Bean plants ( Phaseolus vulgaris L. cv. Scarlett), germinated in darkness for I week, were transferred to light (200 μmol m−2 s−1) and cultivated for I week in a complete nutrient solution. After this period, cadmium ions in the form of CdSO4 were added at the concentrations of 0.10.20 and 50 μ M . The effects of this metal on the properties of photosystem II photochemistry were studied by means of modulated fluorescence analysis. Steady state photochemical quenching. non-photochemical quenching and terminal fluorescence were determined in control and cadmiumtreated plants. We postulate that, during short term exposure of plants to cadmium in the early stages of growth, the Calvin cycle reactions are more likely than photosystem II to be the primary target of the toxic influence of cadmium. The reduced demand for ATP and NADPH upon Calvin cycle inhibition causes a down-regulation of photosystem II photochemistry and of the yield of linear electron transport.  相似文献

The possibility of a role for phosphate metabolism in the photosynthetic regulation that occurs during frost hardening was investigated in winter rye (Secale cereale L. cv. Musketeer). Leaves of frost-hardened and non-hardened winter rye were studied during photosynthetic induction, and at steady state after being allowed to take up 20 mM orthophosphate through the transpiration stream for 3 h. At the growth irradiance (350 mol·m-2·s-1) frost-hardening increased the stationary rate of CO2-dependent O2 evolution by 57% and 25% when measured at 5 and 20° C, respectively. Frosthardening also reduced the lag phase to stationary photosynthesis by 40% at 5° C and decreased the susceptibility of leaves to oscillations during induction and after interruption of the actinic beam during steady-state photosynthesis. These responses are all indicative of increased phosphate availability in frost-hardened leaves. As reported previously by Öquist and Huner (1993, Planta 189, 150–156), frost-hardening also decreased the reduction state of QA, the primary, stable quinone acceptor of PSII, and decreased the sensitivity of winter rye to photoinhibition of photosynthesis. Non-hardened rye leaves fed orthophosphate also showed an increased photosynthetic capacity (25% at 20° C and light saturation), lower reduction state of QA, a reduced sensitivity to photoinhibition and lower susceptibility to oscillations resulting from a brief interruption of the actinic light. In this study, the plant node was dried in an oven (40, 50 and 60 °C), shade and temperature-controlled microwave (40, 50 and 60 °C) methods. Statistically (p<0.05), the values closest to the color values of fresh grass were determined in an oven at 40 °C drying temperature. Effective diffusion values varied between 8.85×10−8–5.65×10−6 m2 s−1. While the activation energy was 61.28 kJ mol−1 in the oven, it was calculated as 85.24 kJ mol−1 in the temperature-controlled microwave. Drying data was best estimated in the Midilli-Küçük (R2 0.9998) model oven at 50 °C. The highest SMER value was calculated as 0.0098 kg kWh−1 in the temperature-controlled microwave drying method. The lowest SEC value in the temperature-controlled microwave was determined as 24.03 kWh kg−1. It was determined that enthalpy values varied between −2484.66/−2623.38 kJ mol−1, entropy values between −162.04/−122.65 J mol−1 and Gibbs free energy values between 453335.22–362581.40 kJ mol−1. Drying rate values were calculated in the range of 0.0127–0.9820 g moisture g dry matter−1 in the temperature-controlled microwave, 0.0003–0.0762 g dry matter−1 in the oven, and 0.001–0.0058 g moisture g dry moisture matter−1 in the shade. Phenolic content 6957.79 μg GAE g−1 fw - 48322.27 μg GAE g−1 dw, flavonoid content 3806.67 mg KE L−1 fw - 22200.00 mg KE L−1 dw and antioxidant capacity 43.35 μmol TE g−1 fw - 323.47 μmol TE g−1 dw. The highest chlorophyll values were obtained from samples dried in an oven at 40 °C. According to the findings, it is recommended to dry the knotweed (Polygonum cognatum Meissn.) plant in a temperature-controlled microwave oven at low temperatures. In this study, in terms of drying kinetics and energy parameters, a temperature-controlled microwave dryer of 60 °C is recommended, while in terms of quality characteristics, oven 40 °C and shade methods are recommended.  相似文献

Lactoperoxidase enzyme (LPO) is secreted from salivary, mammary, and other mucosal glands including the bronchi, lungs, and nose, which had functions as a natural and the first line of defense towards viruses and bacteria. In this study, methyl benzoates were examined in LPO enzyme activity. Methyl benzoates are used as precursors in the synthesis of aminobenzohydrazides used as LPO inhibitors. For this purpose, LPO was purified in a single step using sepharose-4B-l-tyrosine-sulfanilamide affinity gel chromatography with a yield of 9.91 % from cow milk. Also, some inhibition parameters including the half maximal inhibitory concentration (IC50) value and an inhibition constant (Ki) values of methyl benzoates were determined. These compounds inhibited LPO with Ki values ranging from 0.033±0.004 to 1540.011±460.020 μM. Compound 1 a (methyl 2-amino-3-bromobenzoate) showed the best inhibition (Ki=0.033±0.004 μM). The most potent inhibitor ( 1 a ) showed with a docking score of −3.36 kcal/mol and an MM-GBSA value of −25.05 kcal/mol, of these methyl benzoate derivatives ( 1 a – 16 a ) series are established H-bond within the binding cavity with residues Asp108 (distance of 1.79 Å), Ala114 (distance of 2.64 Å), and His351 (distance of 2.12 Å).  相似文献

Data independent acquisition (DIA/SWATH) MS is a primary strategy in quantitative proteomics. diaPASEF is a recent adaptation using trapped ion mobility spectrometry (TIMS) to improve selectivity/sensitivity. Complex DIA spectra are typically analyzed with reference to spectral libraries. The best-established method for generating libraries uses offline fractionation to increase depth of coverage. More recently strategies for spectral library generation based on gas phase fractionation (GPF), where a representative sample is injected serially using narrow DIA windows that cover different mass ranges of the complete precursor space, have been introduced that performed comparably to deep offline fractionation-based libraries. We investigated whether an analogous GPF-based approach that accounts for the ion mobility (IM) dimension is useful for the analysis of diaPASEF data. We developed a rapid library generation approach using an IM-GPF acquisition scheme in the m/z versus 1/K0 space requiring seven injections of a representative sample and compared this with libraries generated by direct deconvolution-based analysis of diaPASEF data or by deep offline fractionation. We found that library generation by IM-GPF outperformed direct library generation from diaPASEF and had performance approaching that of the deep library. This establishes the IM-GPF scheme as a pragmatic approach to rapid library generation for analysis of diaPASEF data.  相似文献

Summary Kluyveromyces spp. have been found to be more efficient than a CUP1R strain of S. cerevisiae in heavy metal resistance and accumulation. The present study describes the subcellular distribution of the accumulated metals (Ag, Cd, Cu) in S. cerevisiae and K. marxianus. Absorption by insoluble cellular material of the metals appears as the main mechanism of metal accumulation in both organisms.  相似文献

Serum copper, zinc levels, and the Cu/Zn ratio were evaluated in 31 patients with breast cancer and 35 healthy controls. Copper and zinc were determined by atomic absorbtion spectrophotometry. The mean serum copper level and the mean Cu/Zn ratio in patients with breast cancer were significantly higher than the control group (p<0.001 andp<0.001). In addition, the mean serum zinc level in patients with breast cancer was significantly lower than the control group (p<0.001). Neither serum copper and zinc levels nor the Cu/Zn ratio were of value in discriminating of the disease activity and severity. Interestingly, the Cu/Zn ratio in premenopausal patients was higher than postmenopausal patients (p<0.05) and this was not related to age. The further combined biological and epidemiological studies are necessary to investigate the roles of copper and zinc in breast cancer.  相似文献

The growth of humic acids, prepared by a gentle method from two different kinds of soils (I and II), has been studied using small-angle neutron scattering at an acidity corresponding to pH 5.0 and 0.10 M ionic strength (NaCl). Humic acids aggregate either to large clusters with a fractal dimension of 2.35 and an average diameter of 1720 (I) or to clusters with an average diameter of 700 (II). After storage for 2 days at 4 °C, the latter aggregates (II) formed a gel. In a step toward gelation, we observed cluster-cluster interaction from the neutron-scattering data in the form of a correlation peak. These differences in size can be explained by assuming that the smaller particles (II) are trapped into a nonequilibrium state characterized by the temperature-solvent condition. The importance of a humic acid gel network for the conservation of water and nutrients in the environment is discussed.  相似文献

Two reproductively isolated demes of brown trout coexist in a small Swedish mountain lake, Lake Bunnersj?rna. We electrophoretically examined 102 specimens from that lake for 27 enzymes encoded by 54 loci. The two demes are fixed for different alleles at a lactate dehydrogenase locus (LDH-1); statistically significant allele frequency differences at five other loci further support the complete lack of gene flow between these demes. There are significant differences in growth rates between fish in the two demes, but no further morphological differentiation h-s been detected.--In light of these findings, the genetic distance between these populations is surprisingly small (Nei's I = 0.975). These demes represent one of the least genetically divergent, reproductively isolated sympatric pair of vertebrate populations that have been identified. The results are discussed from both an evolutionary and ecological perspective.  相似文献
