We have investigated the DNA binding properties of the anthramycin analogues 4, 5, and 6 using fluorescence spectroscopy. A considerable fluorescence enhancement occurs when pyrrolo [1,4] benzodiazepines (P[1,4]Bs) are covalently attached to duplex DNA, which was used to show that neither the presence of RNA, single-stranded DNA, or protein had any effect on the degree of fluorescence enhancement resulting from the incubation of 5 and 6 with DNA. The enhancement was found to be dependent on the presence of the imine functionality in each of the compounds. A wavelength of 320 nm was used to excite the chromophore and its emission wavelength maximum was 420 nm. Additionally, we have discovered that the P[1,4]B ring system exhibits exceptionally favorable fluorescence polarization anisotropy (FPA) decay characteristics. For these more detailed fluorescence measurements, we used the structurally simpler analogue 4,. The time resolved maximum FPA for 4 in glycerol at 25 degrees C is 0.28. This result indicates that the P[1,4]B family of antibiotics could serve as sensitive probes of DNA dynamics in the 0.1 to 35 ns time scale.  相似文献

It has been proposed that in addition to production of a penicillin-binding protein with low affinity for beta-lactam antibiotics, control of autolysin activity is involved in the mechanism of staphylococcal methicillin resistance. A homogeneous methicillin-resistant Staphylococcus aureus strain (DU4916) had lower rates of unstimulated, NaCl- and Triton X-100-stimulated autolysis, and daptomycin (LY146032)-induced lysis than a heterogeneous methicillin-resistant strain (DU4916-K7) and a methicillin-susceptible strain (DU4916S) derived from DU4916.  相似文献

The population dynamics of interior ice microalgae were investigated at a snow-free site on annual land-fast sea ice in McMurdo Sound, Antarctica, during the austral spring and summer of 1995 – 96. A dynamic successional sequence was observed with life history transformations playing an important role. During late November and early December (austral spring), cryo- and halotolerant dinoflagellates and chrysophytes bloomed in brine channels within the upper ice. At this time, competition and grazing pressure are low because of the inability of most marine species to grow under the extreme environmental conditions found in the upper ice during the austral spring. In November and December, dinoflagellates, chrysophytes, and prasinophytes contributed an average of 66%, 44%, and < 1% of the phytoflagellate biomass, respectively. Both the dinoflagellates and the chrysophytes encysted in December, with cyst formation most intense just prior to surface melt and flushing of the ice. The cysts appear to be an adaptation for survival and dispersal in the plankton during ice decay and/or overwintering in the sea ice. In January (austral summer), when ice temperatures were similar to those in the water column, pennate diatoms replaced flagellates as the photosynthetic dominants in the upper sea ice. The upper land-fast sea ice undergoes dramatic seasonal changes in light availability, temperature, brine salinity, and inorganic nutrient availability. Ephemeral blooms of cyst-forming phytoflagellates exploit this habitat in the austral spring, when both inorganic nutrients and light are available but temperatures <− 2° C and brine salinities elevated.  相似文献

To follow the biodistribution of exogenous hyaluronan in tumor-bearing animals, a total of seventeen inbred rats with hepatic metastases from a colonic adenocarcinoma received 125I-labelled hyaluronan by intravenous injections. Group I received only labeled hyaluronan (25 g), whereas group II received 2.5 mg chondroitin sulphate prior to labeled hyaluronan, to block receptor uptake in normal liver endothelial cells. Animals in group III received intravenous, as well as intraperitoneal chondroitin sulphate (2.5 mg), to see if a better and prolonged blocking could be achieved. Radioactivity was visualized by whole body autoradiography, using phosphorimaging and the average radioactivity determined as phosphoimaging density units of the total area of hepatic metastases, normal liver, and skeletal muscle by computer-based image analysis. At 5 h, tumors in groups II and III showed higher uptake (4.8 ±1.8, P = .01 and 3.6 ±1.1, P = .01, respectively), in comparison to group I (1.8 ±0.6), and the mean normal liver/tumor concentration ratio was reduced from 21.4 ±10.1 in group I to 5.7 ±2.7 in group II and 3.5 ±1.1 in group III (P = .008 and P = .01, respectively).Our study shows that hyaluronan targets liver metastases of a colon adenocarcinoma. Furthermore, chondroitin sulphate pretreatment increases tumor uptake, while uptake at normal receptor sites is significantly reduced. The results also suggest that after blocking of normal hyaluronan/chondroitin sulphate receptors in healthy tissue, hyaluronan may be used to deliver drugs to specific hyaluronan receptor-positive sites of pathology.  相似文献

Tea tree oil (TTO) stimulates autolysis in exponential and stationary phase cells of Escherichia coli . Electron micrographs of cells grown in the presence of TTO showed the loss of electron dense material, coagulation of cell cytoplasm and formation of extracellular blebs. Stationary phase cells demonstrated less TTO-stimulated autolysis and also had greater tolerance to TTO-induced cell death, compared to exponentially grown cells. It was also revealed that a subpopulation of stationary phase cells demonstrated increased tolerance to TTO-bactericidal effects.  相似文献

This study was designed to use biotin labelling in situ hybridization to physically map the 5S rDNA genes to a chromosome arm location in rice. Chromosome preparations were made using an improved protoplast technique, which resulted in more mitotic cells with less overlying cytoplasmic and cellular debris. Cells in which both chromatids were labelled were observed. The hybridization detection level for the 5S rDNA gene complex was 17.22%. The results established that the 5S rDNA gene complex of rice is located at the end of the short arm of chromosome 9 in rice cultivar IR36. The similarities and differences of the 5S rDNA gene complex location between rice and other cereals and advantages of in situ hybridization for physical mapping are discussed.  相似文献

A F2 mapping population was created from a cross between 'UC-90' and E-line ryes (Secale cereale L.), two lines that showed polymorphism for eight C-band loci. Clones from rye, as well as other grasses, were used as probes. RFLP maps of rye chromosomes 6R and 7R were generated that include the 6RS and 6RL terminal C-bands and the 7RS terminal C-band. The 6R map spans 230 cM and includes 9 loci. The 7R map covers 225 cM and includes 21 loci. Segregation distortion was detected for several chromosomal regions. Heterochromatic C-bands did not appear to be responsible for the distortion.  相似文献
