Anode Break Excitation in Space-Clamped Squid Axons   总被引:1,自引:0,他引:1  
Strength-duration curves for space-clamped squid axons, using square wave anode breaks as stimuli, established the existence of four distinct regions. For the average experimental axon the intersection of the first two regions, τ1, occurs at about 7 msec. This agrees with computations based on the Hodgkin-Huxley (HH) equations and corresponds to the accommodation time constant found previously for a linearly rising ramp, as given by the HH equations and as found experimentally. The second break in the curve, τ2, at about 200 msec, and the third break, τ3, at 1 sec, are far beyond the range of the HH equations and may be the counterpart in the excitability of the long time constants, which have been apparent from a number of other types of experiments. The regions of the curve before 1 msec and beyond 2 or 3 sec are quite variable and may represent breakdown. Rheobase increases in both experimental and computed axons when temperature is raised. In both experimental and computed axons τ1 descreases slightly when the temperature is raised from 10 to 15°C. At 20 and 25°C, τ1 of the experimental axon increases markedly.  相似文献

The lowering of external sodium raised both the constant quantity threshold, Qo, and the rheobase, Io, in both real space-clamped squid axons and the theoretical axon as computed on the basis of the standard Hodgkin-Huxley equations. In both real and theoretical axons the minimum intensity for excitability for short pulses, which occurs at about 15°C, was still present when low sodium replaced seawater. Low sodium did not affect the temperature dependence of the strength-duration relationship in the range, 5° to 25°C. The excitability of tetrodotoxin-treated real axons was found to be more temperature-dependent than that of normal real axons. Also the data on dosage-response to TTX of real axons fit the dose-response relationship of a hypothetical system in which one TTX ion binds reversibly to its receptor to produce a fraction of the inhibitory effect, the curve being identical to a simple adsorption isotherm. The Hodgkin-Huxley equations describe the broad outline of events occurring during excitation quite well.  相似文献

Electrical rectification was demonstrated in whole sartorius muscle and sciatic nerve of Rana pipiens and also in the single giant nerve fiber of the northern squid, Ommastrephes illecibrossus. It is probably a property of the plasma membrane. Rectification decreases reversibly under the influence of increased concentrations of the potassium ion and with chloroform, veratrine sulfate and isoamyl carbamate. No effect was found with lack of calcium, excess calcium, or barium chloride. Decrease in rectification is invariably accompanied by simultaneous decrease in resting potential. A proposed explanation of the mechanism of rectification is discussed. Rectification in a living membrane, viz. a change in resistance with change in direction of current flow, may possibly be explained in terms of a change in the concentration of potassium ions in the membrane.  相似文献

Rosen (Bull. Math Biophysics. 1959) has argued that a self-reproducing automaton of the type originally described by von Neumann is impossible because of a logical paradox inherent in its definition. The paradox is resolved by explicitly allowing errors (mutations) in the system and thus introducing evolution. There is no paradox in an automaton, originating from a slightly different ancestor through mutation. The effect of trapidil derivative AR12456 on intracellular cholesterol metabolism was investigated in human hepatoma cell line HepG2. AR12456 enhanced the uptake and degradation of125I-LDL in a dose-dependent manner. The drug inhibited cholesterol synthesis and esterification without affecting cellular cholesterol content and bile acid synthesis; cholesterol efflux was slightly increased. These results show that the inhibition of cholesterol synthesis together with the enhanced expression of LDL receptors may partially explain the hypocholesterolemic activity of compound AR12456.  相似文献

Attaching and effacing (A/E) pathogens such as enterohemorrhagic Escherichia coli (EHEC) and enteropathogenic E. coli (EPEC) cause serious global health problems. These bacteria colonize the gastrointestinal system, attach to intestinal epithelial cells, efface (collapse) infected cell microvilli and cause overt diarrhoea that may ultimately result in death of the host. Although pathogenically induced diarrhoea is a significant global health issue, the molecular mechanisms that underlie this disease remain largely unknown. A natural murine infection model, employing the A/E pathogen Citrobacter rodentium, has been helpful in studying the diseases in vivo. C. rodentium colonize the colon at high levels, attach to colonocytes, efface microvilli and cause hyperplasia and inflammation in infected mice. As the disease progresses, the mice develop a diarrhoea-like phenotype. Aquaporin (AQP) water channels have been proposed to play a role in the normal dehydration of faecal contents. Here we examine whether C. rodentium infection may alter AQP localization in colonocytes. We demonstrate that during infection, AQP2 and AQP3 are mislocalized from their normal location along cell membranes to the cell cytoplasm. The change in localization of these proteins correlates with the diarrhoea-like phenotype present in infected mice. Mice that recover from the infection at 28-35 days post inoculum regain their normal membrane AQP localization. The altered localization of AQPs is partially dependent on the bacterial type III effector proteins EspF and EspG. We conclude that altered AQP localization may be a contributing factor to diarrhoea during bacterial infection.  相似文献

Quantitative trait loci (QTL) affecting carcass and meat quality located on SSC2 were identified using variance component methods. A large number of traits involved in meat and carcass quality was detected in a commercial crossbred population: 1855 pigs sired by 17 boars from a synthetic line, which where homozygous (A/A) for IGF2. Using combined linkage and linkage disequilibrium mapping (LDLA), several QTL significantly affecting loin muscle mass, ham weight and ham muscles (outer ham and knuckle ham) and meat quality traits, such as Minolta-L* and -b*, ultimate pH and Japanese colour score were detected. These results agreed well with previous QTL-studies involving SSC2. Since our study is carried out on crossbreds, different QTL may be segregating in the parental lines. To address this question, we compared models with a single QTL-variance component with models allowing for separate sire and dam QTL-variance components. The same QTL were identified using a single QTL variance component model compared to a model allowing for separate variances with minor differences with respect to QTL location. However, the variance component method made it possible to detect QTL segregating in the paternal line (e.g. HAMB), the maternal lines (e.g. Ham) or in both (e.g. pHu). Combining association and linkage information among haplotypes improved slightly the significance of the QTL compared to an analysis using linkage information only.  相似文献
