Short interval intracortical inhibition (SICI) of motor cortex, measured by transcranial magnetic stimulation (TMS) in a passive (resting) condition, has been suggested as a neurophysiological marker of hyperactivity in attention-deficit/hyperactivity disorder (ADHD). The aim of this study was to determine motor excitability in a go/nogo task at stages of response preparation, activation and suppression in children with ADHD, depending on the level of hyperactivity and impulsivity. Motor evoked potentials were recorded in 29 typically developing children and 43 children with ADHD (subdivided in two groups with higher and lower levels of hyperactivity/impulsivity; H/I-high and H/I-low). In the H/I-high group, SICI was markedly reduced in the resting condition and during response preparation. Though these children were able to increase SICI when inhibiting a response, SICI was still reduced compared to typically developing children. Interestingly, SICI at rest and during response activation were comparable, which may be associated with their hypermotoric behaviour. In the H/I-low group, response activation was accompanied by a pronounced decrease of SICI, indicating reduced motor control in the context of a fast motor response. In summary, different excitability patterns were obtained for the three groups allowing a better understanding of dysfunctional response activation and inhibition processes within the motor cortex in children with ADHD.  相似文献

Bioenergy crops may help achieve multiple energy, economic, and environmental objectives in the US Midwest, but a portfolio of crops must first be developed and tested in comparison to the current standard, maize (Zea mays L.). The nascent, but long-term and ongoing Landscape Biomass Project in Boone County, Iowa, USA examines five cropping systems including continuous maize, a modified maize–soy [Glycine max (L.) Merr.] rotation, nurse cropped maize–switchgrass (Panicum virgatum L.), double-cropped triticale (Triticosecale?×?Whit.)/sorghum (Sorghum bicolor (L.) Moench), and intercropped triticale–aspen (Crandon [Populus alba?×?Populus grandidentata]) over an elevation gradient. Here, we report perennial establishment and crop productivity across five landscape positions during the first 4 years (2009–2012) of this experiment. Perennials (switchgrass and aspen) established successfully across the gradient with minimal effect of landscape position. Continuous maize had the highest biomass yields both within and over growing seasons, but they declined over time. In comparison, the diversified and perennial systems had lower, but stable or increasing yields over time, despite extreme weather conditions. Landscape position did not consistently influence biomass yield; its effect depended on year and cropping system. Cropping system productivity was generally consistent across the landscape within a given year with greater variability between years. Findings help explain why landscape is often disregarded in Midwestern crop management: it does not seem to substantially drive crop, and thus economic, performance in the short term. Conversely, related Landscape Biomass studies find landscape influences important ecosystem functions (e.g., soil carbon storage) and should be an integral management consideration.  相似文献

The TIM23 translocase mediates the deltaPsi- and ATP-dependent import of proteins into mitochondria. We identified Tim14 as a novel component of the TIM23 translocase. Tim14 is an integral protein of the inner membrane with a typical J-domain exposed to the matrix space. TIM14 genes are present in the genomes of virtually all eukaryotes. In yeast, Tim14 is essential for viability. Mitochondria from cells depleted of Tim14 are deficient in the import of proteins mediated by the TIM23 complex. In particular, import of proteins that require the action of mtHsp70 is affected. Tim14 interacts with Tim44 and mtHsp70 in an ATP-dependent manner. A mutation in the HPD motif of the J-domain of Tim14 is lethal. Thus, Tim14 is a constituent of the mitochondrial import motor. We propose a model in which Tim14 is required for the activation of mtHsp70 and enables this chaperone to act in a rapid and regulated manner in the Tim44-mediated trapping of unfolded preproteins entering the matrix.  相似文献

The reaction between metmyoglobin and hydrogen peroxide results in the two-electron reduction of H2O2 by the protein, with concomitant formation of a ferryl-oxo heme and a protein-centered free radical. Sperm whale metmyoglobin, which contains three tyrosine residues (Tyr-103, Tyr-146, and Tyr-151) and two tryptophan residues (Trp-7 and Trp-14), forms a tryptophanyl radical at residue 14 that reacts with O2 to form a peroxyl radical and also forms distinct tyrosyl radicals at Tyr-103 and Tyr-151. Horse metmyoglobin, which lacks Tyr-151 of the sperm whale protein, forms an oxygen-reactive tryptophanyl radical and also a phenoxyl radical at Tyr-103. Human metmyoglobin, in addition to the tyrosine and tryptophan radicals formed on horse metmyoglobin, also forms a Cys-110-centered thiyl radical that can also form a peroxyl radical. The tryptophanyl radicals react both with molecular oxygen and with the spin trap 3,5-dibromo-4-nitrosobenzenesulfonic acid (DBNBS). The spin trap 5,5-dimethyl-1-pyrroline N-oxide (DMPO) traps the Tyr-103 radicals and the Cys-110 thiyl radical of human myoglobin, and 2-methyl-2-nitrosopropane (MNP) traps all of the tyrosyl radicals. When excess H2O2 is used, DBNBS traps only a tyrosyl radical on horse myoglobin, but the detection of peroxyl radicals and the loss of tryptophan fluorescence support tryptophan oxidation under those conditions. Kinetic analysis of the formation of the various free radicals suggests that tryptophanyl radical and tyrosyl radical formation are independent events, and that formation of the Cys-110 thiyl radical on human myoglobin occurs via oxidation of the thiol group by the Tyr-103 phenoxyl radical. Peptide mapping studies of the radical adducts and direct EPR studies at low temperature and room temperature support the conclusions of the EPR spin trapping studies.  相似文献

The Organization for Economic Co-operation and Development (OECD) has recently adopted Test Guideline 487 (TG487) for conducting the in vitro micronucleus (MNvit) assay. The purpose of this study is to evaluate and validate treatment conditions for the use of p53 competent TK6 human lymphoblastoid cells in a TG487 compliant MNvit assay. The ten reference compounds suggested in TG487 (mitomycin C, cytosine arabinoside, cyclophosphamide, benzo-a-pyrene, vinblastine sulphate, colchicine, sodium chloride, nalidixic acid and di(2-ethylhexyl)phthalate and pyrene) and noscapine hydrochloride were chosen for this study. In order to optimize the micronucleus response after treatment with some positive substances, we extended the recovery time after pulse treatment from 2 cell cycles recommended in TG487 to 3 cell cycles for untreated cells (40h). Each compound was tested in at least one of four exposure conditions: a 4h exposure followed by a 40h recovery, a 4h exposure followed by a 24h recovery, a 4h exposure in the presence of an exogenous metabolic activation system followed by a 40h recovery period, and a 27h continuous direct treatment. Results show that the direct acting clastogens, clastogens requiring metabolic activation and aneugens caused a robust increase in micronuclei in at least one test condition whereas the negative compounds did not induce micronuclei. The negative control cultures exhibited reproducibly low and consistent micronucleus frequencies ranging from 0.4 to 1.8% (0.8±0.3% average and standard deviation). Furthermore, extending the recovery period from 24h to 40h produced a 2-fold higher micronucleus frequency after a 4h pulse treatment with mitomycin C. In summary, the protocol described in this study in TK6 cells produced the expected result with model compounds and should be suitable for performing the MNvit assay in accordance with guideline TG487.  相似文献

Cronobacter spp. (formerly Enterobacter sakazakii) and Salmonella spp. are increasingly implicated internationally as important microbiological contaminants in low-moisture food products, including powdered infant formula. Estimates indicate that 40 to 80% of infants infected with Cronobacter sakazakii and/or Salmonella in the United States may not survive the illness. A systematic approach, combining literature-based data mining, comparative genome analysis, and the direct sequencing of PCR products of specific biomarker genes, was used to construct an initial collection of genes to be targeted. These targeted genes, particularly genes encoding virulence factors and genes responsible for unique phenotypes, have the potential to function as biomarker genes for the identification and differentiation of Cronobacter spp. and Salmonella from other food-borne pathogens in low-moisture food products. In this paper, a total of 58 unique Salmonella gene clusters and 126 unique potential Cronobacter biomarkers and putative virulence factors were identified. A chitinase gene, a well-studied virulence factor in fungi, plants, and bacteria, was used to confirm this approach. We found that the chitinase gene has very low sequence variability and/or polymorphism among Cronobacter, Citrobacter, and Salmonella, while differing significantly in other food-borne pathogens, either by sequence blasting or experimental testing, including PCR amplification and direct sequencing. This computational analysis for Cronobacter and Salmonella biomarker identification and the preliminary laboratory studies are only a starting point; thus, PCR and array-based biomarker verification studies of these and other food-borne pathogens are currently being conducted.  相似文献

Summary We have established in vitro culture conditions for globular zygotic wheat embryos (Triticum aestivum L.). Their nutritional requirements have been systematically investigated. The initial sucrose concentration, as well as the sucrose concentration during the culture, a 6-benzylaminopurine supplement, the use of nitrates and ammonium as nitrogen source have a major influence on the embryo development. Proline has an inhibitory effect on the germination. A double layer system with different media was used to give a continuous variation of the medium composition with time. K+ gradient-dependent rubidium flux in vesicles obtained from stimulated rabbit stomach distinguishes two cation pathways. Selective inhibition by vanadate and the (1,2-alpha)-imidazopyridine, SCH 28080 identifies one pathway as H,K-ATPase-mediated passive cation exchange. A second pathway, additive to the first, is inhibited by the protonophore, tetrachlorosalicylanilide and is identified as a K+ conductance pathway present in these vesicles. The conductance was limited to vesicle populations obtained from the stimulated rabbit gastric mucosa and was distributed into both a light microsomal fraction and a heavier membrane fraction. 86Rb+ transport through the cation conductance exhibited a trans-stimulated cation selectivity sequence of K+ greater than Rb+ = Cs+ much greater than Li+. Potential sensitive flux was inhibited by the cyanine dye 3,3'-dipropyl-2,2'-thiodicarbo cyanine iodide, Ba2+, quinine, and the guanidinium compound 1,8-bis-guanidinium-n-octane. The presence of the conductance was correlated with K+-dependent H+ transport which did not require prolonged equilibration in K+ medium for activation. A role for the stimulus-dependent K+ conductance in gastric acid secretion could be its provision of a pathway for net K+ movement to the luminal site of the H,K-ATPase.  相似文献
