1. Variations in pH value between 4 and 8 produce practically no effect on the normal rate of respiration (the rate at neutrality is called normal). 2. Increasing the pH value to 8.80 causes respiration to fall to 60 per cent of the normal, after which it remains stationary for the duration of the experiment. 3. Decreasing the pH value to 2.65 causes a gradual rise and a gradual return to normal; at pH 1.10 to 1.95 the preliminary rise amounts to 20 per cent and is followed by a fall to below the normal. 4. The decrease in respiration brought about by solutions of a pH value of 1.95 or less are irreversible, while a similar decrease which occurs at pH 8.80 is reversible, the rate coming back to practically normal after the material is replaced in a neutral solution. 5. Determinations by means of Winkler''s method showed an increase in the consumption of oxygen in acid solutions and a decrease in alkaline solutions.  相似文献

The effects of a potent HMG CoA reductase inhibitor, lovastatin, was studied in male Syrian Golden hamsters. Lovastatin (0.1% in food for 6 days) increased hamster serum triglycerides by 12-fold with 2.4-fold increase in serum cholesterol. On continuous treatment serum triglyceride and cholesterol levels gradually decreased to below control values by 12-18 days. When hamsters were fed a mixture of lovastatin and Na-mevalonate no increase in serum triglyceride was observed. [14C]Cholesterol synthesis was increased by 266-fold in livers of hamsters fed lovastatin for 6 days. The increased synthesis of endogenous mevalonate metabolites may be a reason for the decrease in triglyceride levels after 6 days in our studies. The present study suggests that a mevalonate metabolite(s) is necessary for normal triglyceride metabolism in hamsters.  相似文献

Previous research established a relationship between circulating sulfoconjugated norepinephrine (NE-SO4) and oxygen consumption at various exercise intensities. In this study, the stability of the NE-SO4 response was examined during sustained exercise at a constant relative intensity. Seven trained men bicycled at 78 +/- 3% of their maximal O2 consumption for 28 min and then rested on the ergometer for a comparable duration. After a 30-min rest, plasma samples were collected through an indwelling catheter at 7-min intervals during the exercise and recovery periods. Free NE and epinephrine increased sixfold during exercise. These changes were accompanied by increases in sulfoconjugated catecholamines, but only NE-SO4 achieved statistical significance (rest, 712 +/- 602; exercise, 1,329 +/- 1,163 pg/ml). This occurred at three collection periods (14, 21, and 28 min). Approximately 35, 52, and 95% of NE, epinephrine, and dopamine, respectively, existed as sulfoconjugated during exercise. Subject variation was present in the sulfoconjugated catecholamine response that could not be attributed to corresponding differences in circulating free catecholamine release. These findings implicate blood flow as a factor in the sulfoconjugation of NE, but not epinephrine or dopamine.  相似文献

A microassay is demonstrated for functional characterization of the Ca2+-release channel (CRC) of sarcoplasmic reticulum (SR) of skeletal muscle using swine with susceptibility to malignant hyperthermia (MH). Diluted muscle homogenates, indo-1 and ratiometric dual-emission spectrofluorometry are used to monitor Ca2+-lowering activity in real-time in the presence and absence of ryanodine at exposures that open and close the CRC. Reactions are initiated with 50 µM CaCl2 to raise ionized Ca2+ concentration near 1 µM and MgATP to activate the Ca2+-ATPase pump. Oxalate is included to precipitate Ca2+ within the SR. The assay requires less than 30 mg muscle, which may be cryopreserved, and is completed within 20 min of thawing the tissue. Maximum SR Ca2+-ATPase pumping and CRC activities, degree of CRC activation, and Ca2+-buffering capacity can be determined. Using this assay we studied muscle from MH-susceptible swine and demonstrated that whereas maximal Ca2+-ATPase pumping and CRC activities are normal, the CRC activity after addition of a bolus of Ca2+ is 50% greater in heterozygotes and 100% greater in homozygotes for the MH mutation. Hypersensitivity to CRC agonists, such as caffeine, and an associated hyposensitivity to CRC antagonists such as Mg2+ is also demonstrated. Genotypes for the MH mutation site can be discriminated from each other by determining Ca2+-lowering activities and the effect of ryanodine on them. (Mol Cell Biochem 167: 61-72, 1997)  相似文献

 A polymerase chain reaction (PCR) application, involving the directed amplification of minisatellite-region DNA (DAMD) with several minisatellite core sequences as primers, was used to detect genetic variation in 17 species of the genus Oryza and several rice cultivars (O. sativa L.). The electrophoretic analysis of DAMD-PCR products showed high levels of variation between different species and little variation between different cultivars of O. sativa. Polymorphisms were also found between accessions within a species, and between individual plants within an accession of several wild species. The DAMD-PCR yielded genome-specific banding patterns for the species studied. Several DAMD-PCR-generated DNA fragments were cloned and characterized. One clone was capable of detecting multiple fragments and revealed individual-specific hybridization banding patterns using genomic DNA from wild species as well as rice cultivars. A second clone detected only a single polymorphic locus, while a third clone expressed a strong genome specificity by Southern analysis. The results demonstrated that DAMD-PCR is potentially useful for species and genome identification in Oryza. The DAMD-PCR technique also allows for the isolation of informative molecular probes to be utilized in DNA fingerprinting and genome identification in rice. Aluminum (Al) stress was examined in three lines of wheat ( Triticum aestivum L.) by measuring root lengths, protein synthesis and protein accumulation in seedling root tips grown in a hydroponic system. An Al-sensitive, recurrent wheat parent (cv. Katepwa) showed very little root growth in low Al concentrations. In contrast, an Al-tolerant near isogenic line (Alikat) and Al-tolerant donor (cv. Maringa) had much greater root growth. Segregation data from an F2 population (Katepwa × Alikat) showed that one major gene controlled Al tolerance based on root growth ( X 2= 0.651). All three lines showed an approximately 2-fold increase in [35S]-Met incorporation in root tips after 3 days in Al and a comparable increase in root-tip dry weight. Maringa and Alikat root tips showed an increased total protein content while Katepwa root tips showed no increase in total protein content during the Al stress. Based on higher specific activities, insoluble proteins were preferentially translated in all three lines during Al stress. Proteinase activity in Katepwa root tips was 1.7-fold higher during Al stress, with Maringa and Alikat showing no change in proteinase activity. The Al-induced, increased proteinase activity in Katepwa appeared to inhibit soluble protein accumulation.  相似文献

An examination of variables affecting the yield of DNA from blood was undertaken in order to improve sample processing and to evaluate alternative methods of mailing blood samples for DNA analysis. A rapid, high-yield method was developed for the isolation of high-molecular-weight DNA from fresh and frozen blood. In addition, the following observations were made: (1) Of the anticoagulants examined, acid citrate dextrose (ACD) solution B was found to be superior to EDTA and heparin for preserving yields of DNA during incubation at room temperature. If DNA is isolated from frozen blood, high yields of undegraded DNA are achieved after incubation at 23 degrees C for 5 days with ACD solution B. (2) High yields of undegraded DNA are obtained from blood stored with ACD solution B for at least 1 day at 42 degrees C, 5 days at 0 degrees C, or 1 month at -20 degrees C. (3) Three cycles of freezing and thawing may have little if any affect on the yield of DNA. The results indicate that blood for DNA extraction may be mailed in an ambient temperature container and, in many cases, sent by first-class mail rather than by overnight delivery services.  相似文献
