The essential oil obtained from fresh leaves of Eucalyptus teretecornis (family Myrtaceae) was analysed by gas chromatography/mass spectrometry (GC/MS). Twenty eight compounds were identified and ??-pinene (22.55%), ??-pinene (22.50%), 1,8-cineole (19.84%), limonene (5.62%), ??-fenchol (3.10%), ??-phellandrene (2.90%), ??-eudesmol (2.66%) and 4-(2-methylcyclohex-1-enyl)-but-2-enal (2.34%) were the main components. The antifungal activity of the essential oil was assayed against Alternaria alternata using bioautography. Two main bioactive components namely a1 (R f ?=?0.27) and a2 (R f ?=?0.33) were observed that produced inhibition zone of 4?mm and 8?mm in diameter respectively. The minimum inhibitory amount (MIA) of a1 and a2 against A. alternata was determined as 28???g and 10???g, respectively using bioautography assay. Components corresponding to a1 and a2 were determined as ??-fenchol (oxygenated monoterpene) and ??-eudesmol (oxygenated sesquiterpene) respectively using GC/MS analysis. The antioxidant activity of the essential oil and its bioactive fraction was evaluated by DPPH radical scavenging assay, ??-carotene/linoleic acid bleaching assay, reducing power assay and metal chelating assay. In addition fraction of the essential oil that showed antioxidant activity was analyzed using GC/MS and ??-fenchol, 4-terpineol and carvacrol were the main components.  相似文献

Development of micronutrient enriched staple foods is an important breeding goal in view of the extensive problem of ‘hidden hunger’ caused by micronutrient malnutrition. In the present study, kernel iron (Fe) and zinc (Zn) concentrations were evaluated in a set of 31 diverse maize inbred lines in three trials at two locations – Delhi (Kharif 2007 & 2008) and Hyderabad (Rabi 2007–08). The ranges of kernel Fe and Zn concentrations were 13.95–39.31 mg/kg and 21.85–40.91 mg/kg, respectively, across the three environments. Pooled analysis revealed significant genotype × environment (G × E) interaction in the expression of both the micronutrient traits, although kernel Fe was found to be more sensitive to G × E as compared to kernel Zn. Seven inbred lines, viz., BAJIM-06-03, DQPM-6, CM212, BAJIM-06-12, DQPM-7, DQPM-2 and CM129, were found to be the most stable and promising inbred lines for kernel Zn concentration, while for kernel Fe concentration, no promising and stable genotypes could be identified. Analysis of molecular diversity in 24 selected inbred lines with phenotypic contrast for the two kernel micronutrient traits, using 50 SSR markers covering the maize genome, revealed high levels of polymorphisms (214 SSR alleles; mean PIC value?=?0.62). The phenotypically contrasting and genetically diverse maize inbred lines identified in this study could be potentially utilized in further studies on QTL analysis of kernel micronutrient traits in maize, and the stable and most promising kernel micronutrient-rich maize genotypes provide a good foundation for developing micronutrient-enriched maize varieties suitable for the Indian context.  相似文献

Induction and maintenance of immunologic tolerance in humans remains a desirable but elusive goal. Therefore, understanding the physiologic mechanisms of regulation of immune responses is highly clinically relevant for immune-mediated diseases (e.g., autoimmunity and asthma/allergy) and for cell and organ transplantation. Acceptance of the fetus, which expresses paternally inherited alloantigens, by the mother during pregnancy is a unique example of how the immune system reshapes a destructive alloimmune response to a state of tolerance. Understanding the complex mechanisms of fetomaternal tolerance has important implications for developing novel strategies to induce immunologic tolerance in humans in general and for prevention of spontaneous abortion in at-risk populations in particular.  相似文献

Acceptance of the fetus expressing allogeneic paternal Ags by the mother is a physiologic model of transplantation tolerance. Various mechanisms contribute to fetal evasion from immune attack by maternal leukocytes. We have recently demonstrated that the inhibitory costimulatory molecule PDL1 plays a critical role in fetomaternal tolerance in that PDL1 blockade or deficiency resulted in decreased allogeneic fetal survival rates. CD4(+)CD25(+) T regulatory cells (Tregs) have also been demonstrated to play an important role in fetomaternal tolerance. Since PDL1 is expressed on Tregs, we explored the interactions between PDL1 and Tregs in vivo in a mouse model of fetomaternal tolerance. Depletion of CD25(+) T cells abrogated the effect of anti-PDL1 Ab indicating that the effect of PDL1 is possibly mediated by CD25(+) Tregs. Adoptive transfer of Tregs from wild-type but not PDL1-deficient mice into PDL1-deficient recipients significantly improved fetal survival. The frequency, phenotype and placental trafficking of Tregs from PDL1-deficient mice were similar to those of wild-type controls, but were defective in inhibiting alloreactive Th1 cells in vitro. This is the first report providing evidence for a link between PDL1 and T regulatory cells in mediating fetomaternal tolerance.  相似文献

There is no clear relation between portal systemic shunting, reduced hepatic insulin extraction leading to an increased systemic delivery of insulin, and, resultant peripheral hyperinsulinemia and insulin resistance. Extrahepatic portal vein obstruction is a natural human model of portal systemic shunting with essentially normal liver function. To investigate the role of portal systemic shunting of insulin in creating systemic hyperinsulinemia and insulin resistance, we studied nine subjects with portal systemic shunting and nine controls matched for age (+/- 2 years), body weight (+/- 2 kg) and height (+/- 5 cm). We carried out an oral glucose tolerance test and hyperinsulinemic euglycemic clamp study at insulin infusion rate of 40 mU/m2/ min. Comparable (p = 0.61) basal insulin concentrations in the two groups (Mean (SE): 21.0 (3.98) vs. 24.1 (4.28) mU/L) demonstrated a lack of hyperinsulinemia in the presence of portal systemic shunting. The lower (p = 0.03) insulin area under curve on oral glucose tolerance test in presence of portal systemic shunting (7.40 (0.95) vs. 10.83 (1.15) U/L-min) indicated that lower extraction of insulin by the liver leads to a lower requirements in the periphery. The coefficient of variation for plasma glucose between 60 and 120 min of the clamps was 4.44 (0.55)%. Comparable (p = 0.82) M-values (6.21 (0.67) vs. 6.38 (0.45) mg/kg/min) in the two groups proved a lack of significant insulin resistance in the presence of portal systemic shunting. We conclude that isolated portal systemic shunting leads to neither hyperinsulinemia nor insulin resistance.  相似文献

The essential oil from fresh leaves of Thuja orientalis L. grown in the north-western Himalaya was isolated by means of hydrodistillation and analyzed by GC and GC/MS. Twenty-two compounds representing 94.0% of the total oil were identified. The leaf oil contained alpha-pinene (29.2%), Delta-3-carene (20.1%), alpha-cedrol (9.8%), caryophyllene (7.5%), alpha-humulene (5.6%), limonene (5.4%), alpha-terpinolene (3.8%) and alpha-terpinyl acetate (3.5%) as major constituents. The essential oil showed antifungal activity against Alternaria alternata in a direct bioautography assay. Two main bioactive compounds named as b1 (Rf = 0.54) and b2 (Rf = 0.80) were observed and tested for antifungal activity; they produced an inhibition zone of 5 and 10 mm in diameter, respectively. The components b1 and b2 were further purified by preparative thin layer chromatography and their antifungal efficacy was re-tested. The minimum inhibitory amount (MIA) of b1 and b2 against A. alternata was determined as 30.5 and 4.5 microg, respectively, using a bioautography assay. The bioactive constituent corresponding to b1 was determined as alpha-cedrol by using GC/MS analysis. The potential of essential oils as a source of natural biocides is discussed.  相似文献

The synthesis and cytotoxic studies of a new series of 16E-arylidene androstene derivatives are reported herein. The impact of incorporating bis-tertiary amino functionalities in the steroid skeleton on cytotoxicity has also been observed. The compounds have been evaluated at National cancer Institute, Bethesda, Maryland, USA for their antineoplastic activity against various tumor cell lines. The synthesized 16E-arylidenosteroids exhibited significant cytotoxicity. Bis-tertiary amino steroid 29 possessing a diethylaminoalkoxy functionality was the most promising compound of the series with a total IP and SC score of 20 in in vivo hollow fiber assay and was selected for further detailed in vivo xenograft testing.  相似文献
