Aerosolized or aspirated manufactured carbon nanotubes have been shown to be cytotoxic, cause pulmonary lesions, and demonstrate immunomodulatory properties. CD-1 mice were used to assess pulmonary toxicity of helical carbon nanotubes (HCNTs) and alterations of the immune response to subsequent infection by Pseudomonas aeruginosa in mice. HCNTs provoked a mild inflammatory response following either a single exposure or 2X/week for three weeks (multiple exposures) but were not significantly toxic. Administering HCNTs 2X/week for three weeks resulted in pulmonary lesions including granulomas and goblet cell hyperplasia. Mice exposed to HCNTs and subsequently infected by P. aeruginosa demonstrated an enhanced inflammatory response to P. aeruginosa and phagocytosis by alveolar macrophages was inhibited. However, clearance of P. aeruginosa was not affected. HCNT exposed mice depleted of neutrophils were more effective in clearing P. aeruginosa compared to neutrophil-depleted control mice, accompanied by an influx of macrophages. Depletion of systemic macrophages resulted in slightly inhibited bacterial clearance by HCNT treated mice. Our data indicate that pulmonary exposure to HCNTs results in lesions similar to those caused by other nanotubes and pre-exposure to HCNTs inhibit alveolar macrophage phagocytosis of P. aeruginosa. However, clearance was not affected as exposure to HCNTs primed the immune system for an enhanced inflammatory response to pulmonary infection consisting of an influx of neutrophils and macrophages.  相似文献

The most common response of a host to pathogens is arguably the asymptomatic response. However, the genetic and molecular mechanisms responsible for asymptomatic responses to pathogens are poorly understood. Here we report on the genetic cloning of two genes controlling the asymptomatic response to tobacco mosaic virus (TMV) in cultivated tobacco (Nicotiana tabacum). These two genes are homologous to tobamovirus multiplication 2A (TOM2A) from Arabidopsis, which was shown to be critical for the accumulation of TMV. Expression analysis indicates that the TOM2A genes might play fundamental roles in plant development or in responses to stresses. Consistent with this hypothesis, a null allele of the TOM2A ortholog in tomato (Solanum lycopersicum) led to the development of bent branches and a high tolerance to both TMV and tomato mosaic virus (ToMV). However, the TOM2A ortholog in Nicotiana glauca did not account for the asymptomatic response to TMV in N. glauca. We showed that TOM2A family is plant-specific and originated from Chlorophyte, and the biological functions of TOM2A orthologs to promote TMV accumulation are highly conserved in the plant kingdom—in both TMV host and nonhost species. In addition, we showed that the interaction between tobacco TOM1 and TOM2A orthologs in plant species is conserved, suggesting a conserved nature of TOM1–TOM2A module in promoting TMV multiplication in plants. The tradeoff between host development, the resistance of hosts to pathogens, and their influence on gene evolution are discussed. Our results shed light on mechanisms that contribute to asymptomatic responses to viruses in plants and provide approaches for developing TMV/ToMV-resistant crops.

The endosperm culture of wheat-rye hybrid was studied in order to explore a new pathway of chromosome engineering. The preliminary results were obtained to show that the endosperm callus formation could be induced from the young endosperm within 7–14 days after crossing on the medium supplemented with 2 ppm 2,4-D, 0.5 ppm kinetin and 3%–8% sucrose. The induction frequency of callus amounts to 35.3%. When the calli were transfered onto an auxin step-down medium containing 0.5 ppm IAA and 1 ppm kinetin, both shoots and roots were formed. 4 endosperm plantlets were obtained. The chromosome number in somatic cells of endosperm plantlets was very unstable. The numbers varied from 6—42, but there is no 49 to be found. The chromosome number with 1—4 times of 7 can be found in higher percentage.  相似文献

Based upon our previous work on the relationship between structure and function of chloroplast of wheat in connection with PSⅡ reaction, we studied the effects of MgCl2 and KC1 toward two kinds of thylakoid membranes. After exposing etiolated wheat seedlings to intermittent light (cycle of 2 min. light, 118 min. dark) for 24 hr, we obtain ed an incompletely developed chleroplast membrane. Completely developed chloroplast membrane was obtained from wheat seedlings grown under normal light-dark regime. Thylakoid membranes of plants grown under intermittent light failed to form grana stacks they remained as single lamellae in the suspension containing Mg++ or K+ of high concentration although simple stackings not more than two thylakoids c.ould be found. However, thylakoids grown under normal light-dark regime showed well developed grand stacks. Isolated chloroplast samples from two kinds of seedlings were suspended in 5 mM MgCl2 and 100 mM KC1 solutions for a definite time, portions of each samples were processed for electron microscopic observations and their photosynthetic activities were measured at the same time (It will be dealt with in another article). When these two kinds of isolated plastids were suspended either in MgCl2 (5 mM) or KC1 (100), the normally developed grana thylakoids stacked closely but the incompletely developed thylakoid- membranes did not stack. The incompletely developed chloroplast thylakoid membranes,, in either Mg++ or K+ ions could not induce stacking of the scattered thylakoid membranes to form grana. Therefore, we presume that light- harvesting chlorophyll a/b protein complex is on internal factor to induce thylakoid- membranes stacking and a definite concentration of caionions is an important factor in maintaining the stacking of thylakoid membranes. These results further prove the close association between structure and function in our previous studies on the mesophyll cell of the winter wheat.  相似文献

A hydroxyproline (HYP) resistant cell line of Chinese aneurolepidium (Aneurolepidium chinense (Trin.) Kitag. ) was isolated under selection pressure of 20 mmol/L HYP. Comparison of the free amino acid pool levels in the cell line with that of donor showed substantial accumulation of proline (6.6 × ). Enzyme examination indicated that γ-glutamyl kinase controlling proline biosynthesis in HR20-8 cell line had 2.5 times as much activity as that of the donor. Exogenous L-proline inhibited the enzyme activities both in the HR20-8 and the donor by the same rate of 30% at 100 mmol/L. The responses of HR20-8 cell line to NaC1, PEG and cold temperature (5 ℃) were also compared with those of donor and the former exhibited remarkably increased tolerance to the tested stress condition. The results showed that changes of γ-glutamyl kinase property exhibited the phenomenon of extra accumulation of proline which might favor the increased tolerance to NaC1, PEG and cold temperature in the resistant cell line.  相似文献

The nutritional quality of seed proteins from cereals, such as wheat and rice, is comparatively low due to its deficency in lysine and some essential amino acids. In this research extensive varieties of plant seed samples were collected and screened by analysis of amino acid composition. Three lysing-rich species which contain more than 6.7% of lysine in total seed proteins were found. 31 kinds of proteins were purified from a species which contains 7.9% of lysine using the modified methods of IEF and SDS electrophoresis. One protein with PI 6.1 and 18 kD was identified which contains 11.4% of lysine and was rich in threonine, valine and isoleucine. This is the first example of the protein which could complement several limiting amino acids of wheat or rice. Further research on the structural gene encoding this protein would have great potential value for improvement of protein quality of these cereals.  相似文献

By comparison of thylakoid membrane lipids and their fatty acid composition, the supermolecular structure of light harvesting chlorophyll a/b-protein complex of Photosystem Ⅱ (LHC Ⅱ ) and the spectroscopic characteristics of thylakoids in winter wheat (Yanda 1817) with those in spring wheat (8901) before and after cold-hardening, it was found that after cold-hardening: (1)The trans-3-hexadeeenoic acid content of phosphatidyl alycerol (PG) in both cultivars decreased significantly, the ratio of monogalactosyl diglyceride (MGDG)/digalactosyl diglyceride (DGDG) in the thylakoid of Yanda 1817 decreased, but had no distinct change in 8901. (2)The lipid/chlorophyll ratio in thylakoids of Yanda 1817 increased significantly, but had no distinct change in 8901. (3) The LHC Ⅱ oligomer content decreased in thylakoids of both cultivars. (4) The A683/A652 ratio of the 4th derivative absorption spectra increased in both cultivars. (5)The F685/F738 ratio of low temperature (77K) fluorescence spectra of thylakoids in 8901 increased but was not affected in Yanda 1817. It was concluded that one of the major strategies of wheat to adapt low temperature was the increase of thylakoid membrane fluidity, and that the decrease of MGDG content may play an important role in stabilizing the bilayer structure of the thylakoid membrane at low temperature.  相似文献
