Scanning electron microscopy of microcorrosion casts was used to visualize circulatory pathways in the sinusal spleen of dog. The examination of contracted versus dilated organs and variations of the volume of material injected gave an indication of flow dynamics. Minimal injections of material into contracted spleens produced filling of mainly the fastest routes for flow, whereas injections into dilated spleens primarily filled slower routes. This procedure yielded a more complete, three-dimensional picture of the arterial, intermediate, and venous pathways as a whole, and of the relative amounts of flow through different arterial routes. Evidence of flow from capillary lumina out into ellipsoid sheaths was plentiful in casts from dilated spleens, but rare in casts from contracted organs. The pattern of flow within and out of the marginal sinus has been elucidated: A circumferential filling occurs first, followed by a flow that radiates outward into the marginal zone and red pulp. Venous sinuses filled via two routes in addition to the generally accepted path from the reticular meshwork via fenestrations in sinus walls. First, many venous sinuses extending out from the marginal sinus and surrounding marginal zone originated as open-ended tubes continuous with the reticular spaces of the marginal sinus or marginal zone. Second, direct connections of arterial capillaries with venous sinuses in the red pulp were found. Evidence indicating that some mechanism is controlling the flow via these routes is discussed. The strikingly different arrangement of venous sinuses in the subcapsular region is demonstrated.  相似文献

Forest biomass represents 25–30 % of the annual biomass available in the USA for conversion into bio-based fuels, bio-based chemicals, and bioproducts in general. The USDA Forest Service Research and Development (R&D) has been focused on producing products from forest biomass since its inception in 1905, with direct combustion, solid sawn lumber, pulp and paper, ethanol as fuel, and silvichemicals all among the mission areas of product research and development. The renewed national interest in biomass conversion to fuels and chemicals is supportive of the most critical need of USDA Forest Service R&D, uses for small-diameter trees and other forest biomass that needs to be removed in the fuel mitigation–fire suppression and forest restoration work of the USDA Forest Service. This paper will summarize the recent USDA Forest Service research on direct combustion, fuel pellets, and conversion of forest biomass to ethanol, both as stand-alone biorefinery processes and as an addition to the traditional wood pulping process.  相似文献

The distribution of sedimentation velocities was determined, by a photoelectric method, for human erythrocytes at low concentrations in Ringer solution. The light absorption at 414 nm was measured, as a function of time, 10 mm below the top of the column. From the frequency distribution of cell velocities that of Rs √ρ-σ was found; Rs being the Stokes' radius, ρ the cell density and σ the density of the solution. Cell density was measured by the phthalate method and the mean Stokes' radius was found to be 2.58 μm. The size distributions showed some skewness but were in good general agreement with those measured by Celloscope counter, and with reported measurements from photomicrographs of cells in hanging drop suspensions. The skewness was much less than that encountered with electric sensing zone instruments (e.g. Celloscope) and the sedimentation method, being based on entirely different premises, provides an important check on such data. The skewness is due to a bias in the orientation of human erythrocytes during sedimentation. This bias may be a characteristic of biconcave cells; it could be absent in many species and reliable measurements of size distribution would then be obtained.  相似文献

We hypothesized that protein turnover would be substantially suppressed during estivation in the land snail, Otala lactea, as part of a wholesale move to conserve ATP in the hypometabolic state, and that decreased rates of protein synthesis and degradation would be mediated by altering the phosphorylation state of key proteins. Rates of protein translation, measured in vitro, decreased by ~80% in extracts of foot muscle and hepatopancreas after 2 days of estivation, and this reduction was associated with strong increases in the phosphorylation of ribosomal factors, eIF2α and eEF2, as well as decreased phosphorylation of 4E-BP1. Reductions in levels of markers of ribosomal biogenesis and a tissue-specific reduction in the phosphorylation state of eIF4E and eIF4GI were also evident after 14 days of estivation. Activity of the 20S proteasome decreased by 60–80% after 2 days of estivation and this decrease was mediated by protein kinase G in vitro, whereas protein phosphatase 2A activated the proteasome. Levels of protein carbonyls did not change in snail tissues during estivation whereas the expression heat shock proteins increased, suggesting that protein resistance to damage is enhanced in estivation. In conclusion, protein synthesis and degradation rates were coordinately suppressed during estivation in O. lactea and this is associated with the phosphorylation of ribosomal initiation and elongation factors and the 20S proteasome.  相似文献
