The effect of anti-idiotype antibodies (a-IdpI) on primary immune response to ovalbumin (OVA) was studied. A-IdpI against OVA antibodies were obtained with pI 6.2-6.7 from BALB/c mice. About 30-40% of IgG plaque cell formation (PCF) was inhibited if a-IdpI antiserum was added in situ, which served as a test for the evaluation of idiotype-positive (Id+) PCF. Up to 70% of common PCF and 70-80% of Id+ PCF were suppressed in mice that were injected with a-IdpI antibodies prior to immunization. This suppression was antigen- and allo-specific and depended upon the time of a-IdpI injection. If a-IdpI antibodies were disaggregated (DA) the Id+ suppression increased. A-IdpI antibodies also decreased IgE response to OVA, the suppression being most pronounced with the use of DA samples. 相似文献

Chronic heart failure (CHF) is a common complication of patients with heart dysfunction and of those suffering from various chronic illnesses. Although recently developed therapies revolutionized treatment of CHF, life expectancy of the survivors is still significantly reduced. Proposed neurohoromonal approach in CHF treatment is on the whole rather unsatisfactory. Recently the interest of clinical investigators has been focused on immune mechanisms in the pathogenesis of CHF. In this review we tried to summarize data concerning contribution of inflammatory cytokines to pathogenesis of CHF. The source and site of action of pro- and antiinflammatory cytokines and number of interactions among cytokines and neuroendocrine systems under CHF are considered. The N. Blokhin National Cancer Research Center is one of the few Russian scientific institutions in which hybridoma technology of monoclonal antibody (mAb) production has been successfully established. Using this technology, several dozens of mAbs to various antigens of human leukocytes have been elaborated. These mAbs are widely used for immune status evaluation and for differential diagnostics of leukemias. Two mAbs were used to develop therapeutic drugs. Imuteran is a pharmaceutical form of mAb ICO-25 against a mucin-like antigen of human milk fat globules and proposed for treatment of epithelial cell-originating cancers (breast, intestinal, ovarian, lung cancer, etc.). ThePhase II clinical study of this agent is now nearly completed, and preliminary results suggest Imuteran to be a promising anticancer agent with tumor-stabilizing activity, but patients should be carefully monitored for signs of allergic reactions. mAb ICO-90 against the CD3 antigen of human T lymphocytes was used to develop the therapeutic agent Atemonate proposed for treatment of acute transplant rejection. At present, the Phase II clinical study of this agent is over, and the results confirm the drug safety and efficacy for this indication. The drug is being registered at the Ministry of Healthcare and Social Development, and transfer to serial production is expected shortly. 相似文献

In studies of currents in perfused cells or their membrane fragments containing potential-dependent Ca channels and Ca2+-activated channels, a buffer slowly binding Ca2+ appears useful in some cases. A buffer with EDTA in excess of Mg2+ has been proposed, and its kinetic characteristics have been calculated. It has been shown that this buffer, depending on its component proportions, may provide Ca2+ binding with a characteristic time of up to tens of milliseconds.
