Nanoviruses have multipartite, circular, single-stranded DNA genomes and cause huge production losses in legumes and other crops. No viral suppressor of RNA silencing (VSR) has yet been reported from a member of the genus Nanovirus. Here, we demonstrate that the nanovirus U2 protein is a VSR. The U2 protein of milk vetch dwarf virus (MDV) suppressed the silencing of the green fluorescent protein (GFP) gene induced by single-stranded and double-stranded RNA, and the systemic spread of the GFP silencing signal. An electrophoretic mobility shift assay showed that the U2 protein was able to bind double-stranded 21-nucleotide small interfering RNA (siRNA). The cysteine residues at positions 43, 79 and 82 in the MDV U2 protein are critical to its nuclear localization, self-interaction and siRNA-binding ability, and were essential for its VSR activity. In addition, expression of the U2 protein via a potato virus X vector induced more severe necrosis symptoms in Nicotiana benthamiana leaves. The U2 proteins of other nanoviruses also acted as VSRs, and the three conserved cysteine residues were indispensable for their VSR activity. 相似文献

Photo-induced halide segregation in wide-bandgap (WBG) perovskite leads to poor stability and limits its application in high-efficiency tandem solar cells. Here, a simple solution strategy to achieve photostable WBG perovskite solar cells (PSCs) with bandgap of ≈1.67 eV by ionic coupling potassium sorbate with defects at the buried perovskite interface is reported. Moreover, the ionic coupled potassium sorbate (ICPS) enables to control the formation of N-methyl formamidinium ions that can selectively passivate the perovskite defects at grain boundaries. As a result, the photo-induced halide segregation in the target perovskite films is frozen under intense light. The target single-junction WBG PSC achieves a record efficiency of 22.00% with an open-circuit voltage (VOC) of 1.272 V and photostability of less than 2% decay over 2000 h of operation. Perovskite/Silicon tandem solar cells are also fabricated that achieve an efficiency of 30.72% (certified 30.09% @1.087 cm2), which is the highest efficiency reported to date with a tunneling oxide passivating contact (TOPCon) c-Si substrate. G-protein-coupled receptor 41 (GPR41) and G-protein-coupled receptor 43 (GPR43) are important short-chain fatty acids (SCFAs) receptors. Previous studies indicated that GPR41 and GPR43 are involved in the secretion of gastrointestinal peptides, and glucose and lipid metabolism, and are closely related to obesity and type II diabetes, and other diseases. The purpose of the study was to explore the relationship of the GPR41 and GPR43 with seasonal breeding, and provide new prospects for further exploring the nutritional needs of breeding. We identified the localization and expression levels of GPR41 and GPR43 in the colon of the wild ground squirrels (Spermophilus dauricus) both in the breeding season and non-breeding season. The histological results revealed that the lumen diameter of the colon had obvious seasonal changes, and the diameter of the colonic lumen in the non-breeding season was larger than that in the breeding season. Immunohistochemical staining suggested that GPR41 and GPR43 are expressed in the simple layer columnar epithelium. In addition, compared with the breeding season, the mRNA and protein expression levels of GPR41 and GPR43 in the colon were higher during the non-breeding season.
