A series of novel substituted 1-benzhydryl-piperazine sulfonamide 8(a-f) and benzamides 9(a-h) were synthesized and their antimicrobial activities evaluated in vitro by paper disc diffusion and micro dilution method against standard strains of Gram-positive (Staphylococcus aureus ATCC 25953, Staphylococcus epidermis 25212, Bacillus cereus 11778, Bacillus substilis 6051) and Gram-negative (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 2853, Proteus vulgaris ATCC 2853 and Salmonella typhi ATCC 9484) bacteria. Among the synthesized new compounds 8d, 8e, 9c, 9e, 9f and 9 h showed potent antimicrobial activities compared to the standard drug streptomycin.  相似文献

In order to explore the anticancer effect associated with the thiazolidinone framework, several 2-(5-((5-(4-chlorophenyl)furan-2-yl)methylene)-4-oxo-2-thioxothiazolidin-3-yl)acetic acid derivatives 5(al) were synthesized. Variation in the functional group at C-terminal of the thiazolidinone led to set of compounds bearing amide moiety. Their chemical structures were confirmed by 1H NMR, IR and Mass Spectra analysis. These thiazolidinone compounds containing furan moiety exhibits moderate to strong antiproliferative activity in a cell cycle stage-dependent and dose dependent manner in two different human leukemia cell lines. The importance of the electron donating groups on thiazolidinone moiety was confirmed by MTT and Trypan blue assays and it was concluded that the 4th position of the substituted aryl ring plays a dominant role for its anticancer property. Among the synthesized compounds, 5e and 5f have shown potent anticancer activity on both the cell lines tested. To rationalize the role of electron donating group in the induction of cytotoxicity we have chosen two molecules (5e and 5k) having different electron donating group at different positions. LDH assay, Flow cytometric analysis and DNA fragmentation suggest that 5e is more cytotoxic and able to induce the apoptosis.  相似文献

The regulation of endogenous levels of ascorbic acid in soybean by far-red absorbing form of phytochrome (Pfr) and by cryptic red light signal (CRS) was studied. Cryptic red light signal is produced by red light pre-irradiation of a photoreceptor other than far-red absorbing form of phytochrome (Pfr) and CRS amplifies the action of phytochrome. The endogenous level of ascorbic acid levels enhanced by phytochrome was amplified by CRS. The lifetime of CRS was from 0 to 2 h and the peak of enhancement of ascorbic acid due to CRS was between 16 to 24 h of dark incubation after the end of the treatment. CRS was found to be ineffective on UV-B enhanced endogenous levels of ascorbic acid.Key words: ascorbic acid, cryptic red light signal, glycine max, phytochrome, ultraviolet-BThe phytochrome mediated morphogenesis involves the conversion of Pr [red absorbing form] to Pfr [far-red absorbing form] and the magnitude of the response is dependent on Pfr/P tot ratio established at the end of the irradiation.1 In broom Sorghum anthocyanin synthesis induced by red light [R1] is reversible with far-red light. But a second red pulse [R2] given after the reversal resulted in increased anthocyanin production compared to the first pulse [R1]. When the red pulse was repeatedly given after every reversal with far-red, the anthocyanin production increased proportionately to the number of previously given pulses.2 Thus red pre-treatment induced a change in the cellular physiological state or change in content of a relevant substance[s] which is designated as Cryptic Red Light Signal [CRS] associated with red signal transduction.2 CRS was first characterized in detail in Broom Sorghum as Pfr amplifying signal produced by red pre-irradiation. CRS is inactive in the absence of Pfr but enhances the action of Pfr. CRS escapes reversal when the plants are exposed to far-red and is probably produced by a different species of phytochrome, distinct from the conventional reversible phytochrome.3We have investigated whether CRS influences other phytochrome regulated processes in plants in addition to anthocyanin synthesis. We chose another process, the synthesis of endogenous ascorbic acid, which is also regulated by conventional phytochrome.4 In soybean, the endogenous level of ascorbic acid is enhanced by conventional far-red reversible form of phytochrome. In addition, an independent UV-B photoreceptor [non reversible with far-red light] also enhances the endogenous synthesis of ascorbic acid in soybean. By using repeated pulses of red light, we have demonstrated that the Cryptic Red Signal is operative in soybean also and it amplifies the red light induced enhancement in the level of ascorbic acid. That CRS is active only in the presence of Pfr is demonstrated by the fact that pre-irradiation with red light is ineffective in amplifying UV-B induced enhancement of ascorbic acid levels. A similar observation on UV-B induced anthocyanin synthesis has been made in Broom Sorghum.2 A separate UV-B photoreceptor independent of phytochrome operates in the plants.5 Although CRS is presumably produced by pre-irradiation with red light, it does not enhance UV-B induced anthocyanin synthesis or ascorbic acid synthesis in the absence of formation of Pfr by the second red pulse.The life-time of CRS was determined as 6 h in 20°C and 3 h in 24°C grown seedlings of Broom Sorghum with reference to anthocyanin synthesis.2 The life-time of CRS determined in soybean seedlings grown at 25°C was upto 1 h.6 Since growing seedlings at a low temperature enhanced the effectiveness of CRS in Broom Sorghum, it was concluded that low temperature may either extend the lifetime of CRS or generate higher amount of CRS.2 Although the exact nature of CRS is yet to be analyzed, work in our laboratory has established the universal nature of this signal and evidences have been obtained for CRS effect in promoting red light induced hypocotyls inhibition in Cucumber seedlings and also red light induced synthesis of betacyanins in Amaranthus seedlings (submitted for publication).  相似文献

Background

An increase in the average age of dengue hemorrhagic fever (DHF) cases has been reported in Thailand. The cause of this increase is not known. Possible explanations include a reduction in transmission due to declining mosquito populations, declining contact between human and mosquito, and changes in reporting. We propose that a demographic shift toward lower birth and death rates has reduced dengue transmission and lengthened the interval between large epidemics.

Methods and Findings

Using data from each of the 72 provinces of Thailand, we looked for associations between force of infection (a measure of hazard, defined as the rate per capita at which susceptible individuals become infected) and demographic and climactic variables. We estimated the force of infection from the age distribution of cases from 1985 to 2005. We find that the force of infection has declined by 2% each year since a peak in the late 1970s and early 1980s. Contrary to recent findings suggesting that the incidence of DHF has increased in Thailand, we find a small but statistically significant decline in DHF incidence since 1985 in a majority of provinces. The strongest predictor of the change in force of infection and the mean force of infection is the median age of the population. Using mathematical simulations of dengue transmission we show that a reduced birth rate and a shift in the population''s age structure can explain the shift in the age distribution of cases, reduction of the force of infection, and increase in the periodicity of multiannual oscillations of DHF incidence in the absence of other changes.

Conclusions

Lower birth and death rates decrease the flow of susceptible individuals into the population and increase the longevity of immune individuals. The increase in the proportion of the population that is immune increases the likelihood that an infectious mosquito will feed on an immune individual, reducing the force of infection. Though the force of infection has decreased by half, we find that the critical vaccination fraction has not changed significantly, declining from an average of 85% to 80%. Clinical guidelines should consider the impact of continued increases in the age of dengue cases in Thailand. Countries in the region lagging behind Thailand in the demographic transition may experience the same increase as their population ages. The impact of demographic changes on the force of infection has been hypothesized for other diseases, but, to our knowledge, this is the first observation of this phenomenon. Crop genome sequencing projects generate massive amounts of genomic sequence information, and the utilization of this information in applied crop improvement programs has been augmented by the availability of sophisticated bioinformatics tools. Here, we present the possible direct utilization of sequence data from a sorghum genome sequencing project in applied crop breeding programs. Based on sequence homology, we aligned all publicly available simple sequence repeat markers on a sequence-based physical map for sorghum. Linking this physical map with already existing linkage map(s) provides better options for applied molecular breeding programs. When a new set of markers is made available, the new markers can be first aligned on a sequence-based physical map, and those located near the quantitative trait locus (QTL) can be identified from this map, thereby reducing the number of markers to be tested in order to identify polymorphic flanking markers for the QTL for any given donor × recurrent parent combination. Polymorphic markers that are expected (on the basis of their position on the sequence-based physical map) to be closely linked to the target can be used for foreground selection in marker-assisted breeding. This map facilitates the identification of a set of markers representing the entire genome, which would provide better resolution in diversity analyses and further linkage disequilibrium mapping. Filling the gaps in existing linkage maps and fine mapping can be achieved more efficiently by targeting the specific genomic regions of interest. It also opens up new exciting opportunities for comparative mapping and for the development of new genomic resources in related crops, both of which are lagging behind in the current genomic revolution. Zinc deficiency impairs cellular immunity. Up-regulation of mRNA levels of IFN-γ, IL-12Rβ2, and T-bet are essential for Th1 differentiation. We hypothesized that zinc increases Th1 differentiation via up-regulation of IFN-γ and T-bet expression. To test this hypothesis, we used zinc-deficient and zinc-sufficient HUT-78 cells (a Th0 cell line) under different condition of stimulation in this study. We also used TPEN, a zinc-specific chelator, to decrease the bioavailability of zinc in the cells. We measured intracellular free zinc, cytokines, and the mRNAs of T-bet, IFN-γ, and IL-12Rβ2. In this study, we show that in zinc-sufficient HUT-78 cells, mRNA levels of IFN-γ, IL-12Rβ2, and T-bet in PMA/PHA-stimulated cells were increased in comparison to zinc-deficient cells. Although intracellular free zinc was increased slightly in PMA/PHA-stimulated cells, Con-A-stimulated cells in 5 μM zinc medium showed a greater sustained increase in intracellular free zinc in comparison to cells incubated in 1 μM zinc. The cells pre-incubated with TPEN showed decreased mRNA levels of IFN-γ and T-bet mRNAs in comparison to cells without TPEN incubation. We conclude that stimulation of cells by Con-A via TCR, release intracellular free zinc which functions as a signal molecule for generation of IFN-γ and T-bet, and IL-12Rβ2 mRNAs required for Th1 cell differentiation. These results suggest that zinc increase Th1 cell differentiation by up-regulation of IFN-γ and T-bet, and IL-12Rbβ2 mRNAs.  相似文献

Wolbachia are cytoplasmically inherited endosymbionts known to cause several reproductive alterations in insects which allow their spread in host populations. In the Uzifly Exorista sorbillans, endoparasites of silkworms, the prevalence of Wolbachia is high in the field. In the present study, we investigated Wolbachia’s effects on the Uzifly fitness traits by measuring fecundity and hatching rate in crosses involving infected and cured individuals. We found evidence for positive fitness effects associated with Wolbachia infection in females which could help promote the spread of Wolbachia in E. sorbillans populations. We tested two types of treatments for removing Wolbachia, antibiotic therapy and high temperature treatment and found an influence on the reproduction: females treated by antibiotics have a lower fecundity than females cured by high temperature which could indicate a negative effect of the antibiotherapy on females’ fitness. Furthermore, the monitoring of the Uzifly populations during 2 years revealed seasonal variations of the offspring production which may be linked to temperature.  相似文献

An efficient stepwise synthesis of homo-oligomers and mixed oligomers of gabapentin and pregabalin on solid support using Fmoc-protected derivatives and HBTU/HOBt/DIEA as coupling agent is described. The synthesis was also carried out using solution phase methodology. The Gpn/Pgn homo oligomers and mixed oligomers forms C9 helix in solution as determined by NMR study. Chiral as well as achiral gamma amino acids were used for the synthesis of oligomers in order to investigate the secondary structural preferences.  相似文献
