. The effect of culture conditions on growth and immunogenicity of Clostridium chauvoei were examined. The pH control and partial feeding of the carbon source at high concentrations were beneficial for growth. The biomass yield was significatively improved, however the butanol concentration reached toxic levels hampering further growth. For each experimental condition the immunogenicity of cells was tested. No differences were found with cells obtained at different temperatures, but it decreased significatively with the partial supply of the carbon source and pH control.  相似文献

Summary Platelet interactions with cultured bovine endothelial cells were studied following freeze-thaw damage or detergent tratment. Platelets from whole blood, platelet-rich plasma, or gel-filtered plasma did not interact directly with freeze-thaw-damaged endothelial cells. Freezing and thawing did result in the exposure of an extracellular matrix located beneath the cells, which proved very thrombogenic. Platelets from all sources attached to both microfilament and amorphous components of the extracellular matrix, although only platelets from whole blood demonstrated aggregation and extensive pseudopodia formation. Treatment of cells with Triton-X detergent resulted in exposure of an intracellular cytoskeleton. Most platelets attached to the cytoskeleton were located near the cell border and had one or more pseudopodia either in contact with extracellular or intracellular material. Adhesion of platelets to the extra-cellular matrix may represent platelet-collagen or plateletfibronectin interactions since both are produced by an incorporated into the extracellular matrix. Platelet interaction with endothelial cytoskeletons may represent contact of pseudopodia with the now exposed matrix located beneath the cells. The possibility that platelets also adhered to intra-cellular components could not be eliminated. These findings are in agreement with data from a freeze-thaw injury model of perfused aorta. In addition, they tend to indicate that physical insult is not sufficient to induce platelet interaction with the endothelial surface, but that chemical modification enhances platelet deposition. This work proposes a new online monitoring method for an assistance during laser osteotomy. The method allows differentiating the type of ablated tissue and the applied dose of laser energy. The setup analyzes the laser-induced acoustic emission, detected by an airborne microphone sensor. The analysis of the acoustic signals is carried out using a machine learning algorithm that is pre-trained in a supervised manner. The efficiency of the method is experimentally evaluated with several types of tissues, which are: skin, fat, muscle, and bone. Several cutting-edge machine learning frameworks are tested for the comparison with the resulting classification accuracy in the range of 84–99%. It is shown that the datasets for the training of the machine learning algorithms are easy to collect in real-life conditions. In the future, this method could assist the doctors during laser osteotomy, minimizing the damage of the nearby healthy tissues and provide cleaner pathologic tissue removal.  相似文献

: The properties of the uptake of nucleosides and nucleotides by brain cells were examined in slices of mouse brain. Of the compounds tested, adenine and adenosine had the most rapid uptake and reached the highest levels. Uptake was mediated, as shown by saturability and strong inhibition, by low temperature, or by cyanide, and was only partially sodium- or calcium-dependent. The inhibition pattern by analogues indicated the presence of several uptake systems (possibly four), as shown by differences between adenine and guanine uptake, between adenine and adenosine uptake, and between adenosine and cytidine uptake. The properties of uptake systems for nucleotides and nucleosides were somewhat different from those for amino acids.  相似文献

Lactose present in whey UF-permeates was hydrolysed by an immobilised enzyme reactor and the formation of monosaccharides (glucose + galactose) and oligosaccharides was monitored. The enzyme used was β-galactosidase from A. oryzaeimmobilised in a porous film. The reactor, run in the flow-through mode, allowed large conversions at short residence times (60% conversion in 1 min). The conversion to oligosaccharides as a function of the reaction time (or degree of conversion) reaches a maximum and then declined as oligosaccharides were converted back to mono- and disaccharides. The higher the initial lactose concentration the higher the conversion to oligosaccharides and these maxima appear at higher degrees of conversion. Some trials were carried out on the concentration of the oligosaccharides present in the hydrolysates by means of membrane filtration (nanofiltration) .  相似文献

The haemocyanin of the left-handed whelk Busycon contrarium (Conrad) exists largely as six or more multi-decameric aggregates characterized by sedimentation coefficients of approximately 105S, 132S, 155S, 170S, 185S and about 200-220S. 2. These aggregates represent di- to hepta- or octa-decameric assemblies of the basic haemocyanin decamer having a mol. wt of 4.3 x 10(6)-4.5 x 10(6). 3. The fully dissociated subunits in 8.0 M urea (pH 8.5) and at pH 11.1, 0.01 M EDTA have mol. wts of 4.78 x 10(5) and 4.62 x 10(5), close to one-tenth of the mol. wt of the basic decameric unit of most gastropod haemocyanins. 4. The pH dependence of the mol. wts (Mw), studied by light-scattering at the constant protein concentration of 0.010%, exhibit bell-shaped pH transition profiles with mol. wt values of about 16 x 10(6) in the presence of 0.01 M Mg2+, in the pH region from about pH 4.5-8.0; in the absence of stabilizing divalent ions the observed mol. wt is about 10 x 10(6) at pH 4.5-7.0. Below pH 4.5 and above 7.0-8.0 there is a sharp drop in mol. wt to about 4 x 10(5)-4.5 x 10(5). 5. The transition profiles observed with both the urea and salt series of probes investigated at concentration = 0.010% are found to produce aggregation at low reagent concentrations with mol. wt changes from about 9 x 10(6)-12 x 10(6)-14 x 10(6), followed by a decrease in mol. wt below 4.3 x 10(6)-4.5 x 10(6) of the haemocyanin decamers.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
