The deconvolution method, i.e., reconstruction of noise-free discrete amplitude distributions of excitatory postsynaptic potentials (EPSPs), has been tested in computer experiments. A generalized quantal model without constraints on distribution of transmitter release probabilities (p) was used. Complex binomial amplitude distributions with a number of discrete components n between 2 and 10 and with different p were simulated. The distance between the discrete components (i.e., the quantum value, v) was determined from simulated distributions with the number of amplitudes N equal to 1000 or 100. For samples with N=100, mean v values were calculated from 10 distributions. The solutions obtained by deconvolution were close to (within ±20% of) the simulated v values when the standard deviation of the noise Sn0.5v. For Sn=0.5–2.0v, v grew in proportion to Sn and was overestimated. A similar correlation was found for v calculated from amplitudes of minimal EPSPs recorded from CA1 area neurons of guinea pig hippocampal slices. However, no significant correlation between v and Sn was found for v>135 µV, i.e., for estimates exceeding the total mean value. A substantial increase in the mean quantal content (m) with a relatively slight increase in v during longterm potentiation (LTP) was confirmed for those cases. Methods of elimination of erroneous v estimates were considered. Rejection of cases with v>2.5Sn (when N=1000) or v>3Sn (when N=100) eliminated erroneous estimates with a high probability but demanded very low noise levels. A new criterion for recognition of correct estimates is suggested. It is based on artificial contamination of the measured amplitudes by the Gaussian noise (noise addition) and on the double-step dependence of v on Sn described above. The establishment of the steady-state rate of photosynthetic O2 evolution by cells of Anabaena variabilis and other cyanobacteria was found to be preceded by a lag-phase the duration of which depended on the time of cell preincubation in the dark. Electron acceptors (benzoquinone, N,N,N,N-tetramethyl-p-phenylenediamine, 2,3,5,6-tetramethyl-p-phenylenediamine or 2,6-dichlorophenolindophenol) abolished the lag-phase as well as the inhibitory effect of cyanide on electron transfer. Mono-, di-and trivalent cations added to the cell suspension markedly reduced the lag-phase. As cation concentrations were increased, acceleration and subsequent deceleration of the O2 evolution rate were observed. The efficient concentrations of cations decreased as their valency increased. The lag-phase and the rate of photosynthetic O2 evolution by the blue-green algae are suggested to depend on the value of the membrane surface charge governing the electrostatic interaction between unidentified membrane-bound redox components. Bovine cardiac troponin C was cleaved at residues cysteine-35 and cysteine-84. Three peptides, N-terminal (residues 1-34), central (residues 35-83) and C-terminal (residues 84-161), of cardiac troponin C were obtained in a homogeneous state. Saturation of troponin C or its C-terminal peptide with Ca2+ or Mg2+ is accompanied by an increase in the ellipticity at 222 nm in the c.d. spectrum. The half-maximal changes in the ellipticity of troponin C were observed at 32 nM-Ca2+ or 56 microM-Mg2+. The corresponding values for the C-terminal peptide are 7.1 nM for Ca2+ and 4.5 microM for Mg2+. The ellipticity of the central peptide (residues 35-83) containing the second cation-binding site was decreased on saturation with Ca2+. The half-maximal changes in the ellipticity occur at 80 microM-Ca2+. Study of the c.d. spectra suggests that the alpha-helices flanking the second cation-binding site of cardiac troponin C exist independently of Ca2+. Saturation of the third and fourth sites with these cations is associated with a considerable increase in the alpha-helix content, probably due to the formation of an alpha-helix flanking the third site on the N-terminus.  相似文献

Phosphorylation of avian gizzard caldesmon by casein kinase II was investigated. The enzyme incorporates about 1 mol of phosphate per mol of caldesmon. All sites of phosphorylation are located in short chymotryptic peptides with Mr 25-27 kDa or in the short N-terminal peptide formed after cleavage of chicken gizzard caldesmon at Cys153. The primary structure of the tryptic peptide containing the main site of duck gizzard caldesmon phosphorylation is S-E-V-N-A-Q-N-X-V-A-E-D-E-T-K, where X is an unidentified residue, presumed to be phosphoserine. Thus, Ser73 is the main site phosphorylated by casein kinase II in avian gizzard caldesmon.  相似文献

Experimental endophthalmitis was treated with single intravitreous administrations of cefotaxime (claforan) and gentamicin. It was found that a single administration of cefotaxime to the vitreous body prevented development of endophthalmitis in rabbits previously infected with Staphylococcus aureus, E. coli and Pseudomonas aeruginosa. Vitrectomy in the treatment of endophthalmitis was shown to be promising and provide satisfactory anatomical and functional results.  相似文献

The suggested earlier complexity approach for detecting structural regularities in primary structures of nucleic acids is illustrated by using lambda phage as an example. Among the most interesting regularities detected in the lambda phage genome are the following: (a) the presence of "extended homology zones" i.e. fragments in which block transpositions of duplicative type predominate explicitly; (b) the abundance of palindrome-hairpin structures and duplications in the origins and termini of many genes; (c) the hierarchy in the repeats and inversions organization.  相似文献

Abstract. The effect and accumulation of cadmium (Cd) in lettuce grown by means of the hydroponic technique was investigated by multivariate analysis, and was found to be affected by the concentration of other trace elements. Particularly iron acted in a strongly antagonistic way against Cd. Consequently, no absolute toxicity limits for Cd can be drawn without considering other trace elements.  相似文献

Pyocyanin added to suspensions of Pseudomonas aeruginosa P changes the structural organization of the cells depending on their physiological characteristics, in particular, the capability to liberate pyocyanin into the cultural broth. Exogenous pyocyanin does not interact with the cells of the parent strain producing the pigment. However, the structure of the isolated cell walls deformed after the fractionation becomes similar, upon contact with pyocyanin, to the structure of the freshly isolated cell walls. The fraction of the cytoplasmic membranes of the parent strain also slightly changes its spatial organization after addition of pyocyanin. The cells of the mutant which do not produce pyocyanin display the ability for structural interaction with exogenous pyocyanin. In contrast, the fraction of their cell walls does not react to addition of the pigment. The cytoplasmic membranes of the mutant interact with pyocyanin in the same manner as the whole cells. These changes caused by pyocyanin always involve certain parts of the molecules. As a result, the following features are observed in their spectra: the absorption at 1475 cm-1 becomes more intensive; the "knee" at the band Amide II is more pronounced in the range of 1500 cm-1; the absorption at 1520 cm-1 decreases as well as the intensity of the band Amide II as a whole. The interaction of pyocyanin with biological objects stems, apparently, from the presence of certain proteins in them since peroxidase was found to respond to pyocyanin in a manner similar to that described for the cells and their fractions.  相似文献

Summary Unidirectional fluxes of 204Tl+ through the human red blood cell membrane were measured. The inward rate coefficient measured in a K+-free saline was 15.6±0.6 hr–1. The influx of Tl+ could be partially inhibited with 0.1mm ouabain (by 28%), 0.1mm DIDS (by 50%) or 1mm furosemide (by 51%). The inhibitory effects of ouabain and DIDS or furosemide were additive. Half-maximal responses were seen at 0.72 m and 0.22mm concentrations of DIDS and furosemide, respectively. A similar action of these blockers on Tl+ influx was observed in the erythrocytes incubated in MgCl2-sucrose media. The outward rate coefficient of 204Tl was also inhibited by DIDS and furosemide (by 65 and 52%, respectively). Rate coefficients of 204Tl influx and efflux decreased significantly in the red cells exposed to Cl-free media (NaNO3 or Mg(NO3)2-sucrose). Under these conditions addition of DIDS and furosemide led to only a small inhibition of Tl+ fluxes. There was a linear increase in Tl+ influx with rising of external Cl concentration within 80–155mm or HCO 3 concentration from 20 to 40mm when the sum of anions was kept constant (155mm) with NO 3 . The HCO 3 -stimulated Tl+ influx was completely blocked by 0.05mm DIDS but only 67% by 1mm furosemide. The present study provides direct evidence for the occurrence of Cl (HCO 3 )-dependent, DIDS-sensitive movement of Tl+ across the human erythrocyte membrane in both directions. Under physiological conditions, about half of net Tl+ fluxes occurs due to an anion exchange mechanism. Our data fail to detect a contribution of the Na-K-Cl cotransport system to Tl+ transport in human erythrocytes.  相似文献
