Tissue samples ofMacrocystis pyrifera from 2 sites in southern New Zealand and harvested over a period of 12 months were analysed for alginate content and composition (M:G ratio). Plants were divided into three frond classes of different length and each frond was further separated into age categories of blades and stipes (viz young, mature and old blades; mature and old stipes). Within each size class, younger blades had higher alginate content than older blades. Stipes did not show such variation with age. Alginate from younger blades and stipes had higher proportions of mannuronic acid residues than those from old blades and stipes. The range of M:G ratios for age categories of either blades or stipes from longer fronds was greater than those for smaller fronds. Alginate content and M:G ratios of stipes were always higher than for blades. The difference between M:G ratios of blades and stipes was greater for smaller fronds than for longer ones. In previousstudies, our laboratory has utilized a cell line derived from the ratinner medullary collecting duct (IMCD) as a model system for mammalianrenal epithelial cell acid secretion. We have provided evidence, from aphysiological perspective, that acute cellular acidification stimulatesapical exocytosis and elicits a rapid increase in proton secretion thatis mediated by an H+-ATPase. Thepurpose of these experiments was to examine the effect of acutecellular acidification on the distribution of the vacuolar H+-ATPase in IMCD cells in vitro.We utilized the 31-kDa subunit of theH+-ATPase as a marker of thecomplete enzyme. The distribution of this subunit of theH+-ATPase was evaluated byimmunohistochemical techniques (confocal and electron microscopy), andwe found that there is a redistribution of these pumps from vesicles tothe apical membrane. Immunoblot evaluation of isolated apical membranerevealed a 237 ± 34% (P < 0.05, n = 9) increase in the 31-kDa subunitpresent in the membrane fraction 20 min after the induction of cellularacidification. Thus our results demonstrate the presence of this pumpsubunit in the IMCD cell line in vitro and that cell acidificationregulates the shuttling of cytosolic vesicles containing the 31-kDasubunit into the apical membrane.  相似文献

The distribution of meiotic pairing sites on a Drosophila melanogaster autosome was studied by characterizing patterns of prophase pairing and anaphase segregation in males heterozygous for a number of 2-Y transpositions, collectively coveringall of chromosome arm 2R and one-fourth of chromosome arm 2L. It was found that all transpositions involving euchromatin from chromosome 2, even short stretches, increased the frequency of prophase I quadrivalents involving the sex and second chromosome bivalents above background levels. Quadrivalent frequencies were the same whether the males carried both elements of the transposition or just the Dp (2;Y) element along with two normal chromosome 2s, indicating that pairing is non-competitive. The frequency of quadrivalents was proportional to the size of the transposed region, suggesting that pairing sites are widely distributed on chromosome 2. Moreover, all but the smallest transpositions caused a detectable bias in the segregation ratio, in favor of alternate segregations, indicating that the prophase associations were effective in orienting centromeres to opposite poles. One transposition involving only heterochromatin of chromosome 2 had no effect on quadrivalent frequency, consistent with previous evidence that autosomal heterochromatin lacks meiotic pairing ability in males. One region at the base of chromosome arm 2L proved to be especially effective in stimulating quadrivalent formation and anaphase segregation, indicating the presence of a strong pairing site in this region. Seedlings of three mangrove species—Rhizophora mangle L., Avicennia germinans (L.)Stearn., and Laguncularia racemosa (L.) Gaertn. f.—were subjected to different light and nutrient regimes in two separate growth chamber experiments. At higher nutrient or light availability, relative growth rate, leaf production, and branch growth differed significantly among species in the following order: Rhizophora < Avicennia < Laguncularia. At lower nutrient or light levels, however, species’ differences were greatly minimized. Lower nutrient or light levels caused greater investment in root biomass by all species, whereas higher nutrient availability resulted in greater investment in leaf area and maximized species’ differences in total leaf area, number of leaves, and leaf area ratio. Mangrove leaves also differed among species in quantity and composition of secondary compounds that may protect seedlings against herbivores or stress factors such as excessive solar radiation. Relative amounts of condensed tannins, gallotannins, and nitrogen were significantly affected by light and nutrient regimes, but patterns of response differed among species. The results indicate that these sympatric species differ substantially in their potential for growth, acquisition of resources, stress tolerance, and susceptibility to herbivores during the seedling stage, but that these characteristics are significantly modified by availability of resources.  相似文献

Relapse to smoking occurs at higher rates in women compared with men, especially when triggered by stress. Studies suggest that sex‐specific interactions between nicotine reward and stress contribute to these sex differences. Accordingly, novel treatment options targeting stress pathways, such as guanfacine, an α2‐adrenergic receptor agonist, may provide sex‐sensitive therapeutic effects. Preclinical studies are critical for elucidating neurobiological mechanisms of stress‐induced relapse and potential therapies, but rodent models of nicotine addiction are often hindered by large behavioral variability. In this study, we used nicotine conditioned place preference to investigate stress‐induced reinstatement of nicotine preference in male and female mice, and the effects of guanfacine on this behavior. Our results showed that overall, nicotine induced significant place preference acquisition and swim stress‐induced reinstatement in both male and female mice, but with different nicotine dose‐response patterns. In addition, we explored the variability in nicotine‐dependent behaviors with median split analyses and found that initial chamber preference in each sex differentially accounted for variability in stress‐induced reinstatement. In groups that showed significant stress‐induced reinstatement, pretreatment with guanfacine attenuated this behavior. Finally, we evaluated neuronal activation by Arc immunoreactivity in the infralimbic cortex, prelimbic cortex, anterior insula, basolateral amygdala, lateral central amygdala and nucleus accumbens core and shell. Guanfacine induced sex‐dependent changes in Arc immunoreactivity in the infralimbic cortex and anterior insula. This study demonstrates sex‐dependent relationships between initial chamber preference and stress‐induced reinstatement of nicotine conditioned place preference, and the effects of guanfacine on both behavior and neurobiological mechanisms.  相似文献

1. Male golden hamsters were induced to hibernate. 2. During stimulated arousal (i) the time course of increase in deep body temperature was related to the time-course of increasing oxygen consumption and (ii) brown adipose tissue lipid content was determined at several time points. 3. In early arousal (0-30 min) more lipid was mobilised (14.6 kJ) than was utilised in metabolism (3.5 kJ). BAT lipids accounted for 88% of the total energy requirement of arousal (90 min). 4. We conclude that BAT not only supplies heat for arousal but fuel for other arousing tissues.  相似文献

Preparations of covalently closed mitochondrial DNA of rat liver contain 10-30% of molecules that are converted into relaxed circular molecules after treatment with ribonuclease. Control experiments, with covalently closed bacteriophage PM2 DNA, indicate that ribonuclease-sensitivity cannot be induced either by depurination or by incubation with reducing agents.  相似文献
