The presence of a high and nonlinear Ca2+-independent (or basal) ATPase activity in rat heart preparations makes difficult the reliable measurement of sarcoplasmic reticulum (SR) Ca2+-ATPase activity by usual methods. A spectrophotometric assay for the accurate determination of SR Ca2+-ATPase activity in unfractionated homogenates from rat heart is described. The procedure is based on that reported by Simonides and van Hardeveld (1990, Anal. Biochem. 191, 321-331) for skeletal muscle homogenates. To avoid overestimation of the Ca2+-ATPase activity of cardiac homogenates that occurs when sequential measurements of total and basal ATPase activities are performed, two parallel and independent assays are required: one with low (micromolar) and other high (millimolar) calcium concentration. Addition of thapsigargin (0.2 microM) blocked totally the activity considered as Ca2+-ATPase activity. Using this method, the rat heart homogenate Ca2+-ATPase activity was 10.5 +/- 2.0 micromol. min-1 x g-1 tissue wet weight (n = 8). Likewise, a spectrophotometric assay for measuring E-type Mg2+-ATPase activity in cardiac total homogenates has been developed, comparing the following characteristics of the enzymatic activity in homogenate and a membrane-enriched fraction: first-order rate constant for ATP-dependent inactivation, Km for ATP, and effects of concanavalin A, Triton X-100, and specific inhibitors.  相似文献

Based on a computational model for 5-HT(1A)R-ligand interaction and QSAR studies, we have designed and synthesized a new series of arylpiperazines 2-8 which exhibit high 5-HT(1A)R affinity and selectivity over alpha(1)-adrenergic receptors. Among them, compound CSP-2503 (4) has been pharmacologically characterized as a 5-HT(1A)R agonist at somatodendritic and postsynaptic sites, endowed with anxiolytic properties.  相似文献

Colorimetric serum bactericidal assay (cSBA), based on the addition of glucose and a pH indicator to the culture medium after the bactericidal reaction, was validated. The precision measured as repeatability, intermediated precision, and reproducibility was determined as a percentage in titer coincidence between replicas >/=50. Moreover the use of the freeze-dried complement was evaluated in comparison to the traditionally stored by freezing. The results were the following: precision: titer +/-1 two-fold dilution (except for the highly positive serum against serogroup B, where there was titer +/-2 dilutions) percentage in titer coincidence: >/=50. The known titer or +/-1 two-fold dilution was found in the sera titrated with the complement either frozen or freeze-dried. Concluding, cSBA showed to be highly precise, allowing also the use of freeze-dried complement which is another important advantage for this kind of assay.  相似文献

This study was undertaken as a prerequisite to investigations on tooth differentiation in a squamate, the Canarian scincid Chalcides. Our main goal was to determine whether the pattern of tooth replacement, known to be regular in lizards, could be helpful to predict accurately any stage of tooth development. A growth series of 20 laboratory-reared specimens, aged from 0.5 month after birth to about 6 years, was used. The dentition (functional and replacement teeth) was studied from radiographs of jaw quadrants. The number of tooth positions, the tooth number in relation to age and to seasons, and the size of the replacement teeth were recorded. In Chalcides, a single row of pleurodont functional teeth lies at the labial margin of the dentary, premaxillary, and maxillary. Whatever the age of the specimens, 16 tooth positions were recorded, on average, in each quadrant, suggesting that positions are maintained throughout life. Replacement teeth were numerous whatever the age and season, while the number of functional teeth was subject to variation. Symmetry of tooth development was evaluated by comparing teeth two by two from the opposite side in the four jaw quadrants of several specimens. Although the relative size of some replacement teeth fitted perfectly, the symmetry criterion was not reliable to predict the developmental stage of the opposite tooth, whether the pair of teeth compared was left-right or upper-lower. The best fit was found when comparing the size of successive replacement teeth from the front to the back of the jaw. Every replacement tooth that is 40-80% of its definitive size is followed, in the next position on the arcade, by a tooth that is, on average, 20% less developed. Considering teeth in alternate positions (even and odd series), each replacement tooth was a little more developed than the previous, more anterior, one (0.5-20% when the teeth are from 10-40% of their final size). The latter pattern showed that tooth replacement occurred in alternate positions from back to front, forming more or less regular rows (i.e., "Zahnreihen"). In Chalcides, the developmental stage of a replacement tooth in a position p can be accurately predicted provided the developmental stage of the replacement tooth in position p-1 or, to a lesser degree, in position p-2 is known. This finding will be particularly helpful when starting our structural and ultrastructural studies of tooth differentiation in this lizard.  相似文献

In the present study we analyzed by PCR-SSO technique the HLA-B gene frequencies in 281 healthy individuals from four Mexican Amerindian populations (66 Mayos, 90 Mazatecans, 72 Nahuas and 53 Teenek). The most frequent alleles in all studied populations were HLA-B35, HLA-B39, and HLA-B40; however, some differences were observed between populations. The HLA-B35 allele was the most frequent in three of the four populations studied (Mayos, Nahuas and Teenek), whereas in Mazatecans the most frequent allele was HLA-B39. HLA-B40 presented frequencies higher than 10% in all groups. On the other hand, only Mayos presented an HLA-B51 gene frequency higher than 10%. When comparisons were made, important differences between groups were observed. The Teenek group presented an increased frequency of HLA-B35 when compared to Mazatecans and the HLA-B52 allele was increased in Nahuas and Teenek when compared to Mayos. An increased frequency of HLA-B39 was observed in Mazatecans when compared to Nahuas, Mayos and Teenek. Also, an increased frequency of HLA-B51 was observed in Mayos when compared to Mazatecans and Nahuas. These data corroborate the restricted polymorphism of HLA-B alleles and the high frequency of HLA-B35, HLA-B39 and HLA-B40 alleles in autochthonous American populations. In spite of the restriction in this polymorphism, differences in frequencies of HLA-B alleles could be helpful in distinguishing each of these populations.  相似文献

Sapogenins from the 25R and 25S series show a marked difference on the E/F regioselectivity of the spiroketal cleavage with BF(3)/Ac(2)O. In contrast to the high yield of single E-ring cleavage products from diosgenin (3) and hecogenin (5), sapogenins of the 25R series (equatorial C-27 methyl), sarsasapogenin (1, 25S series, axial C-27 methyl) yields the corresponding acetyldihydropyran, (25S)-23-acetyl-22,26-epoxy-5beta-cholest-22-ene-3beta,16beta-diyl diacetate (8), two isomeric furostenes: (E)- and (Z)-(25S)-23-acetyl-5beta-furost-22-ene-3beta,26-diyl diacetate (9 and 10) and a third one bearing an additional acetyl group: (E)-(20S,25S)-20,23-diacetyl-5beta-furost-22-ene-3beta, 26-diyl diacetate (11). The structures of the compounds were unambiguously established using two dimensional NMR techniques. The lower E/F selectivity in the cleavage of 1 is attributed to steric hindrance resulting from the axial methyl in F ring on a beta elimination forming the dihydropyran double bond in the major product 8.  相似文献

The wood of 11 plant species was evaluated as a food source significantly impacting the growth and survival of incipient colonies of the Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae). Colonies of C. formosanus feeding on pecan, Carya illinoensis (Wangenh.), and red gum, Liquidambar styraciflua L., produced significantly more progeny than colonies feeding on other wood species tested. Progeny of colonies feeding on pecan and American ash, Fraxinus americana L., had significantly greater survival than progeny of colonies feeding on other wood species. Colonies feeding on a nutritionally supplemented cellulose based matrix showed similar fitness characteristics as colonies feeding on the best wood treatments. These results indicate that differences observed in colony fitness can be partially explained by nutritional value of the food treatment, raising the possibility that wood from different tree species have different nutritional values to the Formosan subterranean termites. Colonies feeding on loblolly pine, Pinus taeda L., and ponderosa pine, Pinus ponderosa Laws., had significantly lower survival and produced significantly fewer workers and soldiers than colonies feeding on other wood species. Colony survival from 90 to 180 d of age and from 90 to 360 d of age was significantly correlated with the number of workers present at 90 d of colony age, indicating that colony survival depends on the presence of workers. Wood consumption in a multiple-choice study was significantly correlated with colony fitness value. This suggests that feeding preference of C. formosanus is at least partially influenced by the nutritional value of the food source.  相似文献

The cell wall-associated glyceraldehyde-3-phosphate dehydrogenase (cwGAPDH) activity in Saccharomyces cerevisiae increases (two- to 10-fold, depending on the strain) in response to starvation and temperature upshift. Assays using transformants carrying pTDH, a yeast centromer derivative plasmid containing the Candida albicans TDH3 gene (encoding GAPDH) fused in frame with the yeast SUC2-coding region for internal invertase, showed that starvation and/or temperature upshift result in a similar increase in both cwGAPDH and cell wall-associated invertase activities. In addition, this incorporation of GAPDH protein into the cell wall in response to stress does not require (i) de novo protein synthesis, indicating that preexisting cytosolic enzyme is incorporated into the cell wall, (ii) nor the participation of the ubiquitin yeast stress response system, as no differences were observed between wild-type and polyubiquitin-depleted (Deltaubi4) strains.  相似文献

Several structurally related anisidine and dibenzanthracene isomers were evaluated for genotoxic effects in the somatic w/w+ assay of Drosophila melanogaster employing insecticide-susceptible (IS) and insecticide-resistant (IR) tester strains. In addition, and in order to find whether or not at the genetic level a regulatory effect is found, crosses between ISxIR strains and IRxIS strains were done. Chemicals tested were the aromatic amines (AAs) ortho-anisidine (o-AN), meta-anisidine (m-AN), and para-anisidine (p-AN) and the polycyclic aromatic hydrocarbons (PAHs) 1,2;3,4-dibenzanthracene (1,2;3,4-DBA) and 1,2;5,6-dibenzanthracene (1,2;5,6-DBA). As positive control N-nitrosodimethylamine (DMN) was used. Our results show that the genotoxic activity of DMN was higher in the IR than in the IS strain. There seems to be a tendency for slightly lower values as measured by clone induction in crosses between ISxIR and IRxIS. o-AN was positive in the IS strain and in crosses between ISxIR and IRxIS but negative in the IR strain. m-AN, p-AN and 1,2;3,4-DBA proved to be not recombinogenic in all strains and crosses while 1,2;5,6-DBA was positive at the highest concentration tested in all the crosses assayed. These findings show that the recombinogenic activity of the anisidine isomers depends on the position of the chemical group relative to one another and that the position of the benzene ring seems to be structurally relevant for genotoxicity of DBA isomers. With respect to IR and IS strains it remains to be determined to what extent the spectrum of metabolizing capacity really differs between the strains of the test assay. Thus more information is needed about the regulation and expression of the cytochrome-P450 genes and action at the molecular level taking place in the eye imaginal disc as well as between insecticide susceptible and resistant strains after exposure to genotoxic chemicals.  相似文献

Methods of adapting micro-organisms to an inhibiting factor in an active industrial bioprocess were examined with an acetic acid fermentation as model. With the aim of automatic control, a fuzzy-logic system was developed on the basis of the collected knowledge of skilled vinegar brewers. In a first step, this fuzzy system was to assess the actual adaptation degree of the bacteria on the basis of data from robust and reasonably priced sensors. From this information an appropriate setpoint value for the inhibiting factor 'final acid concentration' was derived for each batch cycle. As a result a further acid tolerance was found after several batch cycles. This adaptation effect should be used to increase the product concentration to more than 20 g per 100 ml acetic acid with a high productivity. The stepwise adapted culture was productive over the aimed acetic acid concentration, a 10% improvement of the product formation rate could be found compared with the status before conditioning. High product concentration and increased productivity finally result in shorter cycle times, less transport and storage volumes, an improved utilization of energy and material resources, and, last but not least, they are an essential steps towards the fulfillment of economical and ecological demands.  相似文献
