Lipid pattern and Na+−K+-dependent adenosine triphosphatase activity in the salt gland of duck before and after adaptation to hypertonic saline

Summary Ducks (Anas platyrhynchos) were fed hypertonic saline for eight days, resulting in an activation and hypertrophy of the salt gland. The Na⁺–K⁺-dependent adenosine triphosphatase, an enzyme generally assumed to be part of the active Na transport system, increased its specific activity by about 200% during this activation. Sulfatides, the major glycolipids of the salt gland, increased their concentration to the same extent. Cholesterol, cerebrosides, and six phospholipid classes showed an increase of 20–80%.A preliminary report on this work was given at the Second International Meeting of the International Society for Neurochemistry, Milan, September 1–5, 1969, and at the XIIIth International Conference on the Biochemistry of Lipids, Athens, September 7–12, 1969.     

Microdensitometer system for microradiography

Summary A microdensitometer system for point measurements in areas down to a few square micrometers is described. The detector system is connected to an integrating digital voltmeter, constituting a very sensitive system. The digital output signals and a digital identification are registered on a recorder and on a tape punch, thus facilitating computer analysis.The precision of the equipment varied from 1.1 per cent, for low signal levels, to 0.3 percent for the highest signal levels. Stray light, the only important source of systematic errors, was found to influence the output signal level. Concerning determinations of the densities, however, a variation of the order of 1 per cent was only obtained for small areas with densities differing from that of the background.     

Changes in the anatomical structure of the shoot apex ofSenecio vulgaris L. during ontogenis in relation to the formation of leaves and inflorescence

An investigation was made of the anatomical structure of the shoot apex ofSenecio vulgaris L. a photoperiodically neutral plant, and compared with the formation of successive leaf primordia along the axis up to the initiation of the terminal inflorescence. In the shoot apex of a germinating plant a central zone can first be distinguished from the peripheral zone which is composed of small and intensely stained cells. Later, a rib meristem appears. At the time of the initiation of the middle (the largest) leaves, the shoot apex has a distinct small central zone and a well developed peripheral zone and rib meristem. Between these zones there is a group of cells dividing in all directions, the subcentral zone. At the time of initiation of the last leaves, the central zone extends to the flanks and gradually ceases to be distinguishable. At the same time, the subcentral zone increases in size. This is caused first by cell division and later, with the initiation of the last, most reduced leaves, by enlargement of the cells. Vacuolization in the inner part of the apex and the arrangement of the superficial cells in rows parallel to the surface of the apex, is a preparatory step to the initiation of the inflorescence.     

Purification of the microsomal Ca2+-ATPase from rat liver

Summary The Ca²⁺-ATPase from rat liver microsomes has been solubilized in Triton X-100 and purified to homogeneity by ficollsucrose treatment, column chromatography with agarose-hexane adenosine 5-triphosphate Type 2, and high pressure liquid chromatography (HPLC). The purified enzyme obtained by this sequential procedure exhibited a 183-fold increase in specific activity. After ficoll-sucrose treatment, the activity of the Ca²⁺-ATPase was stable for at least two weeks when stored at –70°C. In SDS-polyacrylamide gels, several fractions from HPLC chromatography showed a single band at a position corresponding to a molecular weight of about 107 kDa. This value is consistent with the molecular weight of the phosphoenzyme intermediate of endoplasmic reticulum (ER) Ca²⁺-ATPase. Further characterization of the ER Ca²⁺-ATPase was performed by western immunoblots. Antiserum raised against the 100-kDa sarcoplasmic reticulum (SR) Ca²⁺-ATPase cross-reacted with the purified Ca²⁺-ATPase from rat liver ER membranes.     

洪湖野菰及其化学成分分析

野菰(Zizania latifolia)是湖北省洪湖中优势水生维管束植物,其群落占全湖355平方公里面积的127平方公里。茎和叶的年生物量为4379克鲜重/平方米,全湖总年产量121700吨干重,目前未被利用。野菰各器官的蛋白质和氨基酸含量分别以百分干重表示:根,7.0和4.76;根状茎,11.3和8.85;茎,9.5和7.15;嫩茎梢,22.4和16.53;叶,16.8和14.61。500克干叶的必需氨基酸含量接近100克干重草鱼幼鱼背肌的必需氨基酸含量。脂肪:叶中3.4～4.2,茎中2.2;粗纤维:叶中26.8～28,茎中24.2;灰分:叶中10.0,茎中5.8。菰茎含可溶性糖类30％以上,其中葡萄糖,果糖、蔗糖和麦芽糖是主要成分。结果表明野菰是一种优质饲料。     

Monoclonal antibody AA4, which inhibits binding of IgE to high affinity receptors on rat basophilic leukemia cells, binds to novel alpha-galactosyl derivatives of ganglioside GD1b

Mouse monoclonal antibody AA4 inhibits the binding of IgE to high affinity IgE receptors on the rat basophilic leukemia cell line RBL-2H3. As shown by immunostaining of thin layer chromatograms, antibody AA4 binds avidly to two disialogangliosides (antigen I and antigen II) that occur in this cell line. The two antigens were purified by anion exchange chromatography followed by short-bed continuous thin-layer chromatography. About 230 micrograms of antigen I and 60 micrograms of antigen II were obtained from 20 g (wet weight) of leukemia cells. The structures of both purified antigens were determined to be alpha-galactosyl derivatives of the ganglioside GD1b by fast atom bombardment-mass spectrometry, by chemical ionization-mass spectrometry of permethylated samples, by gas chromatography-mass spectrometry of partially methylated alditol acetates, and by treatment with exoglycosidases and mild acid hydrolysis. The structure of antigen I is: (formula; see text) Antigen II has an additional alpha-galactosyl residue as follows: (formula; see text) The ceramide of antigen I contains approximately equal amounts of C24:0, C22:0, C20:0, C18:0, and C16:0 N-acyl fatty acids. The ceramide base is predominantly sphingosine along with a small amount of dihydrosphingosine. In contrast, the ceramide of antigen II contains mainly C24:0 N-acyl fatty acid with much lower amounts of C22:0, C20:0, and C18:0 fatty acids. Moreover, the ceramide base is approximately 55% sphingosine and 45% dihydrosphingosine. No unsaturated N-acyl fatty acids were detected in either antigen.

A general method for highly selective cross-linking of unprotected polypeptides via pH-controlled modification of N-terminal alpha-amino groups

A method is described for the highly selective modification of the alpha-amino groups at the N-termini of unprotected peptides to form stable, modified peptide intermediates which can be covalently coupled to other molecules or to a solid support. Acylation with iodoacetic anhydride at pH 6.0 occurs with 90-98% selectivity for the alpha-amino group, depending on the N-terminal residue (as shown with a series of model hexapeptides containing a competing Lys residue). Although Cys residues must be protected (reversibly or irreversibly) before the anhydride reaction, there are no detectable side reactions of the alpha-amino moiety--of the reagent or of modified peptide--with the side chains of His, Met, or Lys. The reaction works well in denaturants, so that inhibitory effects of noncovalent structure can be minimized. In a second step the iodoacetyl-peptide can be reacted with a thiol group on a protein, on a solid chromatography matrix, on a spectroscopic probe, etc. This is illustrated by reaction of a series of N alpha-iodoacetyl-peptides with murine interferon-gamma, which contains a C-terminal Cys residue. Data are presented which suggest that this iodoacetic anhydride scheme is superior in selectivity for alpha-amino groups to conventional chemical approaches to cross-linking such as use of 2-iminothiolane or N-hydroxysuccinimide-activated carboxylic acid esters. The reaction is ideally suited for modifying peptide fragments, as pure species or as mixtures, derived from proteolytic or chemical fragmentation of proteins. Furthermore, polypeptides synthesized biosynthetically, for example via recombinant DNA techniques, can be cross-linked in this way.(ABSTRACT TRUNCATED AT 250 WORDS)     

Sensory cues for the gradual frequency fall responses of the gymnotiform electric fish, Rhamphichthys rostratus

The sensory cues for a less known form of frequency shifting behavior, gradual frequency falls, of electric organ discharges (EODs) in a pulse-type gymnotiform electric fish, Rhamphichthys rostratus, were identified. We found that the gradual frequency fall occurs independently of more commonly observed momentary phase shifting behavior, and is due to perturbation of sensory feedback of the fish's own EODs by EODs of neighboring fish. The following components were identified as essential features in the signal mixture of the fish's own and the neighbor's EOD pulses: (1) the neighbor's pulses must be placed within a few millisecond of the fish's own pulses, (2) the neighbor's pulses, presented singly at low frequencies (0.2–4 Hz), were sufficient, (3) the frequency of individual pulse presentation must be below 4 Hz, (4) amplitude modulation of the sensory feedback of the fish's own pulses induced by such insertions of the neighbor's pulses must contain a high frequency component: sinusoidal amplitude modulation of the fish's own EOD feedback at these low frequencies does not induce gradual frequency falls. Differential stimulation across body surfaces, which is required for the jamming avoidance response (JAR) of wave-type gymnotiform electric fish, was not necessary for this behavior. Continuity,Collapse or Metamorphosis? Demographic anthropology and the study of change within human populations.Foreword

this first paper introduces the topic of the volume, outcomes of a symposium organised at the 13th ICAES held in Mexico in August 1993. It briefly reminds of the convergence between anthropology and demography up to date and stresses on the need of real transdisciplinary work in this challenging domain. The aim of the symposium was to make a contribution on the central theme chosen by the congress — the cultural and biological dimensions of global change — while examining the place of demographic anthropology in the study of change. The papers presented at the symposium have been organised in three parts which form the present volume: the composition of population, the choice of spouse and mobility, the reproduction and dynamics of populations. The basic mechanisms of change are considered through examples at the level of local populations. This also leads to question the definitions of human groups and to make a “declaration” stressing on the importance of individual heterogeneity and the arbitrariness and reductive nature of any grouping of individuals, stating therefore the misapprehension of the most recent scientific work inherent to the rationale of programs of “ethnic cleasing”.
