major antigens (65-28 kilodaltons). Membrane proteins and antigens were stable under the conditions tested; the major proteins were detected in all membranes, and the antigen profiles tested with different antisera showed maximum similarities of 82-95%. Differences did occur in the amounts of membrane proteins synthesized; cells at high growth rates and those growing on the surfaces in the chemostat showed increased amounts of two proteins (40 and 32 kilodaltons) and possibly novel proteins of 24 and 25 kilodaltons. In addition, these membranes reflected increased synthesis or a change to increased reactivity of antigens between 20.5 and 24 kilodaltons. The results indicate stability of the expression of outer-membrane proteins and antigens in environments of differing pH and under different growth rates. However, the amount of these molecules synthesized can vary, and increases in certain proteins and antigens occur as the growth rate increases and the organisms grow on surfaces.  相似文献

Soil redox potentials and pore water sulfide concentrations on a mangrove island in the Belizean barrier reef system were significantly correlated with the presence of the aerial roots of mangrove trees. Sulfide concentrations were three to five times lower near the prop roots of Rhizophora mangle (red mangrove) and the pneumatophores of Avicennia germinans (black mangrove) than in adjacent (≤ 1 meter away) unvegetated sediment. Soil redox potentials were also significantly higher near the aerial roots. A comparison of the two species revealed that sulfide concentrations in the rhizosphere of R. mangle were as low as that of A. germinans. However, sulfide concentrations in areas occupied by the black mangrove were variable and a function of pneumatophore density. The occurrence of an oxidized rhizosphere around the roots of both species suggests that the adult trees are equally capable of exploiting reduced sediments as long as their respective pathways for root aeration are functional.  相似文献

A sensitive, quantitative assay for phosphenolpyruvate carboxylase which utilizes microtiter plates is described. The assay depends upon the production of a colored compound in the reaction between oxaloacetate, the product of the phosphoenolpyruvate reaction, and the dye Fast Violet B. The method is particularly appropriate for monitoring chromatographic eluates and its utility for this purpose is demonstrated by the detection of phosphoenolpyruvate carboxylase in fractions of crude maize extract separated by size-exclusion chromatography.  相似文献

Aim We modelled the relationship of breeding evidence for five species of forest songbirds (ruby-crowned kinglet (Regulus calendula) Blackburnian warbler (Dendroica fusca), black-throated blue warbler (Dendroica caerulescens), bay-breasted warbler (Dendrioca castanea) and Connecticut warbler (Oporornis agilis)) and a variety of macro-climate variables to examine the importance of climate as a factor determining distribution of breeding in these species and to assess the usefulness of spatial predictions generated from these models. Location Modelling was conducted over the entire province of Ontario, Canada, an area of ≈900,000 km2. Methods Data on the distribution of breeding in the province was derived from the Breeding Bird Atlas of Ontario. We used logistic regression to model the relationship between the probability of breeding (assessed in 10 km×10 km blocks) and estimates of a variety of climate variables at the same scale. Models were selected that had the least number of explanatory variables while at the same time having close to the best possible classification accuracy. Results The final models for these five species had from one to six explanatory variables and an overall concordance of 70.4% to 86.3% indicating a good classification accuracy. Results from subsampling 50% of the original data ten times indicate that (1) the classification accuracy of the model for data used to generate the model is not very sensitive to the specific observations used to generate the model (2) the classification accuracy of test data is close to the classification accuracy of the model data and (3) the classification accuracy of the test data is not dependent on the specific observations used to generate the model. We generated a spatial prediction of the probability of occurrence of each species for Ontario using the relationships defined by the logistic regression models and using 1 km gridded estimates of the necessary climate variables. These probability maps closely matched the maps of observed evidence of breeding from the Atlas. Main conclusions Although mechanisms controlling breeding distribution cannot be determined using this method, we can conclude that (1) macro-climate is an important factor directly and/or indirectly determining distribution of breeding in these species and (2) spatial predictions of probability of breeding are accurate enough to be useful in predicting probability of breeding in unsampled areas.  相似文献
