Antiviral activity of a two-spiral RNA (ts RNA), a new natural interferon inductor was studied. It was shown that ts RNA extracted from a phage infected E. coli culture was an active inductor of interferon and resistance to infection with the forestspring encephalitis virus experimental animals. In experiments on 10-12 g mice ts RNA administered in a dose of 50 micrograms/mouse 6 hours after the infection induced up to 1280 units/ml of the serum interferon. When the inductor was administered repeatedly, the experimental animals developed hyporeactivity resulting in a marked decrease in interferon production after the 3rd subsequent injection. The most pronounced effect with respect to the forest-spring encephalitis virus was observed when the inductor was administered intraperitoneally in a dose of 20 micrograms/mouse 4 hours before the infection. The protective effect was less pronounced when the inductor was administered 24 and 48 hours before the infection. A two-fold administration of the inductor did not increase the antiviral effect. When the inductor was administered in a dose of 100 micrograms 14 days before the infection, the animals showed an increase in the nonspecific resistance to the infection resulting in a marked antiviral effect. 相似文献

High-titer antidiphtheria antitoxic rabbit serum has been obtained, and on the basis of this serum a coagglutinating diagnosticum has been developed. The sensitivity of the test has been found to depend on the content of antitoxic antibodies in the serum and on its purity. Diagnostica prepared from native serum containing 500 I. U./ml (a titer of 1:51, 200 in the passive hemagglutination test) permit the detection of 0.02-0.03 Lf/ml of diphtheria toxin. A decrease in antibody titer to 5-25 I. U./ml leads to a drop in sensitivity to 0.2-2 Lf/ml. The use of LgG fraction and pure antibodies increases the sensitivity of the test to 0.002-0.003 Lf/ml. The possibility of detecting toxin in Corynebacterium diphtheriae strains is shown. 相似文献

The intravascular (iv) or intramuscular (im) inoculations of a suspension of Lewis carcinoma cells induced metastatic tumor nodules in the lungs of C57Bl/6 mice. The administration of curantil (dipyridamole) (500 mg per 20 g-mouse, per os) once a week beginning one day after iv tumor cells inoculation (2 X 10(5) cells) reduced 4-fold the number of tumor nodules. The administration of curantil one day after im tumor cells inoculation (2 X 10(6) cells) reduced 2-fold the number of tumor nodules. The Quin fluorescence in gamma-hexachlorocyclohexane-stimulated polymorphonuclear leukocytes is rapidly increased, which points to the increase in Ca2+in concentration during leukotriene B4 synthesis in leukocytes. An addition of EGTA and calcium antagonists (nifedipine, verapamil, diltiazem) to cell suspensions does not affect the basal level of internal Ca2+ but results in the inhibition of the gamma-hexachlorocyclohexane-induced Ca2+ increase. Two mechanisms of calcium homeostasis regulation in neutrophils are proposed. One of them, cAMP regulation, is coupled with a potent inhibiting effect of prostacyclin, an adenylate cyclase activator, on Ca2+in increase in stimulated neutrophils. The other one is the activation of protein kinase C catalyzed by 4 beta-phorbol-12 beta-myristate-13 alpha-acetate. The experimental results suggest that such an activation blocks Ca2+ influx into the cells via the closure of Ca2+ channels. The synergism of action of the above mechanisms in the regulation of calcium homeostasis in neutrophils is demonstrated. 相似文献

The spatial summation in receptive fields (RF) of single neurons in cat's extrastriate area 21a was investigated as a basic neurophysiological substrate for central integration processing of visual information. The results showed that the majority of investigated neurons changed their response patterns with gradual increase of applied stimulus size. In approximately 82% of cases the suppression of neuron discharges was observed when the length of the moving strip exceeded that of the RF. In some neurons the increased size of the moving stimulus leads to the changes in the RF substructure. Receptive fields of neurons recorded at the same microelectrode penetration depth showed a great variety of RF superpositions distributed in a spatially asymmetric manner. As a result, every single RF consists of multiple sub-regions within the RF, differing from each other by the number of superimposed RF-s (density factor). We suggest that such complex spatial organization of the RF provides the neurophysiological basis for central integration processing of the visual information. 相似文献

Functional relationship between wave form of complex spike (CS) and depression time of simple spike (SS) in discharge of cerebellar
Purkinje cells was studied after their activation with afferent climbing fiber at different terms of postnatal ontogenesis
in norm and after treatment with harmaline. The experiments were carried out on three age groups of Wistar rats: rat pups
(2 weeks), the adult (4–6 months), and the old animals (22–26 months). It was established that the SS duration in norm was
approximately equal in rat pups, adult, and old animals, whereas it markedly decreased form the young to the old animals during
the SS depression in the Purkinje cell discharge. Frequency of small action potential (lAP) and their number in the Purkinje
cell discharge were approximately equal in young rat pups and adult animals, while in old animals these parameters were higher,
on average, by 30%. After administration of harmaline, all CS parameters in rat pups and old animals increased in parallel
with the depression time elongation. In adult rats, harmaline did not produce statistically significant changes of the mean
values of CS parameters, but an increase of the simple spike depression time was observed. The obtained results allow concluding
that the SS wave form and the simple spike depression time in norm are functionally coupled and change with age. The effect
of harmaline on the CS wave forms as well as on interrelation of the CS duration and the CS depression time in the Purkinje
cell discharge was more pronounced at the early and the late stages of Wistar rat postnatal ontogenesis. 相似文献
