Global climate change is causing increased climate extremes threatening biodiversity and altering ecosystems. Climate is comprised of many variables including air temperature, barometric pressure, solar radiation, wind, relative humidity, and precipitation that interact with each other. As movement connects various aspects of an animal''s life, understanding how climate influences movement at a fine‐temporal scale will be critical to the long‐term conservation of species impacted by climate change. The sedentary nature of non‐migratory species could increase some species risk of extirpation caused by climate change. We used Northern Bobwhite (Colinus virginianus; hereafter bobwhite) as a model to better understand the relationship between climate and the movement ecology of a non‐migratory species at a fine‐temporal scale. We collected movement data on bobwhite from across western Oklahoma during 2019–2020 and paired these data with meteorological data. We analyzed movement in three different ways (probability of movement, hourly distance moved, and sinuosity) using two calculated movement metrics: hourly movement (displacement between two consecutive fixes an hour apart) and sinuosity (a form of tortuosity that determines the amount of curvature of a random search path). We used generalized linear‐mixed models to analyze probability of movement and hourly distance moved, and used linear‐mixed models to analyze sinuosity. The interaction between air temperature and solar radiation affected probability of movement and hourly distance moved. Bobwhite movement increased as air temperature increased beyond 10°C during low solar radiation. During medium and high solar radiation, bobwhite moved farther as air temperature increased until 25–30°C when hourly distance moved plateaued. Bobwhite sinuosity increased as solar radiation increased. Our results show that specific climate variables alter the fine‐scale movement of a non‐migratory species. Understanding the link between climate and movement is important to determining how climate change may impact a species’ space use and fitness now and in the future.  相似文献

Plants are protected from pathogens not only by their own immunity but often also by colonizing commensal microbes. In Arabidopsis thaliana, a group of cryptically pathogenic Pseudomonas strains often dominates local populations. This group coexists in nature with commensal Pseudomonas strains that can blunt the deleterious effects of the pathogens in the laboratory. We have investigated the interaction between one of the Pseudomonas pathogens and 99 naturally co-occurring commensals, finding plant protection to be common among non-pathogenic Pseudomonas. While protective ability is enriched in one specific lineage, there is also a substantial variation for this trait among isolates of this lineage. These functional differences do not align with core-genome phylogenies, suggesting repeated gene inactivation or loss as causal. Using genome-wide association, we discovered that different bacterial genes are linked to plant protection in each lineage. We validated a protective role of several lineage-specific genes by gene inactivation, highlighting iron acquisition and biofilm formation as prominent mechanisms of plant protection in this Pseudomonas lineage. Collectively, our work illustrates the importance of functional redundancy in plant protective traits across an important group of commensal bacteria.Subject terms: Microbial ecology, Plant ecology  相似文献

Two sex steroid hormone combinations which have been used clinically as tests for detection of early pregnancy were examined for embryotoxic effects in macaques and baboons. Norethisterone acetate and ethinyl estradiol (NEA + EE) were orally administered to rhesus and cynomolgus monkeys and baboons at dosages ranging from one to 1,000 times the human dose equivalent (HDE) during days 20-50 of pregnancy. Progesterone and estradiol benzoate (P + EB) were delivered by two to six intramuscular injections to rhesus and cynomolgus monkeys between gestational days 20 and 35 at 0.1-25 X HDE. Fetuses were examined following cesarean section at 100 +/- 2 days (NEA + EE) or at term (P + EB). The results showed increased embryolethality over controls at 100-1,000 X HDE (NEA + EE) and at 10 and 25 X HDE (P + EB). Besides growth retardation, isolated cases of minor nongenital malformations were observed only in cynomolgus monkeys following treatment with both hormone combinations mainly at embryolethal dose levels and were considered spontaneous in nature. Virilization of female cynomolgus fetuses following NEA + EE treatment was manifested as two cases of clitoral enlargement in the 300 X HDE group and two cases of increased anogenital distance with reduced vaginal opening in the 1,000 X HDE group. The highest dose of NEA + EE was also maternally toxic, as two maternal deaths occurred at the end of the treatment period. One dead female cynomolgus fetus exposed to P + EB (10 X HDE) also exhibited masculinized external genitalia.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Physico-chemical factors that determine tracer substance flux from or into sealed vesicular structures are examined. Flux amplitudes are dependent on the average volume of a vesicle, while flux rates depend on the average number of transmembrane channels per vesicle. Gating processes leading to channel opening and/or closing affect both amplitudes and rates. Averaging over inhomogeneities in vesicle size and channel density leads to an explicit expression for time-dependent tracer content. Means for experimentally determining all variable factors in this expression are discussed.  相似文献

Protein powders that are dehydrated or mixed with a glassy compound are known to have improved thermal stability. We present elastic and quasielastic neutron scattering measurements of the global dynamics of lysozyme and ribonuclease A powders. In the absence of solvation water, both protein powders exhibit largely harmonic motions on the timescale of the measurements. Upon partial hydration, quasielastic scattering indicative of relaxational processes appears at sufficiently high temperature. When the scattering spectrum are analyzed with the Kohlrausch-Williams-Watts formalism, the exponent beta decreases with increasing temperature, suggesting that multiple relaxation modes are emerging. When lysozyme was mixed with glycerol, its beta values were higher than the hydrated sample at comparable temperatures, reflecting the viscosity and stabilizing effects of glycerol.  相似文献

Evidence is accumulating that gastrin precursors may act as growth factors for the colonic mucosa in vivo. The aims of this study were to prepare recombinant human progastrin(6-80) and to investigate its structure and biological activities in vitro. Human progastrin(6-80) was expressed in Escherichia coli as a glutathione S-transferase fusion protein. After thrombin cleavage progastrin(6-80) was purified by reverse phase high pressure liquid chromatography and characterized by radioimmunoassay, amino acid sequencing, and mass spectrometry. Assays for metal ions by atomic emission spectroscopy revealed the presence of a single tightly bound calcium ion. Progastrin(6-80) at concentrations in the pm to nm range stimulated proliferation of the conditionally transformed mouse colon cell line YAMC. The observations that progastrin(6-80) did not bind to either the cholecystokinin (CCK)-A or the gastrin/CCK-B receptor expressed in COS cells and that antagonists selective for either receptor did not reverse the proliferative effects of progastrin(6-80) suggested that progastrin(6-80) stimulated proliferation independently of either the CCK-A or the gastrin/CCK-B receptor. We conclude that recombinant human progastrin(6-80) is biologically active and contains a single calcium ion. With the exception of the well known zinc-dependent polymerization of insulin and proinsulin, this is the first report of selective, high affinity binding of metal ions to a prohormone.  相似文献

The importance of influenza viruses as worldwide pathogens in humans, domestic animals, and poultry is well recognized. Discerning how influenza viruses interact with the host at a cellular level is crucial for a better understanding of viral pathogenesis. Influenza viruses induce apoptosis through mechanisms involving the interplay of cellular and viral factors that may depend on the cell type. However, it is unclear which viral genes induce apoptosis. In these studies, we show that the expression of the nonstructural (NS) gene of influenza A virus is sufficient to induce apoptosis in MDCK and HeLa cells. Further studies showed that the multimerization domain of the NS1 protein but not the effector domain is required for apoptosis. However, this mutation is not sufficient to inhibit apoptosis using whole virus.  相似文献

Viral protein 40 (VP40) of Ebola virus appears equivalent to matrix proteins of other viruses, yet little is known about its role in the viral life cycle. To elucidate the functions of VP40, we investigated its ability to induce the formation of membrane-bound particles when it was expressed apart from other viral proteins. We found that VP40 is indeed able to induce particle formation when it is expressed in mammalian cells, and this process appeared to rely on a conserved N-terminal PPXY motif, as mutation or loss of this motif resulted in markedly reduced particle formation. These findings demonstrate that VP40 alone possesses the information necessary to induce particle formation, and this process most likely requires cellular WW domain-containing proteins that interact with the PPXY motif of VP40. The ability of VP40 to bind cellular membranes was also studied. Flotation gradient analysis indicated that VP40 binds to membranes in a hydrophobic manner, as NaCl at 1 M did not release the protein from the lipid bilayer. Triton X-114 phase-partitioning analysis suggested that VP40 possesses only minor features of an integral membrane protein. We confirmed previous findings that truncation of the 50 C-terminal amino acids of VP40 results in decreased association with cellular membranes and demonstrated that this deletion disrupts hydrophobic interactions of VP40 with the lipid bilayer, as well as abolishing particle formation. Truncation of the 150 C-terminal amino acids or 100 N-terminal amino acids of VP40 enhanced the protein's hydrophobic association with cellular membranes. These data suggest that VP40 binds the lipid bilayer in an efficient yet structurally complex fashion.  相似文献

The heavy metal tolerant Cardaminopsis halleri, grown on Zn and Cu polluted soil, showed electron dense metal containing precipitates (Zn, Cu, Sn, Fe, Al) on the leaf surface, in the intercellular spaces (Zn, Cu, Sn), the cell walls and the cell wall thickenings of the xylem vessels (Zn, traces of Cu and Fe). Large amounts of Zn were measured in the vacuoles, the main storage compartment for this metal in Cardarminopsis. The cytoplasm and nuclei contained small precipitates, including mainly Zn and Si. As shown by ESI Zn was co-localized with Si in these structures. The EEL-spectra of the cytoplasmic precipitates corresponded with the spectra of Zn-silicate. Besides Zn-silicate, electron translucent structures in the cytoplasm were identified as SiO2 by their EEL spectra. It was concluded that in the cytoplasm of Cardaminopsis Zn is transiently accumulated as silicate, being slowly degraded to SiO2. Zn is translocated into the vacuole and accumulated in an unknown form. A second Si and Zn-uptake mechanism was found, excluding a membrane and cytoplasm passage. Pinocytotic vesicles, formed by the plasmamembrane and the tonoplast, enable a direct translocation of Si and Zn from extracellular compartments into the vacuole. The formation of Zn-silicate is part of the heavy metal tolerance mechanism and may be responsible for the amelioration of the Zn toxicity in Cardaminopsis.  相似文献
