Zusammenfassung Die Hypothese, daß der Atlantik zwischen NW-Afrika und der Südküste der Iberischen Halbinsel auf dem Frühjahrszug von Transsaharaziehern nur bei E-Winden infolge von Drift oder Pseudodrift überquert wird, wurde geprüft. An der Fangstation von Sanlucar de Barrameda an der spanischen Südküste wurden 1983 und 1986 maximale Fangzahlen von Transsaharaziehern in Zeiten erreicht, in denen mehrere Tage hintereinander östliche Winde am Boden und in der Höhe herrschten.
Is the Atlantic between NW Africa and Spain crossed by passerine trans-Saharan migrants in spring?
Summary The hypothesis is tested whether the Atlantic between NW Africa and the south coast of the Iberian Peninsula is crossed in spring by passerine trans-Saharan migrants only in easterly winds because of drift or pseudodrift. The analysis is based on the number of daily captured birds during spring migration 1983 and 1986 at a ringing station in Sanlucar de Barrameda, situated at the Spanish south coast. Maximum numbers of captured birds occured only during periods of several days with easterly winds at the bottom and high.

The properties of one ATP-inhibited and one Ca2+-dependent K+ channel were investigated by the patch-clamp technique in the soma membrane of leech Retzius neurons in primary culture. Both channels rectify at negative potentials. The ATP-inhibited K+ channel with a mean conductance of 112 pS is reversibly blocked by ATP (K i = 100 m), TEA (K i =0.8 mm) and 10 mm Ba2+ and irreversibly blocked by 10 nm glibenclamide and 10 m tolbutamide. It is Ca2+ and voltage independent. Its open state probability (P o) decreases significantly when the pH at the cytoplasmic face of inside-out patches is altered from physiological to acid pH values. The Ca2+-dependent K+ channel with a mean conductance of 114 pS shows a bell-shaped Ca2+ dependence of P o with a maximum at pCa 7–8 at the cytoplasmic face of the membrane. The P o is voltage independent at the physiologically relevant V range. Ba2+ (10 mm) reduces the single channel amplitude by around 25% (ATP, TEA, glibenclamide, tolbutamide, and Ba2+ were applied to the cytoplasmic face of the membrane).We conclude that the ATP-dependent K+ channel may play a role in maintaining the membrane potential constant—independently from the energy state of the cell. The Ca2+-dependent K+ channel may play a role in generating the resting membrane potential of leech Retzius neurons as it shows maximum activity at the physiological intracellular Ca2+ concentration.This study was supported by the Deutsche Forschungsgemeinschaft (W.-R. Although neurofilaments are among the most highly phosphorylated proteins extant, relatively little is known about the kinases involved in their phosphorylation. The majority of the phosphates present on the two higher-molecular-mass neurofilament subunits are added to multiply repeated sequence motifs in the tail. We have examined the specificity of a neurofilament-associated kinase (NFAK) partially purified from chicken spinal cord that selectively phosphorylates the middle-molecular-mass neurofilament subunit, NF-M. Two-dimensional phosphopeptide mapping of 32P-labeled NF-M shows that, in vitro, NFAK phosphorylates a subset of peptides phosphorylated in vivo in cultured neurons. The absence of a complete complement of labeled phosphopeptides following in vitro phosphorylation, compared with phosphorylation in vivo, is not due to a lack of availability of phosphorylation sites because the same maps are obtained when enzymatically dephosphorylated NF-M is used as an in vitro substrate. Phosphopeptide maps from in vitro-phosphorylated NF-M and those from a recombinant fusion protein containing only a segment of the tail piece of chicken NF-M reveal identical labeled peptides. The fusion protein lacks a segment containing 17 KXX(S/T)P putative phosphorylation sites contained in the tail of chicken NF-M but contains a segment that includes four KSPs and a KSD site also present in the intact tail. These results suggest (a) that NFAK mediates the phosphorylation of some, but not all, potential phosphorylation sites within the tail of NF-M and (b) that multiple kinases are necessary for complete phosphorylation of the NF-M tail.  相似文献

-Chymotrypsin was modified with cyanuric chloride activated monomethoxypolyethylene glycol (MPEG) with molecular weights 1900 and 5000. Using the higher molecular weight MPEG a product that was soluble in benzene at moderate levels of modification was obtained, whereas with MPEG 1900 almost all the enzyme's amino groups had to be modified for dissolving the conjugate. The catalytic activity decreased with increasing degree of substitution. Apparent Vmax was considerably higher for the less modified enzyme preparation than for the more modified one, while Km,app stayed almost constant. The modified enzyme was used for peptide synthesis. The reaction was dependent on the content of dissolved water. Both Vmax,app and Km,app increased with increasing water content. It was possible to achieve a process with complete conversion of substrate to dipeptide.  相似文献

The signal transduction system that mediates bacterial chemotaxis allows cells to moduate their swimming behavior in response to fluctuations in chemical stimuli. Receptors at the cell surface receive information from the surroundings. Signals are then passed from the receptors to cytoplasmic chemotaxis components: CheA, CheW, CheZ, CheR, and CheB. These proteins function to regulate the level of phosphorylation of a response regulator designated CheY that interacts with the flagellar motor switch complex to control swimming behavior. The structure of CheY has been determined. Magnesium ion is essential for activity. The active site contains highly conserved Asp residues that are required for divalent metal ion binding and CheY phosphorylation. Another residue-at the active site, Lys109, is important in the phosphorylation-induced conformational change that facilitates communication with the switch complex and another chemotaxis component, CheZ. CheZ facilitates the dephosphorylation of phospho-CheY. Defects in CheY and CheZ can be suppressed by mutations in the flagellar switch complex. The macroscopic algae Characeae are usually assumed to occur in waterbodies supplied by groundwater with low phosphate content, but the indicative value of the species is seldom defined in bibliography. Former braided channels of the Rhône river are supplied with groundwater originating from the main channel (seepage) or from hillslope aquifer. The aim of the present paper was to determine if it possible to use the Characeae as indicators of physicochemical characteristies of water in order to assess the origin of groundwater supplying former river channels. Four former braided channels of the Rhône River colonized by Characeae were investigated, and the physico-chemical characteristics of i) the channels, ii) the groundwater and iii) the river were measured over a period of several months. Species are arranged along a gradient of conductivity, alkalinity, ammonium and phosphate content of the water. Charophyte species can indicate the origin of groundwater, either seepage or hillslope nutrient-poor aquifer, and integrate both the average value of the chemical parameter, and their variations. C. hispida occurs in a nutrient-poor channel mainly supplied by highly calcareous groundwater coming from hillslope aquifer. Chara major has requirements close to those of C. hispida, but is more tolerant to periodic inputs of nutrients. C. vulgaris and N. syncarpa both tolerate mesotrophic waters originating from both hillslope aquifer and seepage, and C. globularis is associated to a channel mainly supplied by mesotrophic to eutrophic river seepage.  相似文献

H2–forming N 5,N 10 methylenetetrahydromethanopterin dehydrogenase is a novel type of hydrogenase that contains neither nickel nor iron-sulfur clusters. Evidence has been presented that the reaction mechanism catalyzed by the enzyme is very similar to that of the formation of carbocations and H2 from alkanes under superacidic conditions. We present here further results in support of this mechanism. It was found that the purified enzyme per se did not catalyze the conversion of para H2 to ortho H2, a reaction catalyzed by all other hydrogenases known to date. However, it catalyzed the conversion in the presence of the substrate N 5,N 10 methenyltetrahydromethanopterin (CH≡H4MPT+), indicating that for heterolytic cleavage of H2 the enzyme-CH≡H4MPT+ complex is required. In D2O, the formation of HD and D2 from H2 rather than a paraortho H2 conversion was observed, indicating that after heterolytic cleavage of H2 the dissociation of the proton from the enzyme-substrate complex is fast relative to the re-formation of free H2.  相似文献
