Ca(2+) release from intracellular stores is controlled by complex interactions between multiple proteins. Triadin is a transmembrane glycoprotein of the junctional sarcoplasmic reticulum of striated muscle that interacts with both calsequestrin and the type 1 ryanodine receptor (RyR1) to communicate changes in luminal Ca(2+) to the release machinery. However, the potential impact of the triadin association with RyR1 in skeletal muscle excitation-contraction coupling remains elusive. Here we show that triadin binding to RyR1 is critically important for rapid Ca(2+) release during excitation-contraction coupling. To assess the functional impact of the triadin-RyR1 interaction, we expressed RyR1 mutants in which one or more of three negatively charged residues (D4878, D4907, and E4908) in the terminal RyR1 intraluminal loop were mutated to alanines in RyR1-null (dyspedic) myotubes. Coimmunoprecipitation revealed that triadin, but not junctin, binding to RyR1 was abolished in the triple (D4878A/D4907A/E4908A) mutant and one of the double (D4907A/E4908A) mutants, partially reduced in the D4878A/D4907A double mutant, but not affected by either individual (D4878A, D4907A, E4908A) mutations or the D4878A/E4908A double mutation. Functional studies revealed that the rate of voltage- and ligand-gated SR Ca(2+) release were reduced in proportion to the degree of interruption in triadin binding. Ryanodine binding, single channel recording, and calcium release experiments conducted on WT and triple mutant channels in the absence of triadin demonstrated that the luminal loop mutations do not directly alter RyR1 function. These findings demonstrate that junctin and triadin bind to different sites on RyR1 and that triadin plays an important role in ensuring rapid Ca(2+) release during excitation-contraction coupling in skeletal muscle. 相似文献

Migrations between different habitats are key events in the lives of many organisms. Such movements involve annually recurring travel over long distances usually triggered by seasonal changes in the environment. Often, the migration is associated with travel to or from reproduction areas to regions of growth. Young anadromous Atlantic salmon (Salmo salar) emigrate from freshwater nursery areas during spring and early summer to feed and grow in the North Atlantic Ocean. The transition from the freshwater (‘parr’) stage to the migratory stage where they descend streams and enter salt water (‘smolt’) is characterized by morphological, physiological and behavioural changes where the timing of this parr‐smolt transition is cued by photoperiod and water temperature. Environmental conditions in the freshwater habitat control the downstream migration and contribute to within‐ and among‐river variation in migratory timing. Moreover, the timing of the freshwater emigration has likely evolved to meet environmental conditions in the ocean as these affect growth and survival of the post‐smolts. Using generalized additive mixed‐effects modelling, we analysed spatio‐temporal variations in the dates of downstream smolt migration in 67 rivers throughout the North Atlantic during the last five decades and found that migrations were earlier in populations in the east than the west. After accounting for this spatial effect, the initiation of the downstream migration among rivers was positively associated with freshwater temperatures, up to about 10 °C and levelling off at higher values, and with sea‐surface temperatures. Earlier migration occurred when river discharge levels were low but increasing. On average, the initiation of the smolt seaward migration has occurred 2.5 days earlier per decade throughout the basin of the North Atlantic. This shift in phenology matches changes in air, river, and ocean temperatures, suggesting that Atlantic salmon emigration is responding to the current global climate changes. 相似文献

When examining how the ecosystems of remote islands have developed, it is important to know the timing of when various elements arrived and whether they then diversified. Our understanding of the histories behind the biodiverse south west Pacific (SWP) archipelagos is limited, and further impeded by the complex geological histories of this region. Previous studies of the SWP short-tongued halictine bee fauna suggest their presence is much younger than the geological ages of these archipelagos, which is surprising given their critical role as pollinators in other terrestrial ecosystems. The long-tongued megachilid bees represent a considerable proportion of the known bee species for the region, yet little is known of their origin. Here we use genetic diversity within mitochondrial DNA to infer the likely ages and origins of megachilid species from Vanuatu, Fiji, and Samoa. Our results indicate a very recent origin for megachilids in the SWP, with many species exhibiting small intraspecific genetic distances. Three species share almost identical haplotypes with specimens from Southeast Asia, suggesting multiple human-aided introductions. Combined with data from recent studies on other bee groups present in the region, our results have broad implications for how the Pacific island biota developed and how we should approach its management. 相似文献

The islands of the south west Pacific (SWP) are highly biodiverse, yet records of their bee fauna suggest a region depauperate of a key pollinator suite. Studies of the bees of Fiji based on molecular data have revealed a recent origin with the majority of species having arrived since the last glacial maximum or introduced since human colonization. Here we use DNA barcodes to provide the first detailed account of Apidae bees from Vanuatu, Fiji, and Samoa. We show that most if not all species in these archipelagos have been recently introduced from Australia and south east Asia, with a further species introduced from the New World. Some of these species have become regionally abundant and we discuss the potential impact of introduced pollinators on endemic plant–pollinator associations. Given the wide-reaching role of native pollinators in island systems, yet lack of understanding of SWP pollinator suites, our study highlights the urgent need for more detailed pollinator research in the region. 相似文献

In quiescent cells, mitochondria are the primary source of reactive oxygen species (ROS), which are generated by leakiness of the electron transport chain (ETC). High levels of ROS can trigger cell death, whereas lower levels drive diverse and important cellular functions. We show here by employing a newly developed mitochondrial matrix-targeted superoxide indicator, that individual mitochondria undergo spontaneous bursts of superoxide generation, termed "superoxide flashes." Superoxide flashes occur randomly in space and time, exhibit all-or-none properties, and provide a vital source of superoxide production across many different cell types. Individual flashes are triggered by transient openings of the mitochondrial permeability transition pore stimulating superoxide production by the ETC. Furthermore, we observe a flurry of superoxide flash activity during reoxygenation of cardiomyocytes after hypoxia, which is inhibited by the cardioprotective compound adenosine. We propose that superoxide flashes could serve as a valuable biomarker for a wide variety of oxidative stress-related diseases. 相似文献

The transport protein particle (TRAPP) was initially identified as a vesicle tethering factor in yeast and as a guanine nucleotide exchange factor (GEF) for Ypt1/Rab1. In mammals, structures and functions of various TRAPP complexes are beginning to be understood. We found that mammalian TRAPPII was a GEF for both Rab18 and Rab1. Inactivation of TRAPPII‐specific subunits by various methods including siRNA depletion and CRISPR–Cas9‐mediated deletion reduced lipolysis and resulted in aberrantly large lipid droplets. Recruitment of Rab18 onto lipid droplet (LD) surface was defective in TRAPPII‐deleted cells, but the localization of Rab1 on Golgi was not affected. COPI regulates LD homeostasis. We found that the previously documented interaction between TRAPPII and COPI was also required for the recruitment of Rab18 to the LD. We hypothesize that the interaction between COPI and TRAPPII helps bring TRAPPII onto LD surface, and TRAPPII, in turn, activates Rab18 and recruits it on the LD surface to facilitate its functions in LD homeostasis. 相似文献

A new generation of ocean colour satellites is now operational, with frequent observation of the global ocean. This paper reviews the potential to estimate marine primary production from satellite images. The procedures involved in retrieving estimates of phytoplankton biomass, as pigment concentrations, are discussed. Algorithms are applied to SeaWiFS ocean colour data to indicate seasonal variations in phytoplankton biomass in the Celtic Sea, on the continental shelf to the south west of the UK. Algorithms to estimate primary production rates from chlorophyll concentration are compared and the advantages and disadvantage discussed. The simplest algorithms utilise correlations between chlorophyll concentration and production rate and one equation is used to estimate daily primary production rates for the western English Channel and Celtic Sea; these estimates compare favourably with published values. Primary production for the central Celtic Sea in the period April to September inclusive is estimated from SeaWiFS data to be 102 gC m−2 in 1998 and 93 gC m−2 in 1999; published estimates, based on in situ incubations, are ca. 80 gC m−2. The satellite data demonstrate large variations in primary production between 1998 and 1999, with a significant increase in late summer in 1998 which did not occur in 1999. Errors are quantified for the estimation of primary production from simple algorithms based on satellite-derived chlorophyll concentration. These data show the potential to obtain better estimates of marine primary production than are possible with ship-based methods, with the ability to detect short-lived phytoplankton blooms. In addition, the potential to estimate new production from satellite data is discussed. 相似文献

Aim Our aim was to quantify climatic influences on key leaf traits and relationships at the global scale. This knowledge provides insight into how plants have adapted to different environmental pressures, and will lead to better calibration of future vegetation–climate models. Location The data set represents vegetation from 175 sites around the world. Methods For more than 2500 vascular plant species, we compiled data on leaf mass per area (LMA), leaf life span (LL), nitrogen concentration (Nmass) and photosynthetic capacity (Amass). Site climate was described with several standard indices. Correlation and regression analyses were used for quantifying relationships between single leaf traits and climate. Standardized major axis (SMA) analyses were used for assessing the effect of climate on bivariate relationships between leaf traits. Principal components analysis (PCA) was used to summarize multidimensional trait variation. Results At hotter, drier and higher irradiance sites, (1) mean LMA and leaf N per area were higher; (2) average LL was shorter at a given LMA, or the increase in LL was less for a given increase in LMA (LL–LMA relationships became less positive); and (3) Amass was lower at a given Nmass, or the increase in Amass was less for a given increase in Nmass. Considering all traits simultaneously, 18% of variation along the principal multivariate trait axis was explained by climate. Main conclusions Trait‐shifts with climate were of sufficient magnitude to have major implications for plant dry mass and nutrient economics, and represent substantial selective pressures associated with adaptation to different climatic regimes. 相似文献

The impacts of glacial cycles on the geographical distribution and size of populations have been explored for numerous terrestrial and marine taxa. However, most studies have focused on high latitudes, with only a few focused on the response of biota to the last glacial maximum (LGM) in equatorial regions. Here, we examine how population sizes of key bee fauna in the southwest Pacific archipelagos of Fiji, Vanuatu and Samoa have fluctuated over the Quaternary. We show that all three island faunas suffered massive population declines, roughly corresponding in time to the LGM, followed by rapid expansion post-LGM. Our data therefore suggest that Pleistocene climate change has had major impacts across a very broad tropical region. While other studies indicate widespread Holarctic effects of the LGM, our data suggest a much wider range of latitudes, extending to the tropics, where these climate change repercussions were important. As key pollinators, the inferred changes in these bee faunas may have been critical in the development of the diverse Pacific island flora. The magnitude of these responses indicates future climate change scenarios may have alarming consequences for Pacific island systems involving pollinator-dependent plant communities and agricultural crops. 相似文献
