The review is dedicated to phosphorylation of αB-crystallin (HspB5), one of ubiquitously expressed small heat shock proteins. We describe the structure and properties of αB-crystallin and protein kinases involved in its phosphorylation in different cells and tissues, advantages and drawbacks of pseudophosphorylation mutants in elucidation of the mechanism of αB-crystallin functioning, effects of phosphorylation on the quaternary structure and intracellular location of αB-crystallin, interactions of αB-crystallin with different elements of the cytoskeleton, and effect of phosphorylation on the chap-erone-like activity of αB-crystallin. We also discuss the validity of experimental data obtained by overexpression of pseudophosphorylation mutants for understanding the effect of phosphorylation on physiologically important properties of αB-crystallin, as well as the question why multiple attempts to phosphorylate αB-crystallin in vitro have been unsuccessful so far. 相似文献

A complex of closely related Mallomonas taxa belonging to the section Papillosae, M. kalinae ?ezá?ová and M. rasilis Dürrschmidt, has been studied in detail by molecular and morphometric methods. Our investigations uncovered the existence of a new species found in water bodies in Vietnam, which we describe here as Mallomonas furtiva sp. nov. This taxon is morphologically very similar to M. kalinae , from which it differs by minute, but statistically significant morphological differences on the structure of silica scales. Indeed, the principal component analysis of morphological traits measured on silica scales significantly separates all three species in the complex. Mallomonas kalinae and M . furtiva differ by number of papillae on the shield and the dome, as well as by the scale sizes. Likewise, Mallomonas rasilis and M. furtiva are primarily differentiated by the absence of submarginal anterior ribs on silica scales of the former species. Phylogenetic analyses showed that Mallomonas furtiva is closely related to M . kalinae , with which it formed a highly supported lineage. Radiation losses from the plasma of the Globus-M tokamak are studied by means of SPD silicon photodiodes developed at the Ioffe Institute, Russian Academy of Sciences. The results from measurements of radiation losses in regimes with ohmic and neutral beam injection heating of plasmas with different isotope compositions are presented. The dependence of the radiation loss power on the plasma current and plasma–wall distance is investigated. The radiation power in different spectral ranges is analyzed by means of an SPD spectrometric module. Results of measurements of radiation losses before and after tokamak vessel boronization are presented. The time evolution of the sensitivity of the SPD photodiode during its two-year exploitation in Globus-M is analyzed. 相似文献

The literary and experimental data on the structure and properties of cardiac and skeletal muscle troponin are reviewed. The cation--binding sites of cardiac and skeletal muscle troponin C are distinguished by specificity; the sites localized in the C-terminal part of the protein molecule can bind both Ca2+ and Mg2+, whereas the sites localized at the N-end specifically bind Ca2+. The use of bifunctional reagents revealed a number of helical sites within the structure of cardiac troponin C (residues 84-92 and 150-158) and of skeletal muscle troponin C (residues 90-98 and 125-136). A comparison of experimental data with the results of an X-ray analysis testifies to the presence in the central part of the troponin C molecule of a long alpha-helical sequence responsible for troponin C interaction with the inhibiting peptide of troponin I. The efficiency of interaction of troponin components depends on Ca2+ concentration; the integrity of the overall troponin complex is mainly provided for by troponin C interaction with troponin I and by troponin I interaction with troponin T. The interaction between troponins T and C is relatively weak, especially in the case of cardiac troponin components. Both skeletal and cardiac muscles synthesize several troponin T isoforms differing in length and amino acid composition of N-terminal 40-60 member peptides. Troponin T isoforms can undergo phosphorylation by several protein kinases. The single site of troponin T which exists in a phosphorylated state in vivo (residue Ser-1) undergoes phosphorylation by specific protein kinase (troponin T kinase) related to casein kinases II. It was assumed that the phosphorylation of Ser-1 residue of troponin T as well as the synthesis of troponin T isoforms differing in the structure of the N-terminal peptide, provides for the regulation of interaction between two neighbouring tropomyosin molecules. 相似文献

Potassium (rubidium) influx, sodium and potassium contents, as well as size distribution, DNA and protein contents and synthesis have been examined in PHA-activated human lymphocytes within 0.5-72 h. A complex set of ionic events was found to include at least two stages of the increase in potassium and sodium contents per g cell protein and in ouabain-sensitive potassium influx which are preceded by a decrease in potassium content by almost 17% within the first 2-5 h. The kinetics of potassium and sodium changes has own pattern for each of cations, thus indicating definite changes in the ouabain-resistant transport of potassium and sodium during the G0----G1----S progression. The late increase in potassium content per g cell protein was found to correlate with the growth in cell size. This finding confirms the rule which was stated earlier for other animal cells, i. e. cells that prepare to proliferate are to raise their potassium per g cell protein up to the level of 0.8-1.0 mmole (Vereninov. Within the scope of the concepts on the look control mechanism, i. e. the system of interrelated control of head and eye movements a mathematical model is considered which described the control system over eye involuntary movements with quadratic criterion of its work quality.
