A limitation to the engineering of cellulolytic thermophiles is the availability of functional, thermostable (≥?60 °C) replicating plasmid vectors for rapid expression and testing of genes that provide improved or novel fuel molecule production pathways. A series of plasmid vectors for genetic manipulation of the cellulolytic thermophile Caldicellulosiruptor bescii has recently been extended to Clostridium thermocellum, another cellulolytic thermophile that very efficiently solubilizes plant biomass and produces ethanol. While the C. bescii pBAS2 replicon on these plasmids is thermostable, the use of homologous promoters, signal sequences and genes led to undesired integration into the bacterial chromosome, a result also observed with less thermostable replicating vectors. In an attempt to overcome undesired plasmid integration in C. thermocellum, a deletion of recA was constructed. As expected, C. thermocellum ?recA showed impaired growth in chemically defined medium and an increased susceptibility to UV damage. Interestingly, we also found that recA is required for replication of the C. bescii thermophilic plasmid pBAS2 in C. thermocellum, but it is not required for replication of plasmid pNW33N. In addition, the C. thermocellum recA mutant retained the ability to integrate homologous DNA into the C. thermocellum chromosome. These data indicate that recA can be required for replication of certain plasmids, and that a recA-independent mechanism exists for the integration of homologous DNA into the C. thermocellum chromosome. Understanding thermophilic plasmid replication is not only important for engineering of these cellulolytic thermophiles, but also for developing genetic systems in similar new potentially useful non-model organisms.  相似文献

The Pyst1 and Pyst2 mRNAs encode closely related proteins, which are novel members of a family of dual-specificity MAP kinase phosphatases typified by CL100/MKP-1. Pyst1 is expressed constitutively in human skin fibroblasts and, in contrast to other members of this family of enzymes, its mRNA is not inducible by either stress or mitogens. Furthermore, unlike the nuclear CL100 protein, Pyst1 is localized in the cytoplasm of transfected Cos-1 cells. Like CL100/ MKP-1, Pyst1 dephosphorylates and inactivates MAP kinase in vitro and in vivo. In addition, Pyst1 is able to form a physical complex with endogenous MAP kinase in Cos-1 cells. However, unlike CL100, Pyst1 displays very low activity towards the stress-activated protein kinases (SAPKs) or RK/p38 in vitro, indicating that these kinases are not physiological substrates for Pyst1. This specificity is underlined by the inability of Pyst1 to block either the stress-mediated activation of the JNK-1 SAP kinase or RK/p38 in vivo, or to inhibit nuclear signalling events mediated by the SAP kinases in response to UV radiation. Our results provide the first evidence that the members of the MAP kinase family of enzymes are differentially regulated by dual-specificity phosphatases and also indicate that the MAP kinases may be regulated by different members of this family of enzymes depending on their subcellular location.  相似文献

Unique luminal configurations exhibited by small arterial vessels in contracted spleens of dog and cat were studied by means of vascular corrosion casts examined by scanning electron microscopy. Concertina-like pleating was seen in casts of trabecular arteries/arterioles, whereas within lymphatic nodules arteriolar casts lacked pleating and were smooth and uniformly cylindrical (as were all small arterial vessels in distended spleens). Morphological details of arterial vessels observed in histological sections indicated that pleating is not due to contraction of specially arranged vascular smooth muscle but to overall shortening of trabecular arterial vessels, caused by contraction of longitudinal smooth muscle in trabeculae. Another phenomenon observed in casts from contracted spleens was an almost complete "pinching-off" of many arteriolar lumens; histological evidence indicated that this is due to contraction of vascular smooth muscle, which selectively diverts flow away from certain regions of the organ. Also noted was a markedly convoluted, tortuous configuration of arterioles (penicilli) in the red pulp of contracted spleens.  相似文献

Spleens from normal, healthy cats, dogs and rabbits were perfused with Ringer solution until only a few red cells remained. After fixation of the intact organ, small pieces of tissue were dried by a camphene method and examined under the scanning electron microscope. In all three species the red cells remaining in the spleen were either reticulocytes, spiculated cells, or cells of tear-drop shape and they were found adhering to macrophages and reticulum cells throughout the red pulp. Elongated masses were found on the sinusal surface of fenestrated endothelium (only in dog and rabbit); some of these appeared to be cells of tear-drop shape emerging from the cords into the sinus. This may perhaps denote a pitting process, as suggested by others, but it cannot be a unique function of fenestrated endothelium for red cells of similar shape were found elsewhere in the pulp. In all three species the network of reticulum fibres presents a very large contact surface area for blood cells and it seems likely that increased cell stickiness, rather than decreased deformability, leads to the trapping of immature red cells in the spleen.  相似文献

A new generation of ocean colour satellites is now operational, with frequent observation of the global ocean. This paper reviews the potential to estimate marine primary production from satellite images. The procedures involved in retrieving estimates of phytoplankton biomass, as pigment concentrations, are discussed. Algorithms are applied to SeaWiFS ocean colour data to indicate seasonal variations in phytoplankton biomass in the Celtic Sea, on the continental shelf to the south west of the UK. Algorithms to estimate primary production rates from chlorophyll concentration are compared and the advantages and disadvantage discussed. The simplest algorithms utilise correlations between chlorophyll concentration and production rate and one equation is used to estimate daily primary production rates for the western English Channel and Celtic Sea; these estimates compare favourably with published values. Primary production for the central Celtic Sea in the period April to September inclusive is estimated from SeaWiFS data to be 102 gC m−2 in 1998 and 93 gC m−2 in 1999; published estimates, based on in situ incubations, are ca. 80 gC m−2. The satellite data demonstrate large variations in primary production between 1998 and 1999, with a significant increase in late summer in 1998 which did not occur in 1999. Errors are quantified for the estimation of primary production from simple algorithms based on satellite-derived chlorophyll concentration. These data show the potential to obtain better estimates of marine primary production than are possible with ship-based methods, with the ability to detect short-lived phytoplankton blooms. In addition, the potential to estimate new production from satellite data is discussed.  相似文献

Members of the bacterial genus Caldicellulosiruptor are the most thermophilic cellulolytic microbes described with ability to digest lignocellulosic biomass without conventional pretreatment. The cellulolytic ability of different species varies dramatically and correlates with the presence of the multimodular cellulase CelA, which contains both a glycoside hydrolase family 9 endoglucanase and a glycoside hydrolase family 48 exoglucanase known to be synergistic in their activity, connected by three cellulose-binding domains via linker peptides. This architecture exploits the cellulose surface ablation driven by its general cellulase processivity as well as excavates cavities into the surface of the substrate, revealing a novel paradigm for cellulase activity. We recently reported that a deletion of celA in C. bescii had a significant effect on its ability to utilize complex biomass. To analyze the structure and function of CelA and its role in biomass deconstruction, we constructed a new expression vector for C. bescii and were able, for the first time, to express significant quantities of full-length protein in vivo in the native host. The protein, which contains a Histidine tag, was active and excreted from the cell. Expression of CelA protein with and without its signal sequence allowed comparison of protein retained intracellularly to protein transported extracellularly. Analysis of protein in culture supernatants revealed that the extracellular CelA protein is glycosylated whereas the intracellular CelA is not, suggesting that either protein transport is required for this post-translational modification or that glycosylation is required for protein export. The mechanism and role of protein glycosylation in bacteria is poorly understood and the ability to express CelA in vivo in C. bescii will allow the study of the mechanism of protein glycosylation in this thermophile. It will also allow the study of glycosylation of CelA itself and its role in the structure and function of this important enzyme in biomass deconstruction.  相似文献

OBJECTIVES: To investigate the relation between out of hours activity of general practice and accident and emergency services with deprivation and distance from accident and emergency department. DESIGN: Six month longitudinal study. SETTING: Six general practices and the sole accident and emergency department in Nottingham. SUBJECTS: 4745 out of hours contacts generated by 45,182 patients from 23 electoral wards registered with six practices. MAIN OUTCOME MEASURES: Rates of out of hours contacts for general practice and accident and emergency services calculated by electoral ward; Jarman and Townsend deprivation scores and distance from accident and emergency department of electoral wards. RESULTS: Distances of wards from accident and emergency department ranged from 0.8 to 9 km, and Jarman deprivation scores ranged from -23.4 to 51.8. Out of hours contacts varied by ward from 110 to 350 events/1000 patients/year, and 58% of this variation was explained by the Jarman score. General practice and accident and emergency rates were positively correlated (Pearson coefficient 0.50, P = 0.015). Proximity to accident and emergency department was not significantly associated with increased activity when deprivation was included in regression analysis. One practice had substantially higher out of hours activity (B coefficient 124 (95% confidence interval 67 to 181)) even when deprivation was included in regression analysis. CONCLUSIONS: A disproportionate amount of out of hours workload fell on deprived inner city practices. High general practice and high accident and emergency activity occurred in the same areas rather than one service substituting for the other.  相似文献
