Different sexual morphs of dimorphic and trimorphic populations of plants have different demands for resources correlating with phenotypic and ecological dissimilarities among morphs. Few studies have investigated the variations in mycorrhizal symbiosis across the sexual morphs of trimorphic plant populations. Here, we report seasonal variations in the mycorrhizal colonization of the roots of female, male and hermaphroditic trees of Jacaratia mexicana under field conditions in central Mexico. Mycorrhizal colonization was low in the rainy season, when J. mexicana bears only leaves, but high in the dry season when it flowers and fruits. In the dry season, the mycorrhizal colonization of female and male trees was over twofold higher than that of the hermaphroditic trees, and all three morphs had low mycorrhizal colonization in the rainy season. The lack of seasonal variation in the mycorrhizal colonization of hermaphroditic trees, together with other available evidence, supports the idea that hermaphroditic trees of J. mexicana might have some physiological disadvantages compared with male and female trees.  相似文献

Potato aspartic proteases (StAPs) and their swaposin domain (StAsp-PSI) are proteins with cytotoxic activity which involves plasma membrane destabilization. The ability of these proteins to produce cell death varies with the cellular type. Therefore, StAPs and StAsp-PSI selective cytotoxicity could be attributed to the different membrane lipid compositions of target cells. In this work we investigate the possible mechanism by which StAPs and StAsp-PSI produce selective membrane destabilization. Results obtained from leakage assays show that StAsp-PSI is a potent inducer of the leakage of LUVs containing anionic phospholipids, especially those containing phosphatidylglycerol. Based in these results, we suggest that the cytotoxic activity of StAsp-PSI on pathogenic microorganisms could be mediated by the attraction between the exposed positive domains of StAsp-PSI and the negatively charged microorganism membrane. On the other hand, our circular dichroism spectroscopic measurements and analysis by size exclusion chromatography and followed by electrophoresis, indicate that hydrophobic environment is necessary to StAsp-PSI oligomerization and both StAsp-PSI disulfide bounds and membrane with negative charged phospholipids are required by StAsp-PSI to produce membrane destabilization and then induce cell death in tumors and microorganism cell targets. Additionally, we demonstrate that the presence of cholesterol into the LUV membranes strongly diminishes the capacity of StAsp-PSI to produce leakage. This result suggests that the lack of hemolytic and cytotoxic activities on human lymphocytes of StAsp-PSI/StAPs may be partly due by the presence of cholesterol in these cell membrane types.  相似文献

Background

Influenza vaccination strategies aim at protecting high-risk population from severe outcomes. Estimating the effectiveness of seasonal vaccines against influenza related hospitalisation is important to guide these strategies. Large sample size is needed to have precise estimate of influenza vaccine effectiveness (IVE) against severe outcomes. We assessed the feasibility of measuring seasonal IVE against hospitalisation with laboratory confirmed influenza through a network of 21 hospitals in the European Union.

Methods

We conducted a multicentre study in France (seven hospitals), Italy (one hospital), and Navarra (four hospitals) and Valencia (nine hospitals) regions in Spain. All ≥18 years hospitalised patients presenting an influenza-like illness within seven days were swabbed. Cases were patients RT-PCR positive for influenza A (H3N2); controls were patients negative for any influenza virus. Using logistic regression with study site as a fixed effect we calculated IVE adjusted for potential confounders. We restricted the analyses to those swabbed within four days.

Results

We included, 375 A(H3N2) cases and 770 controls. The overall adjusted IVE was 24.9% (95%CI–1.8;44.6). Among the target group for vaccination (N = 1058) the adjusted IVE was 28.8% (95%CI:2.8;47.9); it was respectively 36.8% (95%CI:−48.8; 73.1), 42.6% (95%CI:−16.5;71.7), 17.8%(95%CI:−40.8; 52.1) and 37.5% (95%CI:−22.8;68.2) in the age groups 18–64, 65–74, 75–84 and more than 84 years.

Discussion

Estimation of IVE based on the pooling of data obtained through a European network of hospitals was feasible. Our results suggest a low IVE against hospitalised confirmed influenza in 2011–12. The low IVE may be explained by a poor immune response in the high-risk population, imperfect match between vaccine and circulating strain or waning immunity due to a late season. Increased sample size within this network would allow more precise estimates and stratification of the IVE by time since vaccination and vaccine types or brands.  相似文献

Annonaceae aporphine alkaloids, of which liriodenine is the most abundant, have not been extensively studied from a biological standpoint. The goal of this study was to investigate the role of liriodenine in antimicrobial defense during early developmental stages in Annona diversifolia. The fungi Rhizopus stolonifer and Aspergillus glaucus, which are responsible for seed deterioration, were isolated during imbibition, and their antifungal activity was determined by diffusion, macrodilution, and metabolic inhibition assays using purified liriodenine and alkaloid extracts obtained from embryos, radicles, and roots at early developmental stages. The presence of liriodenine in extracts was quantified by high-performance liquid chromatography. Purified liriodenine and alkaloidal extracts inhibited both fungi, and there was a positive relationship between extract activity and amount of liriodenine contained therein. The quantity of liriodenine present in extracts suggests its importance in controlling other phytopathogens.  相似文献

The genus Phlebovirus of the family Bunyaviridae consists of approximately 70 named viruses, currently assigned to nine serocomplexes (species) based on antigenic similarities. Sixteen other named viruses that show little serologic relationship to the nine recognized groups are also classified as tentative species in the genus. In an effort to develop a more precise classification system for phleboviruses, we are attempting to sequence most of the named viruses in the genus with the goal of clarifying their phylogenetic relationships. In this report, we describe the serologic and phylogenetic relationships of 13 viruses that were found to be members of the Candiru serocomplex; 6 of them cause disease in humans. Analysis of full genome sequences revealed branching inconsistencies that suggest five reassortment events, all involving the M segment, and thus appear to be natural reassortants. This high rate of reassortment illustrates the inaccuracy of a classification system based solely on antigenic relationships.  相似文献

One of the difficulties in the comparison of results between laboratories working with the comet assay is the great diversity of parameters used to express DNA damage and the lack of conversion factors between the majority of them. Here we report a scorer-independent conversion curve to transform the values of DNA damage reported in arbitrary units (AU) into estimated percentage of DNA in the tail (E%T), and the results obtained in an intercomparison exercise where the effectiveness of this curve and two others proposed in the literature (E%T=AU/4 and E%T=(AU/5)+10) were tested. To obtain the conversion curve, human lymphocytes were first treated with radiation or H(2)O(2). Percentage of DNA in tail (%T) was then measured in 2100 comets (300 comets per treatment) using Casp image analysis software. Subsequently, using these values of %T, categories of 0, 1, 2, 3, and 4 were assigned to comets with %T [0-1), [1-25), [25-45), [45-70), and >70, and DNA damage was calculated in AU, as usual. DNA damage was induced in the interval 24-315AU (1.54-65.23%T). The best-fit conversion curve obtained by regression analysis was E%T=(AU-25.87)/4.46. In the intercomparison exercise, ten scorers from nine laboratories analyzed the same comet images (recorded on a compact disc) visually. The values reported in comet categories were transformed into AU and subsequently into E%T, using the three approaches mentioned above. The best agreement between E%T and %T measured by the software (S%T) was obtained with the conversion curve reported here, where the slope of E%T versus S%T from the ten scorers was not different from 1. Using this conversion curve, the overall mean difference between E%T and S%T was 1.4±2.62 and 57 (81%) of E%T values differ from S%T by less than 5 units. These findings show the strength of the scorer-independent conversion curve as a tool to compare results reported in AU or %T by different laboratories.  相似文献

Specific roles of glycosylation appear to be protein-dependent. Plant aspartic proteases (APs) contain two or more consensus N-glycosylation sites; however, the importance of them is not well understood. StAPs (Solanum tuberosum aspartic proteases) are bifunctional proteins with both proteolytic and antimicrobial activities. These proteins are accumulated into the intercellular washing fluid of potato tubers and leaves after wounding or infection. In this paper we investigated the importance of glycosylation on the StAPs apoplast accumulation, biochemical parameters, and fungicidal activity. Assays to evaluate the importance of StAPs glycosylation groups by using glycosylation inhibitors demonstrate that carbohydrate portions are essential to StAPs accumulation into the apoplast of tubers and leaves after wounding or detachment, respectively. Bifunctional activity of StAPs is differentially affected by this post-translational modification. Results obtained show that not significant changes were produced in the physicochemical properties after StAPs deglycosylation (pH and thermal-optimum activity and index of protein surface hydrophobicity). Otherwise, StAPs antifungal activity is affected by deglycosylation. Deglycosylated StAPs (dgStAPs) fungicidal activity is lower than native StAPs at all concentrations and times assayed. In summary, glycosylation has not a significant role on the StAPs conformational structure. However, it is involved in the StAPs subcellular accumulation and antifungal activity suggesting that it could be necessary for StAPs membrane and/or protein interactions and subsequently its biological function(s).  相似文献

HIV-1 viral load testing from 51 patients was compared in plasma samples simultaneously processed and stored in primary Vacutainer plasma preparation tubes (PPTs) and PPT sample aliquots transferred in secondary tubes before freezing, using RT-PCR quantification with an ultrasensitive method (the Roche AMPLICOR HIV-1 MONITOR). We concluded that freezing the primary sample in the PPT collection tube can artificially elevate the HIV-1 viral load. We therefore store samples as frozen aliquots in a second tube.  相似文献
