Summary Hydrogen is consumed by methanogenic, sulphate-reducing, and homoacetogenic bacteria and members of these bacterial groups are able to grow chemolithotrophically with hydrogen as sole energy source. Cathodic hydrogen consumption by sulphate-reducing bacteria has been proposed as one of the factors in the anaerobic corrosion of metals. Desulfovibrio spp. were able to utilize cathodic hydrogen from mild steel as the only source of energy for growth with sulphate or nitrate as terminal electron acceptor. Other hydrogen-oxidizing bacteria such as Methanospirillum hungatei, Acetobacterium woodii and Wolinella succinogenes were also able to utilize cathodic hydrogen from mild steel for energy generation and growth. Weight loss studies of mild steel coupons under different growth conditions of Desulfovibrio spp. indicated that hydrogen removal alone is not the cause of corrosion and the depolarization phenomenon probably plays a role only in the initiation of the anaerobic microbial corrosion process.  相似文献

A variety of compounds were investigated for use as sulfur sources for the growth of methanogenic bacteria.Methanococcus (Mc.) deltae, Mc. maripaludis, Methanobacterium (Mb.) speciesGC-2B, GC-3B, andMMY, Methanobrevibacter (Mbr.) ruminantium, andMethanosarcina (Ms.) barkeri strain 227 grew well with sulfide, So, thiosulfate, or cysteine as sole sulfur source.Mbr. ruminatium was able to grow on SO 4 = or SO 3 = , andMs. barkeri strain 227 was able to grow on SO 3 = , but not on SO 4 = as a sole sulfur source.Mc. jannaschii grew with sulfide, So, thiosulfate or SO 3 = , but not on cysteine or SO 4 = as sole surface source.Mc. thermolithotrophicus, Mc. jannaschii, Mc. deltae, andMb. thermoautotrophicum strains Marburg and H were able to grow with methanethiol, ethanethiol,n-propanethiol,n-butanethiol, methyl sulfide, dimethyl sulfoxide, ethyl sulfide, or CS2 as a sulfur source, when very low levels (20–30 M) of sulfide were present; no growth occurred on 5–100 M sulfide alone. Byssus thread production ofD. polymorpha under different conditions of temperature, salinity and agitation were studied in the laboratory. The acclimation to salinity and temperature greatly affects the byssus production ofD. polymorpha. Byssus production of mussels was significantly reduced when temperature increased beyond 20°C and decreased below 10°C. Mussels with cut threads (for counting), produced a substantially increased number of threads. However, mussels with uncut byssus threads were comparatively more mobile. Byssus production of mussels did not vary significantly at salinities up to 3. Beyond this salinity byssus production was reduced significantly. Mussels increased their byssus production with increasing frequency of agitation.  相似文献

Root treatments of barley (Hordeum distichum L.) plants with 10-7 to 10-4 M abscisic acid (ABA) caused an increase in proline content, especially at higher concentrations, within 2–3 h. Even 3 h after the removal of ABA from the medium the plants continued to accumulate proline. The higher the concentration of the ABA, the higher was the proline level at 6 h. When the highest ABA concentration, 10-4 M, was tested with polyethylene glycol (PEG) (-5.0 bars) in the medium, the ABA treatment resulted in a higher proline content than in control plants. The treatments PEG alone and PEG + ABA resulted in heavy accumulation of proline, especially, 3 h after releasing the plants from the stress. The proline content in PEG+ABA-treated plants was always higher than plants treated with PGE or ABA alone. In peas (Pisum sativum L. cv Alaska) the same trend occurred although to a lesser degree. Liver-derived progenitor cells (LDPCs) are recently identified novel stem/progenitor cells from healthy, unmanipulated adult rat livers. They are distinct from other known liver stem/progenitor cells such as the oval cells. In this study, we have generated a LDPC cell line RA1 by overexpressing the simian virus 40 (SV40) large T antigen (TAg) in primary LDPCs. This cell line was propagated continuously for 55 passages in culture, after which it became senescent. Interestingly, following transformation with SV40 TAg, LDPCs decreased in size significantly and the propagating cells measured 1 μm in diameter. RA1 cells proliferated in vitro with a doubling time of 5-7 days, and expressed cell surface markers of LDPCs. In this report, we describe the characterization of this novel progenitor cell line that might serve as a valuable model to study liver cell functions and stem cell origin of liver cancers.  相似文献

Abstract

A complete three dimensional model for the LexA repressor dimer bound to the recA operator site consistent with relevant biochemical and biophysical data for the repressor was proposed from our laboratory when no crystal structure of LexA was available. Subsequently, the crystal structures of four LexA mutants Δ1–67 S119A, S119A, G85D and Δ1-67 quadruple mutant in the absence of operator were reported. It is examined in this paper to what extent our previous model was correct and how, using the crystal structure of the operator-free LexA dimer we can predict an improved model of LexA dimer bound to recA operator. In our improved model, the C-domain dimerization observed repeatedly in the mutant operator-free crystals is retained but the relative orientation between the two domains within a LexA molecule changes. The crystal structure of wild type LexA with or without the recA operator cannot be solved as it autocleaves itself. We argue that the ‘cleavable’ cleavage site region found in the crystal structures is actually the more relevant form of the region in wildtype LexA since it agrees with the value of the pre-exponential Arrhenius factor for its auto- cleavage, absence of various types of trans-cleavages, difficulty in modifying the catalytic serine by diisopropyl flourophosphate and lack of cleavage at Arg 81 by trypsin; hence the concept of a ‘conformational switch’ inferred from the crystal structures is meaningless.  相似文献

The meaning and implications of Hindu nationalist expression in the United States are different from those at home, but both are linked through transnational circuits of communication and exchange. In India, the invocation of religion summons up the unresolved debates between nationalism and social reform, and presents Hinduism as an implicitly conservative force. By contrast, in the US, Hindu religion is more self-consciously a medium of cultural reproduction. This article points to the ways in which Hindu nationalism seeks and promotes transnational affiliations even while espousing a rhetoric of insularity, cultural pride and self-sufficiency.  相似文献
