In this study we have addressed the ability of the glycolipid transfer protein (GLTP) to transfer anthrylvinyl-galactosylceramide at different pH and sodium chloride concentrations, and the ability of three different mutants to transfer the fluorescently labeled galactosylceramide between donor and acceptor model membranes. We constructed single tryptophan mutants with site-directed mutagenesis where two of the three tryptophan (W) of wild-type human GLTP were substituted with phenylalanine (F) and named W85 GLTP (W96F and W142F), W96 GLTP (W85F and W142F) and W142 GLTP (W85F and W96F) accordingly. Wild-type GLTP and W96 GLTP were both able to transfer anthrylvinyl-galactosylceramide, but the two variants W85 GLTP and W142 GLTP did not show any glycolipid transfer activity, indicating that the tryptophan in position 96 is crucial for transfer activity. Tryptophan fluorescence emission showed a blue shift of the maximal emission wavelength upon interaction of glycolipid containing vesicle with wild-type GLTP and W96 GLTP, while no blue shift was recorded for the protein variants W85 GLTP and W142 GLTP. The quantum yield of tryptophan emission was highest for the W96 GLTP protein whereas W85 GLTP, W142 GLTP and wild-type GLTP showed a lower and almost similar quantum yield. The lifetime and anisotropy decay of the different tryptophan mutants also changed upon binding to vesicles containing galactosylceramide. Again wild-type GLTP and W96 GLTP showed similar behavior in the presence of vesicles containing glycolipids. Taken together, our data show that the W96 is involved not only in the activity of the protein but also in the interaction between the protein and glycolipid containing membranes.  相似文献

GsMTx-4, a polypeptide from the spider Grammostola spatulata, is an inhibitor of mechanosensitive channels. It is known to interact with lipid membranes, suggesting it partitions into the membrane to alter the channel gating, but the effect of the membrane charge on GsMTx-4 activity remains unknown. In this study, we found that GsMTx-4 more effectively interacts with anionic lipids than zwitterionic ones. The effect of GsMTx-4 on negatively charged membranes was similar to that of the antimicrobial peptide melittin, which led us to assess GsMTx-4's antimicrobial activity. Interestingly, we found that, in contrast to other neurotoxins, GsMTx-4 exhibited antimicrobial properties and was more active against Gram-positive than Gram-negative bacteria. These results suggest that GsMTx-4 exerts its antimicrobial effect by altering the packing of the membrane and/or inhibiting mechanosensitive channels. These findings could point the way towards a new class of antimicrobial peptides.  相似文献

The influence of the degree of cross-linking (DCL) on chitosan beads was studied. Chitosan was prepared from the exoskeleton of Cape rock-lobsters, collected from the surroundings of Cape Town, South Africa. The chitosan beads were characterized; the beads water contents and pKa varied in the range of 90-96% and 4.3-6.0, respectively, and were found to decrease with increasing DCL (0.0-34.0%). A pH-model, which described the reversibility of the metal adsorbed onto the beads, was used to predict the equilibrium properties of copper adsorption onto the cross-linked beads. The model accounts for the effect of pH and the important model parameters, the equilibrium adsorption constant (Kads) and to a lesser extent the adsorbent adsorption capacity (qmax) showed to decrease with the DCL. The adsorbent capacity and the adsorption constant were determined as 3.8-5.0mmol/g chitosan and (9-90)x10(-4), respectively. The adsorption kinetics could be described using a shrinking core model and the effective diffusion coefficient (Deff) was determined as (8.0-25.8)x10(-11)m2/s. It was found that Deff decreases with the DCL mainly due to the decreased in water content of the beads at high DCL.  相似文献

Vero cell cultures are used in the quality control of Diphtheria vaccines: to estimate vaccine potency and to determine residual toxicity and reversion to toxicity. The impact of replacing foetal calf serum containing medium (SCM) by serum free media (SFM) on the sensitivity of Vero cells to Diphtheria Toxin was studied. Compared to SCM, SFM showed an eight-fold decrease in sensitivity to Diphtheria Toxin. This decrease was almost immediate, indicating that this phenomenon was not caused by a change in membrane structure or protein expression. We investigated the effect of SFM on Diphtheria Toxin in order to determine the cause of the decrease in sensitivity. Our results show that oligopeptides, which are often used in SFM as part of the replacement of foetal calf serum, are the most likely cause.  相似文献

The genus Rosa has a complex evolutionary history caused by several factors, often in conjunction: extensive hybridization, recent radiation, incomplete lineage sorting, and multiple events of polyploidy. We examined the applicability of AFLP markers for reconstructing (species) relationships in Rosa, using UPGMA clustering, Wagner parsimony, and Bayesian inference. All trees were well resolved, but many of the deeper branches were weakly supported. The cluster analysis showed that the rose cultivars can be separated into a European and an Oriental cluster, each being related to different wild species. The phylogenetic analyses showed that (1) two of the four subgenera (Hulthemia and Platyrhodon) do not deserve subgeneric status; (2) section Carolinae should be merged with sect. Cinnamomeae; (3) subsection Rubigineae is a monophyletic group within sect. Caninae, making sect. Caninae paraphyletic; and (4) there is little support for the distinction of the five other subsections within sect. Caninae. Comparison of the trees with morphological classifications and with previous molecular studies showed that all methods yielded reliable trees. Bayesian inference proved to be a useful alternative to parsimony analysis of AFLP data. Because of their genome-wide sampling, AFLPs are the markers of choice to reconstruct (species) relationships in evolutionary complex groups.  相似文献

Piscidin 2 (P2), a 22-residue cationic peptide isolated from the mast cells of hybrid striped bass, has potent antibacterial activities. However, its antifungal properties are not completely understood. In the current study, we investigated the antifungal effects and mode of action of P2. P2 exhibited potent antifungal activity against human pathogenic fungi. To understand the fungicidal properties of P2, we focused on a membrane-active mechanism of the peptide by in vivo and in vitro testing. Flow cytometric analysis using bis-(1,3-dibutylbarbituric acid) trimethine oxonol [DiBAC4(3)] and protoplast regeneration experiments showed that P2 caused fungal membrane damage. Furthermore, fluorescence analysis using 1,6-diphenyl-1,3,5-hexatriene (DPH) revealed that P2 created pores in fungal membranes. These results were confirmed with dye leakage tests by using liposomes composed of phosphatidylcholine/phosphatidylserine (3:1, w/w), which mimicked fungal membranes. The present study indicated that P2 exerts its fungicidal effects by perturbing membrane activities.  相似文献

Microscopy has been used to diagnose the effects of air pollutantson conifers. As sampling may be slow, it is important to knowwhether the structure of needles changes during the day. Usinglight- and electron microscopy, diurnal changes in the structureof mesophyll cells of current year needles from 6-year-old Norwayspruce saplings were studied on one day in early autumn, 1995.Saplings had been grown in open-top chambers and exposed toozone and drought stress during the 1992–1995 growingseasons. In all treatments, the proportion of cells with smallcytoplasmic vacuoles and with large cytoplasmic lipid depositsdecreased during the day. Mitochondria increased in size fromearly morning to midday, after which time they shrank progressively.Ozone affected the shape of mitochondria: the proportion oftube-shaped mitochondria decreased from early morning in controlsaplings, whereas it increased in ozone-treated saplings. Starchaccumulated during the day, but to a lesser extent in drought-stressedsaplings than well-watered controls. Microbodies were smallerin drought-stressed saplings than in controls early in the morning,but their size then increased compared to that of the controls.Cellular and sub-cellular disorganization, probably due to sub-optimalfixation procedures, were detected in all treatments, especiallyin the morning, but these decreased later, particularly in well-wateredsaplings. Results show that microscopy is a sensitive and validtool for studying stress responses in plants. However, the timeof sample collection must be considered and documented to avoidmisleading conclusions due to actual diurnal changes and fixationproblems.
