BackgroundThe second wave of avian influenza H7N9 virus outbreak in humans spread to the Guangdong province of China by August of 2013 and this virus is now endemic in poultry in this region.MethodsFive patients with H7N9 virus infection admitted to our hospital during August 2013 to February 2014 were intensively investigated. Viral load in the respiratory tract was determined by quantitative polymerase chain reaction (Q-PCR) and cytokine levels were measured by bead-based flow cytometery.ResultsFour patients survived and one died. Viral load in different clinical specimens was correlated with cytokine levels in plasma and broncho-alveolar fluid (BALF), therapeutic modalities used and clinical outcome. Intravenous zanamivir appeared to be better than peramivir as salvage therapy in patients who failed to respond to oseltamivir. Higher and more prolonged viral load was found in the sputum or endotracheal aspirates compared to throat swabs. Upregulation of proinflammatory cytokines IP-10, MCP-1, MIG, MIP-1α/β, IL-1β and IL-8 was found in the plasma and BALF samples. The levels of cytokines in the plasma and viral load were correlated with disease severity. Reactivation of herpes simplex virus type 1(HSV-1) was found in three out of five patients (60%).ConclusionExpectorated sputum or endotracheal aspirate specimens are preferable to throat swabs for detecting and monitoring H7N9 virus. Severity of the disease was correlated to the viral load in the respiratory tract as well as the extents of cytokinemia. Reactivation of HSV-1 may contribute to clinical outcome.  相似文献

[目的]研究重组骆驼蓬脂转移蛋白(recombinant Peganum harmala lipid transfer protein,r Ph LTP)与顺铂(DDP)协同对宫颈癌He La细胞的抑制作用及可能的作用机制。[方法]四甲基偶氮咗蓝(MTT)法检测He La细胞生长的抑制率;Annexin-V/PI双染法检测细胞凋亡率,流式细胞术检测细胞内活性氧(ROS)的变化情况。[结果]r Ph LTP与DDP联合应用后与相应的DDP组相比,对细胞生长的抑制率显著增强(P0.01),低浓度r Ph LTP联合2.5μg/m L顺铂对He La细胞的抑制率为(42.50±0.059)%,远高于高浓度顺铂(10μg/m L)对He La细胞的抑制率(36.88±0.134)%,细胞的凋亡率显著增加(P0.01),细胞内ROS极明显升高(P0.001)。[结论]r Ph LTP能够显著增强顺铂对He La细胞的化疗敏感性(P0.001),这种作用可能与r Ph LTP增强顺铂诱导细胞内活性氧的产生导致细胞凋亡有关。  相似文献

Increasing drought and extreme rainfall are major threats to maize production in the United States. However, compared to drought impact, the impact of excessive rainfall on crop yield remains unresolved. Here, we present observational evidence from crop yield and insurance data that excessive rainfall can reduce maize yield up to ?34% (?17 ± 3% on average) in the United States relative to the expected yield from the long‐term trend, comparable to the up to ?37% loss by extreme drought (?32 ± 2% on average) from 1981 to 2016. Drought consistently decreases maize yield due to water deficiency and concurrent heat, with greater yield loss for rainfed maize in wetter areas. Excessive rainfall can have either negative or positive impact on crop yield, and its sign varies regionally. Excessive rainfall decreases maize yield significantly in cooler areas in conjunction with poorly drained soils, and such yield loss gets exacerbated under the condition of high preseason soil water storage. Current process‐based crop models cannot capture the yield loss from excessive rainfall and overestimate yield under wet conditions. Our results highlight the need for improved understanding and modeling of the excessive rainfall impact on crop yield.  相似文献

Epigenetics pertains to heritable alterations in genomic structural modifications without altering genomic DNA sequence. The studies of epigenetic mechanisms include DNA methylation, histone modifications, and microRNAs. DNA methylation may contribute to silencing gene expression which is a major mechanism of epigenetic gene regulation. DNA methylation regulatory mechanisms in lens development and pathogenesis of cataract represent exciting areas of research that have opened new avenues for association with aging and environment. This review addresses our current understanding of the major mechanisms and function of DNA methylation in lens development, age-related cataract, secondary cataract, and complicated cataract. By understanding the role of DNA methylation in the lens disease and development, it is expected to open up a new therapeutic approach to clinical treatment of cataract.  相似文献

Autophagy is a membrane-mediated intracellular degradation system. The serine/threonine kinase Atg1 plays an essential role in autophagosome formation. However, the role of the mammalian Atg1 homologues UNC-51-like kinase (ULK) 1 and 2 are not yet well understood. We found that murine ULK1 and 2 localized to autophagic isolation membrane under starvation conditions. Kinase-dead alleles of ULK1 and 2 exerted a dominant-negative effect on autophagosome formation, suggesting that ULK kinase activity is important for autophagy. We next screened for ULK binding proteins and identified the focal adhesion kinase family interacting protein of 200 kD (FIP200), which regulates diverse cellular functions such as cell size, proliferation, and migration. We found that FIP200 was redistributed from the cytoplasm to the isolation membrane under starvation conditions. In FIP200-deficient cells, autophagy induction by various treatments was abolished, and both stability and phosphorylation of ULK1 were impaired. These results suggest that FIP200 is a novel mammalian autophagy factor that functions together with ULKs.  相似文献

2-Substituted 3-nitropropanoic acids were designed and synthesized as inhibitors against carboxypeptidase A (CPA). (R)-2-Benzyl- 3-nitropropanoic acid showed a potent inhibition against CPA (K(i)=0.15 microM). X-ray crystallography discloses that the nitro group well mimics the transition state occurred in the hydrolysis catalyzed by CPA, that is, an O,O'-bidentate coordination to the zinc ion and the two respective hydrogen bonds with Glu-270 and Arg-127. Because the nitro group is a planar species, we proposed (R)-2-benzyl-3-nitropropanoic acid as a pseudo-transition-state analog inhibitor against CPA.  相似文献

3种入侵和本地沉水植物形态和生理性状对螺类牧食的响应 沉水植物水盾草(Cabomba caroliniana)已成为中国太湖流域的优势入侵水生植物。与外来物种的原产地环境相比,引入地新环境中存在的专食性天敌较少。外来物种可能会逃避其原产地环境中的天敌牧食,又因为它们的适口性相对较差,从而导致在引入地外来物种通常比本地物种遭受的牧食者影响更低(天敌逃逸假说)。本研究的目的是比较水盾草与共生的本地沉水植物对本地牧食者的响应。我们进行了一个中宇宙尺度实验,研究了水盾草和两种共生的本地沉水植物黑藻(Hydrilla verticillata)和穗花狐尾藻(Myriophyllum spicatum)对两种本地广食性腹足纲螺类萝卜螺(Radix swinhoei)和环棱螺(Sinotaia quadrata)的牧食响应。记录了它们的形态性状指标(总生物量、冠根比和相对生长率)和生理性状指标(叶片总非结构性碳、木质素和纤维素)。研究结果表明,环棱螺对3种沉水植物性状指标的影响较少。随着本地广食性螺类萝卜螺数量的增加,黑藻和穗花狐尾藻大部分植物性状发生了改变,而水盾草的植物性状表现出相对稳定的趋势。水盾草对萝卜螺的牧食更具抵抗力,这与天敌逃逸假说的假设一致。这一发现说明牧食性螺类促进了水盾草的入侵,这可能会改变沉水植物群落中的物种组成。  相似文献

Animal influenza viruses pose a clear threat to public health. Transmissibility among humans is a prerequisite for a novel influenza virus to cause a human pandemic. A novel reassortant swine influenza virus acquired sustained human-to-human transmissibility and caused the 2009 influenza pandemic. However, the molecular aspects of influenza virus transmission remain poorly understood. Here, we show that an amino acid in hemagglutinin (HA) is important for the 2009 H1N1 influenza pandemic virus (2009/H1N1) to bind to human virus receptors and confer respiratory droplet transmissibility in mammals. We found that the change from glutamine (Q) to arginine (R) at position 226 of HA, which causes a switch in receptor-binding preference from human α-2,6 to avian α-2,3 sialic acid, resulted in a virus incapable of respiratory droplet transmission in guinea pigs and reduced the virus's ability to replicate in the lungs of ferrets. The change from alanine (A) to threonine (T) at position 271 of PB2 also abolished the virus's respiratory droplet transmission in guinea pigs, and this mutation, together with the HA Q226R mutation, abolished the virus's respiratory droplet transmission in ferrets. Furthermore, we found that amino acid 271A of PB2 plays a key role in virus acquisition of the mutation at position 226 of HA that confers human receptor recognition. Our results highlight the importance of both the PB2 and HA genes on the adaptation and transmission of influenza viruses in humans and provide important insights for monitoring and evaluating the pandemic potential of field influenza viruses.  相似文献
