The deletion of T-type Cav3.1 channels may reduce high-fat diet (HFD)-induced weight gain, which correlates positively with obesity and endothelial dysfunction. Therefore, experiments were designed to study the involvement of T-type Cav3.1 channels in HFD-induced endothelial dysfunction in mice. Wildtype (WT) and Cav3.1−/− mice were fed either a normal diet (ND) or an HFD for 8 weeks. Body composition was assessed, and thoracic aortae and mesenteric arteries were harvested for myography to assess endothelium-dependent responses. Changes in intracellular calcium were measured by fluorescence imaging, and behavior was assessed with the open-field test. Cav3.1−/− mice had attenuated HFD-induced weight gain and lower total fat mass compared with WT mice. Cav3.1−/− mice on an HFD had reduced plasma cholesterol levels compared with WT mice on the same diet. Increased feeding efficiency, independent of food intake, was observed in WT mice on an HFD compared with an ND, but no difference in feeding efficiency between diets was observed for Cav3.1−/− mice. Nitric oxide-dependent dilatation was increased in mesenteric arteries of Cav3.1−/− mice compared with WT mice on an HFD, with no difference observed in aortae. No differences in mouse locomotor activity were observed between the experimental groups. Mice on an HFD lacking T-type channels have reduced weight gain, lower total cholesterol levels, and increased dilatation of resistance vessels compared with WT mice on an HFD, suggesting that Cav3.1 deletion protects against endothelial dysfunction in resistance vessels but not in large conduit vessels.  相似文献

The crash phase of vole populations with cyclic dynamics regularly leads to vast areas of uninhabited habitats. Yet although the capacity for cyclic voles to re-colonize such empty space is likely to be large and predicted to have become evolved as a distinct life history trait, the processes of colonization and its effect on the spatio-temporal dynamics have been little studied. Here we report from an experiment with root voles (Microtus oeconomus) specifically targeted at quantifying the process of colonization of empty patches from distant source patches and its resultant effect on local vole deme size variation in a patchy landscape. Three experimental factors: habitat quality, predation risk and inter-patch distance were employed among 24 habitat patches in a 100 × 300-m experimental area. The first-born cohort in the spring efficiently colonized almost all empty patches irrespective of the degree of patch isolation and predation risk, but this was dependent on habitat quality. Just after the initial colonization wave the deme sizes in patches of the same quality were underdispersed relative to Poisson variance, indicating regulated (density-dependent) settlement. Towards the end of the breeding season local demographic processes acted to smooth out the initial post-colonization differences among source and colonization patches, and among patches of initially different quality. However, at this time demographic stochasticity had also given rise to a large (overdispersed) variation in deme sizes that may have contributed to an overshadowing of the effect of other factors. The results of this experiment confirmed our expectation that the space-filling capacity of voles is large. The costs associated with transience appeared to be so low, at least at the spatial scale considered in this experiment, that such costs are not likely to substantially constrain habitat selection and colonization in the increase phase of cyclic patchy populations.  相似文献

The novel synthetic retinoid, CD437, shows potent anti-tumor activity in a range of different cancer cell lines and now serves as a prototype for development of new retinoid related molecules (RRMs). The purpose of this study was to examine the effect and cellular targets of CD437 in the human metastatic melanoma cell lines FEMX-1 and WM239. We showed that treatment with CD437 led to cell cycle arrest and induced apoptosis through both the extrinsic- and intrinsic pathways (caspase 8, -9 and PARP cleavage) in both cell lines. Interestingly, apoptosis was induced independently of DNA-fragmentation in FEMX-1 cells, and appeared partially caspase-independent in the WM239 cells. Additionally, up-regulation of CHOP mRNA and cathepsin D protein expression, following retinoid treatment, suggests involvement of the endoplasmatic reticulum (ER) and lysosomes, respectively. Combination of suboptimal concentrations of CD437 and lexatumumab, a TRAIL death receptor-2 agonist, resulted in synergistic reduction of viable cells, along with increased PARP cleavage. These results indicate that CD437 has a strong anti-neoplastic effect alone and in combination with lexatumumab in melanoma cell lines.  相似文献

Protein kinase C (PKC) δ is a regulator of apoptosis with both pro- and anti-apoptotic effects. The mechanistic basis for the discrepant effects is not completely understood. Here we show that Smac interacts with PKCδ. The interaction depends on the N-terminus of Smac and is disrupted upon treatment with paclitaxel. This is associated with release of Smac into the cytosol. Activation of PKCδ rescues the interaction during paclitaxel exposure and suppresses the paclitaxel-mediated cell death. However, under these conditions the complex is mainly found in the cytosol suggesting that cytosolic Smac can be bound by PKCδ when PKC is activated. The data unravel a previously unrecognized interaction and suggest that PKCδ by associating with Smac may prevent its apoptotic effects.  相似文献

Mechanisms that mediate apoptosis resistance are attractive therapeutic targets for cancer. Protein kinase Cδ (PKCδ) is considered a pro-apoptotic factor in many cell types. In breast cancer, however, it has shown both pro-survival and pro-apoptotic effects. Here, we report for the first time that down-regulation of PKCδ per se leads to apoptosis of MDA-MB-231 cells. Inhibition of MEK1/2 by either PD98059 or U0126 suppressed the induction of apoptosis of PKCδ-depleted MDA-MB-231 cells but did not support survival of MCF-7 or MDA-MB-468 cells. Basal ERK1/2 phosphorylation was substantially higher in MDA-MB-231 cells than in the other cell lines. PKCδ depletion led to even higher ERK1/2 phosphorylation levels and also to lower expression levels of the ERK1/2 phosphatase MKP3. Depletion of MKP3 led to apoptosis and higher levels of ERK1/2 phosphorylation, suggesting that this may be a mechanism mediating the effect of PKCδ down-regulation. However, PKCδ silencing also induced increased MEK1/2 phosphorylation, indicating that PKCδ regulates ERK1/2 phosphorylation both upstream and downstream. Moreover, PKCδ silencing led to increased levels of the E3 ubiquitin ligase Nedd4, which is a potential regulator of MKP3, because down-regulation led to increased MKP3 levels. Our results highlight PKCδ as a potential target for therapy of breast cancers with high activity of the ERK1/2 pathway.  相似文献

We investigated rates and mechanisms of photoacclimation in cultures of Phaeocystis antarctica G. Karst. and Fragilariopsis cylindrus (Grunow) Willi Krieg, phytoplankton taxa that each dominate distinct areas of the Ross Sea, Antarctica. Both P. antarctica and F. cylindrus acclimated to increases in irradiance by reducing the effective size of the pigment antenna (σPSII) via xanthophyll‐cycle activity and reductions in chl. While enhanced photoprotection facilitated increases in specific growth rate and eventually led to higher light‐saturated photosynthetic rates (Pcellm) in P. antarctica, increases in those variables were much smaller in F. cylindrus. In response to a lower irradiance, relaxation of xanthophyll‐cycle activity led to an increase in σPSII in both taxa, which occurred much more slowly in F. cylindrus. A surprising increase in specific growth rate over the first 36 h of acclimation in P. antarctica may have facilitated the significant reductions in Pcellm observed in that taxon. In general, P. antarctica acclimated more quickly to changes in irradiance than F. cylindrus, exhibited a wider range in photosynthetic rates, but was more susceptible to photoinhibition. This acclimation strategy is consistent with growth in deeply mixed water columns with variations in irradiance that allow time for repair. In contrast, the slower acclimation rates, extensive photoprotection, and low photoinhibition exhibited by F. cylindrus suggest that it does not require the same period for repair as P. antarctica and is best suited for growth in habitats with relatively uniform irradiance, such as shallow mixed layers or sea ice.  相似文献

In the Ross Sea, the prymnesiophyte Phaeocystis antarctica G. Karst. dominates deeply mixed water columns, while diatoms dominate shallower mixed layers. Understanding what controls the dynamics of these two phytoplankton taxa is essential because they dominate virtually all coastal polar waters, have different nutrient utilization characteristics, and support dissimilar food webs. We cultured two strains of P. antarctica and one strain of the diatom Fragilariopsis cylindrus (Grunow) Willi Krieg under three dynamic irradiance regimes that simulated different mixed‐layer depths and measured their photosynthetic characteristics, cellular pigment concentrations, and cellular carbon and nitrogen content. In both species, chl a–normalized maximum carbon uptake rate (Pm* ) and specific growth rate were highest in the deeply mixed treatment that had a dark period. In all irradiance treatments, both (Pm* ) and photosynthetic efficiency (α*) were greater for the two P. antarctica strains than for the F. cylindrus strain. In contrast, P. antarctica strains were more susceptible to photoinhibition (β*) than the F. cylindrus strain. When photosynthetic rates of each phytoplankton taxon were normalized by cellular particulate organic carbon (POC), the difference in the maximal photosynthetic rate () was generally reduced. In the dynamic irradiance treatment that simulated the shallowest mixed‐layer irradiance, all three phytoplankton had similar ; however, the diatom had a 2‐fold higher POC‐normalized photosynthetic efficiency (αC). Finally, we performed calculations using the measured POC‐normalized photosynthetic parameters to show that αC and can play a greater role than βC in determining the competitive outcome between P. antarctica and F. cylindrus in both shallow and deep mixed‐layer environments of the Ross Sea.  相似文献

Preferences of sheep for different types of pen flooring   总被引:2,自引:0,他引:2  
In countries where the climate makes it practical with indoor housing during cold periods of the year and access to straw is limited, e.g. Iceland and Norway, housing of sheep on slats or expanded metal floors is common practice. However, European regulations for organic farming require that all animals should have access to a lying area with solid floor. The objective of this experiment was to investigate sheep preferences for different types of pen flooring.

In Experiment 1, a total number of 16 ewes, divided into four groups, were subjected to four different treatments. In each treatment, the ewes could choose between two lying areas with one of the following flooring materials: (1) solid wood versus rubber mats, (2) expanded metal versus solid wood, (3) solid wood versus straw, (4) expanded metal versus straw. In Experiment 2, a total number of eight ewes were individually subjected to the same treatments, first fully coated and then sheared. The ewes were video taped for 48 h in each treatment period.

In Experiment 1, the first animal that lay down after feeding preferred to lie down on straw or wood to expanded metal and straw to wooden floor (P < 0.05), but this first choice did not reflect the overall flooring preference for unsheared ewes. The groups of unsheared ewes showed no significant preferences for lying area.

In Experiment 2, single housed, unsheared ewes preferred wooden floor to rubber mats (P < 0.05), and tended to prefer expanded metal floor to straw (P = 0.08). There were no significant preferences in the two other treatments. After shearing, the ewes’ preferred wooden floor to expanded metal (P < 0.05), straw to wooden floor (P < 0.05), and straw to expanded metal floor (P< 0.0001). There were no significant preferences between rubber mats and wooden floor. Mean lying time (% of observations) for all treatments was 64.7% for unsheared ewes, and there were no significant differences between treatments. Mean lying time (% of observations) for all treatments the first 2–3 days after shearing was 43%. Significant differences in pre-shearing versus post-shearing lying times (% of observations) existed when the ewes were housed in pens with no straw (P < 0.05), this was not the case when the ewes had access to straw.

In conclusion, sheared but not unsheared ewes, preferred softer floors with low thermal conductivity (straw and wood). The less dramatically reduction in lying time (% of observations) after shearing when the ewes had access to straw, suggest that access to straw the first weeks after shearing may improve animal welfare.  相似文献
