Chemical modification of proteins has been crucial in engineering protein‐based therapies, targeted biopharmaceutics, molecular probes, and biomaterials. Here, we explore the use of a conjugation‐based approach to sense alternative conformational states in proteins. Tyrosine has both hydrophobic and hydrophilic qualities, thus allowing it to be positioned at protein surfaces, or binding interfaces, or to be buried within a protein. Tyrosine can be conjugated with 4‐phenyl‐3H‐1,2,4‐triazole‐3,5(4H)‐dione (PTAD). We hypothesized that individual protein conformations could be distinguished by labeling tyrosine residues in the protein with PTAD. We conjugated tyrosine residues in a well‐folded protein, bovine serum albumin (BSA), and quantified labeled tyrosine with liquid chromatography with tandem mass spectrometry. We applied this approach to alternative conformations of BSA produced in the presence of urea. The amount of PTAD labeling was found to relate to the depth of each tyrosine relative to the protein surface. This study demonstrates a new use of tyrosine conjugation using PTAD as an analytic tool able to distinguish the conformational states of a protein.  相似文献

In central Chile, the bur beak chervil (Anthriscus caucalis M. Bieb.; Apiaceae) is an annual naturalized herb introduced from Europe at least 120 years ago. Anthriscus is distributed in vegetation formations such as sclerophyllous shrublands (locally known as “matorral”) and spiny savannas of Acacia caven (locally known as “espinal”). In matorral formations, Anthriscus grows at the edge of native woody fragments. Because these fragments are refuges where native herbs recruit, we studied the impact of Anthriscus on the diversity and survival of native forbs established in these sheltering microsites. First, we characterized the spatial distribution of Anthriscus in the matorral, sampling in different micro-habitat types. We differentiated three microhabitat types: under the canopy of a fragment, at the edge of the canopy of a fragment, and in open sites outside the fragments. A total of 40 1 × 1 m quadrates were randomly distributed in each habitat type. Inside each of them, we recorded the number of seedlings and established plants, including Anthriscus. Then we evaluated experimentally the effect of Anthriscus on diversity and evenness of the local herb assemblages. For this purpose we conducted a field trial using 34 metallic enclosures (0.5 × 0.5 m) arranged in pairs. In each pair, Anthriscus individuals were removed from one plot, the other paired plot acting as control. We periodically recorded the presence and abundance of the remanent species of herbs inside the plots, and then we characterized the species diversity and evenness over time (Shannon’s index, H′ and Pielou’s index, J′). Finally, in a second experiment we measured experimentally the presence or absence of Anthriscus against the survival of four native herb species (Bowlesia incana, Bromus berteroanus, Pectocarya linearis, and Moscharia pinnatifida). Here we used 20 0.5 × 0.5 m plots where we randomly transplanted seedlings of native herbs and Arnthiscus. Then, for each species and plot we determined their survival (%) according to the number of seedlings initially transplanted. The samplings show strong association between the presence of Anthriscus on edge habitat in the matorral. The maximum densities were noted in these microhabitat types whereas in open areas and under-fragment sites Anthriscus shows very low or null densities. At the end of the first trials, the plots with Anthriscus showed a Shannon diversity index H′ = 0.41 (±0.11 SE), while in plots without Anthriscus this value was 1.19 (±0.1 SE), both as averages. Pielou’s evenness index (J′) yielded values of 0.23 (±0.06 SE) and 0.59 (±0.04 SE) for treatments with and without Anthriscus, respectively. Similarly, the second trials shows that the survival of the four native herbs was drastically decreased in the presence of Anthriscus: by 64% for Bowlesia incana, 43% for Bromus berteroanus, 46% for Moscharia pinnatifida, and 76% for Pectocaria linearis. Our study shows that the effects of Anthriscus include an inhibition of the establishment of native plants and a decrease in their survival in edge habitats, therefore affecting the composition and diversity of the local herb layer. Thus, Anthriscus is invading a refuge habitat for native herbs in the Chilean matorral, decreasing the native herb diversity and survival.  相似文献

Lipid metabolism-related gene mutations can cause retinitis pigmentosa, a currently untreatable blinding disease resulting from progressive neurodegeneration of the retina. Here, we demonstrated the influence of adiponectin receptor 1 (ADIPOR1) deficiency in retinal neurodegeneration using Adipor1 knockout (KO) mice. Adipor1 mRNA was observed to be expressed in photoreceptors, predominately within the photoreceptor inner segment (PIS), and increased after birth during the development of the photoreceptor outer segments (POSs) where photons are received by the visual pigment, rhodopsin. At 3 weeks of age, visual function impairment, specifically photoreceptor dysfunction, as recorded by electroretinography (ERG), was evident in homozygous, but not heterozygous, Adipor1 KO mice. However, although photoreceptor loss was evident at 3 weeks of age and progressed until 10 weeks, the level of visual dysfunction was already substantial by 3 weeks, after which it was retained until 10 weeks of age. The rhodopsin mRNA levels had already decreased at 3 weeks, suggesting that reduced rhodopsin may have contributed to early visual loss. Moreover, inflammation and oxidative stress were induced in homozygous KO retinas. Prior to observation of photoreceptor loss via optical microscopy, electron microscopy revealed that POSs were present; however, they were misaligned and their lipid composition, including docosahexaenoic acid (DHA), which is critical in forming POSs, was impaired in the retina. Importantly, the expression of Elovl2, an elongase of very long chain fatty acids expressed in the PIS, was significantly reduced, and lipogenic genes, which are induced under conditions of reduced endogenous DHA synthesis, were increased in homozygous KO mice. The causal relationship between ADIPOR1 deficiency and Elovl2 repression, together with upregulation of lipogenic genes, was confirmed in vitro. Therefore, ADIPOR1 in the retina appears to be indispensable for ELOVL2 induction, which is likely required to supply sufficient DHA for appropriate photoreceptor function and survival.Subject terms: Mechanisms of disease, Developmental neurogenesis  相似文献

Complex structures can be seen associated with large areas of endoplasmic reticulum of the chromaffin cells in the viscacha adrenal medulla. These structures are cylindrical in shape and are formed by chains of globular subunits. A central lumen is observed within which there are bodies resembling chromaffin granules. Parallel cisternae of the endoplasmic reticulum are arranged surrounding these bodies. The functional significance of these structures in unknown.  相似文献

Vitamin A and its derivatives are known to enhance the immune system and affect embryogenesis. In this study, five daily subcutaneous injections of retinol palmitate (0.001 mg/kg body weight) were administered to eight female SW mice before mating. Six more weekly injections of retinol palmitate were given during pregnancy and lactation. Eight controls were similarly treated with saline. Four of the eight vitamin-A-treated females had litters, whereas seven of the eight saline-treated females had litters. Resultant litters did not differ in size or appearance. At 12 weeks of age, serum IgM and IgG1 levels were significantly higher in the progeny of vitamin-A-treated mothers before but not after immunization with a test antigen, sheep red blood cells (SRBC). This difference was not seen when other progeny were tested at the age of one year. Anti-SRBC titers did not differ in the two groups of progeny at the age of 12 weeks or one year. One-year-old progeny of vitamin-A-treated mothers weighed significantly more than did control progeny; significant enlargement of the heart, spleen, and kidneys was observed. However, organ-to-body-weight ratios did not differ significantly. In a separate experiment, adult female mice treated with varying doses of vitamin A (five daily doses of 0.0001, 0.0005, or 0.001 mg/kg body weight) showed a dose-dependent reduction of serum IgG1 and hematocrits, but no change in serum IgM levels or leukocyte counts. Resting untreated mice had IgM levels which were one-half those seen in saline-treated controls. These studies indicate that large doses of vitamin A can affect some aspects of the developing and mature murine immune system.  相似文献

The Dothideomycete Pseudocercospora fijiensis, previously Mycosphaerella fijiensis, is the causal agent of black Sigatoka, one of the most destructive diseases of bananas and plantains. Disease management depends on fungicide applications, with a major contribution from sterol demethylation‐inhibitors (DMIs). The continued use of DMIs places considerable selection pressure on natural P. fijiensis populations, enabling the selection of novel genotypes with reduced sensitivity. The hitherto explanatory mechanism for this reduced sensitivity was the presence of non‐synonymous point mutations in the target gene Pfcyp51, encoding the sterol 14α‐demethylase enzyme. Here, we demonstrate a second mechanism involved in DMI sensitivity of P. fijiensis. We identified a 19‐bp element in the wild‐type (wt) Pfcyp51 promoter that concatenates in strains with reduced DMI sensitivity. A polymerase chain reaction (PCR) assay identified up to six Pfcyp51 promoter repeats in four field populations of P. fijiensis in Costa Rica. We used transformation experiments to swap the wt promoter of a sensitive field isolate with a promoter from a strain with reduced DMI sensitivity that comprised multiple insertions. Comparative in vivo phenotyping showed a functional and proportional up‐regulation of Pfcyp51, which consequently decreased DMI sensitivity. Our data demonstrate that point mutations in the Pfcyp51 coding domain, as well as promoter inserts, contribute to the reduced DMI sensitivity of P. fijiensis. These results provide new insights into the importance of the appropriate use of DMIs and the need for the discovery of new molecules for black Sigatoka management.  相似文献

This study investigated the usefulness of macrophage size determinations in lymphocyte-rich pleural effusions to improve the cytologic diagnosis of tuberculous pleurisy. The size of pleural macrophages was analyzed by quantitative morphometric planimetry in 18 effusions due to tuberculosis, 21 effusions following radiotherapy for malignant disease and 10 effusions due to congestive heart failure. Macrophages were identified and clearly separated from mesothelial cells by latex phagocytosis and immunostaining with the monoclonal antibody My4 (CD14). The mean macrophage area (+/- standard deviation) in tuberculous effusions (92 +/- 14 sq micron) was significantly smaller than in postradiation (141 +/- 28 sq micron) and heart-failure effusions (154 +/- 22 sq micron) (P less than .0001). There was also a smaller ratio of mesothelial cells in tuberculous effusions (0.5 +/- 0.9%) in comparison with effusions following radiotherapy (4 +/- 5%) or congestive heart failure (10 +/- 12%). In summary, this study demonstrated some cytomorphologic parameters that may be helpful in the differential diagnosis of tuberculous effusions.  相似文献
