Antifilaggrin autoantibodies (AFA) have described to be the most specific markers of rheumatoid arthritis (RA) and epitopes containing citrulline within the sequence of filaggrin identified as major antigenic sites recognised by AFA. In this paper we confirm that citrulline is an essential constituent of filaggrin related antigenic determinants recognised by RA specific autoantibodies, thus having an important role for the development of filaggrin peptides based serological tests. Moreover, we describe our findings on the comparative conformational analysis of filaggrin (306–324) peptide sequence and an analogue in which two arginines were simultaneously substituted by citrulline carried out by Circular Dichroism (CD) and Fourier-Transform Infrared Spectroscopy (FTIR). Results might contribute to the understanding of the structural features that may be important to explain the enhanced binding characteristics of citrullinated peptides to the autoantibodies in rheumatoid arthritis. 相似文献
Aim Fagus grandifolia var. mexicana (Martinez) Little has an extraordinarily restricted distribution in the Mexican montane cloud forests. Isolated Fagus (beech) populations have been recorded in less than 10 small areas (2–40 ha) in the eastern Sierra Madre at altitudes from 1400 to 2000 m. The objectives were to determine tree and seedling age, forest structure, phenology, litterfall patterns and the relationship between mast and climatic variables. Location We report on three Fagus stands at the Acatlan Volcano, Veracruz, Mexico. Methods Changes in forest cover were determined using aerial photographs. Within each stand, basal area, density and tree species composition were determined in a 0.1‐ha band transect. Additionally, litterfall production was quantified and phenophases were recorded monthly over a 3‐year period, and 60 tree cores were collected to determine age distribution and tree‐ring growth. Results The forest was atypical in several respects. Fagus was the only dominant tree species in the crater stand, although in the rim and at the top of the volcano it was codominant with other tree species. Juveniles occurred only on the rim, but there was a seedling bank in the crater. Although forest cover in the area increased between 1968 and 1993, the Fagus stands did not change in size. Leaf production peaked in March and April, and leaf fall occurred from October through February. Litterfall production was the highest in November. During mast years, flowering started in February and between mast events there were no flowers or fruits. Minimum temperatures were highly correlated with Fagus litterfall and leaf fall. Seedlings ranged in age from 2 to 18 years and were 13–60 cm tall. Tree cores ranged from 76 to 120 years, but trees were older than the core samples. Main conclusions Although beech is considered a gap regeneration species that reaches the canopy after alternating periods of release and suppression, the trees in the crater were released when less than 1.5 m tall and have suffered few periods of suppression since. The results indicate that the crater stand was established after a severe disturbance destroyed the existing forest. We conclude that the relict beech population should be able to maintain itself, if not severely disturbed by humans or by climatic changes related to global warming. 相似文献
Abstract We have isolated spontaneous mutant strains of Escherichia coli KL16 showing different levels of nalidixic acid (NAL) resistance. From 40 independent mutants, 36 had gyrA and four had gyrB mutations. Most of the gyrA mutations (30/36) conferred high level NAL resistance. In contrast, the only gyrB mutation that conferred a relatively high level of NAL resistance also determined enhanced susceptibility to quinolones with a piperazinyl substituent at C7 position of the quinolone ring (amphoteric quinolones). This gyrB mutation (denoted gyrB1604 ), jointly with a gyrA mutation (denoted gyrA972 ) which confers a high level of quinolone resistance, were used to construct strain IC2476, carrying the two gyr mutant alleles. The susceptibility of this strain to amphoteric quinolones (pipemidic acid, norfloxacin and ciprofloxacin) was similar to that of the gyrA972 single mutant. This result indicates that the change in GyrA subunit which determines a high level of quinolone-resistance has the capacity to mask the hypersusceptibility to amphoteric quinolones promoted by the GyrB1604 mutant subunit. This capacity was further confirmed by studying the effects of ciprofloxacin (CFX) on gyrase inhibition in the gyrA972 gyrB1604 strain. 相似文献
The application of in situ near‐infrared spectroscopy monitoring of xylose metabolizing yeast such as Pichia stipitis for ethanol production with semisynthetic media, applying chemometrics, was investigated. During the process in a bioreactor, biomass, glucose, xylose, ethanol, acetic acid, and glycerol determinations were performed by a transflection probe immersed in the culture broth and connected to a near‐infrared process analyzer. Wavelength windows in near‐infrared spectra recorded between 800 and 2200 nm were pretreated using Savitzky–Golay smoothing, second derivative and multiplicative scattering correction in order to perform a partial least squares regression and generate the calibration models. These calibration models were tested by external validation (78 samples). Calibration and validation criteria were defined and evaluated in order to generate robust and reliable models for an alcoholic fermentation process matrix. Moreover, regressions coefficients (β) and variable influence in the projection plots were used to assess the results. A novelty is the use of β versus VIP dispersion plots to determine which vectors have more influence on the response in order to improve process comprehension and operability. Background: The occurrence of shrub patches, alternating with either bare soil or low herbaceous cover, is a common feature in arid and semi-arid shrublands throughout the world. This patchy pattern of vegetation may result from water limitation, modulated by plant interactions; grazing (offtake and tramping) by livestock may cause further patchiness vegetation structure.
Aims: We hypothesised that vegetation patchiness in the semi-arid shrublands of north-eastern Patagonia would be increased by livestock grazing, but not by positive interactions between adult plants of shrubs and grasses.
Methods: We compared vegetation cover and pattern at three grazing intensities (exclosure, light and heavy grazing) and measured the growth of a representative shrub and grass in the presence and absence of the other to quantify the role of plant-to-plant interactions and its interaction with grazing for vegetation structure.
Results: In the grazing exclosure and in moderately grazed areas, vegetation cover among shrub patches was larger, whereas the top cover of shrubs was lower than in the heavily grazed areas. We did not find any evidence of positive interactions between shrub and grass life forms.
Conclusions: Our results were consistent with the hypothesis that livestock grazing increased the formation of patchy vegetation cover in arid and semi-arid shrublands. 相似文献
