Transforming growth factor β1 (TGF-β1) is a multifunctional cytokine whose potent immunomodulatory activity is well documented. To explore the mechanisms of this activity we examined the effect of TGF-β1 on the production of IFN-γ measured at the mRNA and protein levels in the YAC-1 cell lymphoma. In previous studies, this model proved useful to characterize the mode of action of the immunosuppressant rapamycin (RAP). Here, we found that when induced by IL-1 or IL-1 + PMA, the production of IFN-γ is suppressed by both TGF-β1 (ED50 = 1.9 pM) and RAP (ED50 = 0.2 nM). In contrast, when induced by the calcium ionophore ionomycin, in the absence or in the presence of PMA, this production is enhanced up to 10-fold by TGF-β (ED50 = 1.8 pM) and 1.5—3-fold by RAP. Therefore, in YAC-1 cells, TGF-β1 exerts opposite effects on IFN-γ production depending on the mode of activiation, and these effects parallel those of RAP. To further analyze the mode of action of TGF-β1 in this system, we used okadaic acid (OA), an inhibitor of serine/threonine protein phosphatases. Treatment with OA rendered the expression of IFN-γ mRNA induced by IL-1 insensitive to TGF-β1 or RAP, indicating that activation of a phosphatase may play a role in the suppressive effect of both agents. However, OA did not prevent the augmentation of ionomycin-mediated induction of IFN-β mRNA by either TGF-β1 or RAP. Hence, the up-regulation of IFN-β production by TGF-β1 and RAP may involve a different biochemical mechanism that that mediating their suppressive action. These observations also favor the hypothesis that the two agents act on the same regulatory pathways. This was further supported by the finding that TGF-β1 and RAP modulate IFN-γ production in an additive rather than synergistic fashion. However, their effects could be dissociated in mutants of YAC-1 cells selected for resistance to the inhibition of IL-1-mediated IFN-γ induction by RAP. Moreover, the IFN-γ modulatory action of RAP in YAC-1 cells was accompanied by an antiproliferative effect, whereas TGF-β1 failed to alter the growth of these cells. Cystinuria is a hereditary disorder of amino acid transport and is manifested by the development of kidney stones. In some patients the disease is caused by mutations in the SLC3A1 gene, which is located on the short arm of chromosome 2 and encodes a renal/intestinal transporter for cystine and the dibasic amino acids. In Israel cystinuria is especially common among Jews of Libyan origin. After excluding SLC3A1 as the disease-causing gene in Libyan Jewish patients, we performed a genomewide search that shows that the Libyan Jewish cystinuria gene maps to the long arm of chromosome 19. Significant linkage was obtained for seven chromosome 19 markers. A maximal LOD score of 9.22 was obtained with the marker D19S882. Multipoint data and recombination analysis placed the gene in an 8-cM interval between the markers D19S409 and D19S208. Significant linkage disequilibrium was observed for alleles of four markers, and a specific haplotype comprising the markers D19S225, D19S208, D19S220, and D19S422 was found in 11 of 17 carrier chromosomes, versus 1 of 58 Libyan Jewish noncarrier chromosomes. 相似文献

Candidate-gene association studies in rheumatoid arthritis (RA) have lead to encouraging yet apparently inconsistent results. One explanation for the inconsistency is insufficient power to detect modest effects in the context of a low prior probability of a true effect. To overcome this limitation, we selected alleles with an increased probability of a disease association, on the basis of a review of the literature on RA and other autoimmune diseases, and tested them for association with RA susceptibility in a sample collection powered to detect modest genetic effects. We tested 17 alleles from 14 genes in 2,370 RA cases and 1,757 controls from the North American Rheumatoid Arthritis Consortium (NARAC) and the Swedish Epidemiological Investigation of Rheumatoid Arthritis (EIRA) collections. We found strong evidence of an association of PTPN22 with the development of anti-citrulline antibody-positive RA (odds ratio [OR] 1.49; P=.00002), using previously untested EIRA samples. We provide support for an association of CTLA4 (CT60 allele, OR 1.23; P=.001) and PADI4 (PADI4_94, OR 1.24; P=.001) with the development of RA, but only in the NARAC cohort. The CTLA4 association is stronger in patients with RA from both cohorts who are seropositive for anti-citrulline antibodies (P=.0006). Exploration of our data set with clinically relevant subsets of RA reveals that PTPN22 is associated with an earlier age at disease onset (P=.004) and that PTPN22 has a stronger effect in males than in females (P=.03). A meta-analysis failed to demonstrate an association of the remaining alleles with RA susceptibility, suggesting that the previously published associations may represent false-positive results. Given the strong statistical power to replicate a true-positive association in this study, our results provide support for PTPN22, CTLA4, and PADI4 as RA susceptibility genes and demonstrate novel associations with clinically relevant subsets of RA. 相似文献

Design of attachment devices in insects varies enormously inrelation to different functional loads. Many systems, locatedon different parts of the body, involve surfaces with particularfrictional properties. Such systems evolved to attach partsof the body to each other, or to attach an insect to the substratumby providing fast and reversible attachment/detachment. Amongthese systems, there are some that deal with predefined surfaces,and others, in which one surface remains unpredictable. We have purified and cloned a HeLa cell nuclear protein that strongly stimulates binding of retinoic acid and thyroid hormone receptors (RARs and TRs) to response elements. The purified protein is a human retinoid X receptor beta (hRXR beta). Three murine members of the RXR family (mRXR alpha, beta, and gamma) have also been cloned, and their interactions with RARs and TRs have been investigated. Under conditions where RAR, RXR, and TR bound poorly as homodimers to various response elements, strongly cooperative RAR-RXR and TR-RXR binding was observed. The binding efficiency was dependent on the sequence, relative orientation, and spacing of the repeated motifs of response elements. We show also that unstable RAR-RXR heterodimers were formed in solution, and that C-terminal sequences and the DNA-binding domains of both receptors were required for efficient formation of stable heterodimers on response elements. These findings suggest a convergence of the signaling pathways of some members of the nuclear receptor superfamily. 相似文献

Candida shehatae cells, cultivated on d-glucose and d-xylose, were subjected to a shift from fully aerobic to anaerobic fermentative conditions. After anaerobic conditions were
imposed, growth was limited to approximately one doubling or less as C. shehatae rapidly entered a stationary phase of growth. Following the shift to anoxia, cell viability rapidly declined and the total
cell volume declined in the d-xylose fermentations. Moreover, the cell volume distribution shifted to smaller volumes. Cell viability, measured by plate
counts, declined nine times faster for d-xylose fermentations than for d-glucose fermentations. Anaerobic growth did not occur on either d-glucose or d-xylose. Selected vitamins and amino acids did not stimulate anaerobic growth in C. shehatae, but did enhance anaerobic growth on d-glucose in S. cerevisiae. The decline in cell viability and lack of anaerobic growth by C. shehatae were attributed to oxygen deficiency and not to ethanol inhibition. The results shed light on why C. shehatae anaerobic fermentations are not currently practical and suggest that research directed towards a biochemical understanding
of why C. shehatae can not grow anaerobically will yield significant improvements in ethanol fermentations from d-xylose.

Candida shehatae were sequentially subjected to aerobic conditions for cellular growth, followed by anaerobic conditions for ethanol production
from D-xylose at pH 2.5, 4.5 and 6.0. Ethanol yields increased from 0.25 g/g to 0.37 g/g and xylitol yields decreased from 0.33 g/g
to 0.1 g/g as the pH was increased from 2.5 to 6.0. Cell viability, measured by plate counts and methylene blue staining,
decreased in all of the fermentations, following the switch from aerobic to anaerobic conditions. However, pH 6.0 was shown
to extend cell viability and increase the final ethanol concentration from 45 g/l to 55 g/l, compared to the yield at pH 4.5.

The thalamus is classically viewed as passively relaying information to the cortex. However, there is growing evidence that the thalamus actively regulates information transmission to the cortex and between cortical areas using a variety of mechanisms, including the modulation of response magnitude, firing mode, and synchrony of neurons according to behavioral demands.
