A suite of muscarinic receptor blockers was used to characterize the receptor(s) mediating the contractile effect of acetylcholine (ACh) on isolated rings of ventral aorta from the dogfish shark, Squalus acanthias. The M2/M4-specific inhibitor N,N’-bis(6-{[(2-methoxyphenyl) methyl] amino} hexyl) -1,8- octane diamine tetrahydrochloride (methoctramine) did not reduce the efficacy of ACh, and the M3-specific inhibitor 4-diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP) displaced the ACh concentration-response curve to the right at much lower concentrations than the M1-specific inhibitor (5-11-dihydro-11- [4-methyl-l-piperazinyl)acetyl] -6H-pyrido[2,3-b] [1,4] benzodiazepin-6-one dihydrochloride) (pirenzepine). It appears, therefore, that an M3-type muscarinic receptor is expressed in the aortic vascular smooth muscle of the dogfish shark.  相似文献

In chicken pot pies, it was not possible to promote the growth of appreciable numbers of staphylococci under any condition of defrost. The pies spoiled under the same conditions, however. In macaroni and cheese dinners, tremendous numbers of saprophytic bacteria developed, but only after extended incubation at room temperature in which even spoilage was carried to an extreme. Under these conditions, staphylococci also multiplied vigorously. At the extreme temperature of 37 C, rank spoilage of such an advanced state as to render the product completely inedible was reached in less than 24 hr. The physiological site of ethylene action on CO2 assimilation was investigated in intact plants of Glycine max L., using a whole-plant, open exposure system equipped witha remotely operated single-leaf cuvette. The objective of the study was met by investigating in control and ethylene-treated plants the (a) synchrony in response of CO2 assimilation, stomatal conductance to water vapor, and substomatal CO2 partial pressure; (b) response of CO2 assimilation as a function of a range of substomatal CO2 partial pressures; and (c) response of CO2 assimilation as a function of a range of photon flux densities. After exposure to 410 micromoles per cubic meter of ethylene for 2.0 hours, CO2 assimilation and stomatal conductance declined in synchrony, while substomatal CO2 partial pressure remained unchanged until exposure times equaled and exceeded 3.0 hours. Because incipient changes in CO2 assimilation occurred without a change in the CO2 partial pressure in the leaf interior, it is concluded that both stomatal physiology and the chloroplast's CO2 assimilatory capacity were initial sites of ethylene action. After 3.5 hours the effect of ethylene on stomatal conductance and CO2 assimilation exhibited saturation kinetics, and the effect was substantially more pronounced for stomatal conductance than for CO2 assimilation. Based on the response of CO2 assimilation to a range of substomatal CO2 partial pressures, ethylene did not affect either the CO2 compensation point or carboxylation efficiency at subsaturating CO2 partial pressures. Above-ambient supplies of CO2 did not alleviate the diminished rates of CO2 assimilation. In partitioning the limitations imposed on CO2 assimilation in control and ethylene-treated plants, the stomatal component accounted for only 16 and 4%, respectively. The response of CO2 assimilation to a range of photon flux densities suggests that ethylene reduced apparent quantum yield by nearly 50%. Thus, the pronounced decline in net photosynthetic CO2 assimilation in the presence of ethylene was due more to a loss in the mesophyll tissue's intrinsic capacity to assimilate CO2 than to a reduction in stomatal conductance.  相似文献

Cutaneous basophil hypersensitivity (CBH) reactions are a heterogeneous group of delayed time course basophil-rich immune responses that can be mediated in the guinea pig by T cells, B cells, or IgG1 antibody. This study examined whether guinea pig IgE antibody could also mediate CBH reactions. IgE antibody to picryl or oxazolone determinants was induced by immunizing Hartley strain guinea pigs pretreated with cyclophosphamide. Hyperimmune serum from these animals was passed through a heavy chain-specific anti-IgG1 affinity column. The presence of IgE anti-hapten antibody in the filtrate fraction was verified by passive cutaneous anaphylaxis (PCA) testing with a 7-day period of local passive sensitization and by the heat lability (56 degrees C, 4 hr) of PCA activity. This IgE-rich fraction and the IgG1 fraction eluted from the column with base (0.2 M Na2CO3, pH 11.3) were transferred i.v. to separate groups of normal guinea pigs. Both fractions mediated delayed time course reactions that contained basophils. Macroscopic and microscopic reactions mediated by the IgE-rich fraction were abolished with heat (56 degrees C, 4 hr). Thus, two antigen-specific factors in guinea pig serum can mediate delayed time course basophil-containing reactions: IgG1 and IgE antibodies. IgE-mediated CBH reactions are similar to the late-phase reaction that follows IgE-dependent wheal-and-flare reactions in humans. The finding that guinea pig IgE can mediate a late reaction that contains basophils makes this a possible model for the human late-phase response, and suggests that some forms of CBH may play a role in human allergic disease.  相似文献

A low relative area change (RAC) of the proximal pulmonary artery (PA) over the cardiac cycle is a good predictor of mortality from right ventricular failure in patients with pulmonary hypertension (PH). The relationship between RAC and local mechanical properties of arteries, which are known to stiffen in acute and chronic PH, is not clear, however. In this study, we estimated elastic moduli of three PAs (MPA, LPA and RPA: main, left and right PAs) at the physiological state using mechanical testing data and correlated these estimated elastic moduli to RAC measured in vivo with both phase-contrast magnetic resonance imaging (PC-MRI) and M-mode echocardiography (on RPA only). We did so using data from a canine model of acute PH due to embolization to assess the sensitivity of RAC to changes in elastic modulus in the absence of chronic PH-induced arterial remodeling. We found that elastic modulus increased with embolization-induced PH, presumably a consequence of increased collagen engagement, which corresponds well to decreased RAC. Furthermore, RAC was inversely related to elastic modulus. Finally, we found MRI and echocardiography yielded comparable estimates of RAC. We conclude that RAC of proximal PAs can be obtained from either MRI or echocardiography and a change in RAC indicates a change in elastic modulus of proximal PAs detectable even in the absence of chronic PH-induced arterial remodeling. The correlation between RAC and elastic modulus of proximal PAs may be useful for prognoses and to monitor the effects of therapeutic interventions in patients with PH.  相似文献

Many splicing factors interact with both mRNA and pre-mRNA. The identification of these interactions has been greatly improved by the development of in vivo cross-linking immunoprecipitation. However, the output carries a strong sampling bias in favor of RNPs that form on more abundant RNA species like mRNA. We have developed a novel in vitro approach for surveying binding on pre-mRNA, without cross-linking or sampling bias. Briefly, this approach entails specifically designed oligonucleotide pools that tile through a pre-mRNA sequence. The pool is then partitioned into bound and unbound fractions, which are quantified by a two-color microarray. We applied this approach to locating splicing factor binding sites in and around ∼4000 exons. We also quantified the effect of secondary structure on binding. The method is validated by the finding that U1snRNP binds at the 5′ splice site (5′ss) with a specificity that is nearly identical to the splice donor motif. In agreement with prior reports, we also show that U1snRNP appears to have some affinity for intronic G triplets that are proximal to the 5′ss. Both U1snRNP and the polypyrimidine tract binding protein (PTB) avoid exonic binding, and the PTB binding map shows increased enrichment at the polypyrimidine tract. For PTB, we confirm polypyrimidine specificity and are also able to identify structural determinants of PTB binding. We detect multiple binding motifs enriched in the PTB bound fraction of oligonucleotides. These motif combinations augment binding in vitro and are also enriched in the vicinity of exons that have been determined to be in vivo targets of PTB.  相似文献
