We examined the spatial pattern of an introduced population of Norway maple (Acer platanoides L.) on a temperate forested island in order to quantify the influence of landscape context on invasion pattern. The spatial location of every Norway maple tree and sapling (≥0.5 m tall) that had invaded the island forest (n = 4496) was mapped using a global positioning system. The influence of landscape context was examined with the aid of a geographic information system and indices of spatial association. We found that the coniferous forest type was the most heavily invaded (71.9% of all Norway maple stems) when compared to either the hardwood or mixed conifer–hardwood forest types (5.4% and 19.3%, respectively). Across all forest types (excluding urban trees), the population was highly aggregated around roads and other Norway maple trees. For example, 90% of the population was within 40.8 m of a road with an average distance from road of 21.02 ± 0.40 m. This association around roads was significantly greater than would be predicted by chance alone (P < 0.001). Similarly, nearest neighbor distances averaged 4.5 ± 0.2 m with 90% of individuals within 8.3 m of another Norway maple. Measures of spatial association indicated that the invasion was significantly aggregated at both the stand and island scale. Nevertheless, a comparatively small but potentially influential set of individuals were observed at relatively long distances from the main invasion front. Ramifications of these disjunct establishments and other observed patterns are discussed in the context of current spread pattern theory, invasive species monitoring, and control efforts.  相似文献

Proteolytic degradation of the major cartilage macromolecules, aggrecan and type II collagen, is a key pathological event in osteoarthritis (OA). ADAMTS-4 and ADAMTS-5, the primary aggrecanases capable of cartilage aggrecan cleavage, are synthesized as latent enzymes and require prodomain removal for activity. The N-termini of the mature proteases suggest that activation involves a proprotein convertase, but the specific family member responsible for aggrecanase activation in cartilage in situ has not been identified. Here we describe purification of a proprotein convertase activity from human OA cartilage. Through biochemical characterization and the use of siRNA, PACE4 was identified as a proprotein convertase responsible for activation of aggrecanases in osteoarthritic and cytokine-stimulated cartilage. Posttranslational activation of ADAMTS-4 and ADAMTS-5 was observed in the extracellular milieu of cartilage, resulting in aggrecan degradation. These findings suggest that PACE4 represents a novel target for the development of OA therapeutics.  相似文献

5-epi-Aristolochene dihydroxylase (EAH) catalyzes unique stereo- and regiospecific hydroxylations of a bicyclic sesquiterpene hydrocarbon to generate capsidiol. To define functional and mechanistic features of the EAH enzyme, the utility of a coupled assay using readily available sesquiterpene synthases and microsomes from yeast overexpressing the EAH enzyme was determined. Capsidiol and deoxycapsidiol biosyntheses were readily measured in coupled assays consisting of 5-epi-aristolochene synthase and EAH as determined by the incorporation of radiolabeled farnesyl diphosphate into thin-layer chromatography-isolated products and verified by gas chromatography-mass spectrometry analysis. The assays were dependent on the amounts of synthase and hydroxylase protein added, the incubation times, and the presence of nicotinamide adenine dinucleotide phosphate. The utility of this coupled assay was extended by examining the relative efficiency of the EAH enzyme to catalyze hydroxylations of different sesquiterpene skeletons generated by other terpene synthases.  相似文献

The role of natural killer cells in experimental murine salmonellosis   总被引:1,自引:0,他引:1  
This study was designed to determine if murine natural killer (NK) cells play a role in host protection against a Salmonella typhimurium challenge infection. Outbred ICR mice injected intravenously with either attenuated (RIA strain) or virulent (SR-11 strain) salmonellae elicited enhanced killing of YAC-1 targets, which was maximal at 24 h after challenging. When NK cells were depleted with antiasialo GM1 prior to challenging, the splenic bacterial numbers were significantly less in this group of mice compared to sham-injected and challenged animals. The rabbit antiasialo GM1 sera had no detectable direct or indirect effect on the salmonellae. Our results indicate that the NK or natural suppressor cells may be functioning as down-regulators.  相似文献

The importance of sympathetically mediated coronary vasoconstrictor tone as a determinant of resting coronary blood flow was assessed in the conscious dog by comparing blood flow and oxygen extraction in a normally innervated (I) and a previously sympathectomized (Sx) region of the same left ventricle. The regional ventricular sympathectomy was achieved by the topical application of phenol. The animals were well acclimated to the laboratory environment before regional myocardial blood flow was measured with microspheres or regional myocardial oxygen extraction was determined on blood sampled from chronically implanted coronary venous catheters. Results indicated that blood flow and oxygen extraction were not significantly different in I and Sx regions under these conditions. Regional blood flow data obtained after beta-adrenergic blockade or combined alpha- and beta-adrenergic blockade were not significantly different from control data. Thus we were unable to confirm previous evidence in the literature of significant resting sympathetic coronary vasoconstrictor tone in the conscious animal.  相似文献

We have studied chromosomal aberration production in V-79 Chinese hamster tissue culture cells by UV light administered during the post-DNA-synthetic G2 phase of the cell cycle. The treatment produced achromatic lesions and some chromatid deletions in the first post-irradiation mitosis, but no isochromatid deletions or chromatid exchange aberrations. In contrast, when G2 UV-irradiated cells were examined in their second post-irradiation mitosis, there were significant yields of chromatid-type aberrations of all types, including isochromatid deletions and chromatid exchanges.

We have earlier reported21 that UV-irradiation during the pre-DNA-synthetic G1 phase of the cell cycle induces only chromatid aberrations and also that most chromosomal aberration production by UV in G1 can be photoreactivated in cells possessing the photoreactivating enzyme. We present here a model for chromosomal aberration production by UV. In the model all aberration production is enzymatically mediated, a consequence of the functioning of known molecular repair mechanisms. The important elements in the model are the following:

1. (1) The vertebrate chromosome is mononeme; i.e., contains but a single DNA double helix during the prereplication G1 phase of the cell cycle.

2. (2) The UV-induced DNA lesion leading to the production of most aberrations is the cyclobutane dimer between adjacent pyrimidines in one polynucleotide strand.

3. (3) Single chain breaks appear at metaphase as achromatic lesions.

4. (4) Dimer removal sometimes leaves unrepaired single chain gaps, possibly as a result of incomplete excision repair.

5. (5) The single-stranded DNA opposite a single chain gap can be cleaved by a single-strand DNAase.

6. (6) Gaps are left in newly synthesized DNA polynucleotide chains opposite defective template chains (i.e., opposite dimers and chain breaks).

7. (7) Double-strand breaks present following local DNA replication may “spread” to the other chromatid by a recombinational process between template and new polynucleotide chains, one from each of the homologous double helices.

The model predicts the occurrence of isoachromatic lesions and of chromatid deletions paired (isolocus) with achromatic lesions. Though often not reported, both do, in fact, occur. In addition, the model accounts for the phenomenon of sister-chromatid exchange as a manifestation of a recombinational, or post-replication, repair mechanism. Finally, the model offers a simple interpretation of chromosomal aberration production by a variety of chemical agents.  相似文献

Anesthetized rabbits were subjected to two 4-min periods of asphyxia with a 15-min recovery period intervening. During the recovery period animals received an iv infusion of either isotonic glucose or mannitol. Results indicated that glucose had a beneficial effect on systolic blood pressure during the second period of asphyxia. A direct metabolic action of glucose on the myocardium is favored as the explanation, but other possibilities include peripheral or neurohumoral effects.  相似文献

The mechanism of binding of murine IFN-gamma to its receptor has not been determined. We have studied this mechanism by examining the binding of overlapping synthetic peptides of IFN-gamma to cloned soluble murine IFN-gamma R. IFN-gamma (1-39) and IFN-gamma (95-133) were able to compete with [125I]IFN-gamma for binding to cloned soluble receptor. Peptides corresponding to the inner region of IFN-gamma--IFN-gamma (36-60), IFN-gamma (54-91), and IFN-gamma (78-107)--showed a markedly reduced ability to compete with [125I]IFN-gamma for receptor binding relative to the N-terminal and C-terminal peptides. In direct binding studies, the binding of [125I]-IFN-gamma (1-39) to soluble receptor could only be competed by IFN-gamma (1-39) and IFN-gamma and not by any of the other peptides including IFN-gamma (95-133). This suggests that the N- and C-termini of IFN-gamma bind to different regions of the receptor. These data in conjunction with previous structure/function studies and x-ray crystallographic data have allowed us to formulate a "velcro-key" model of IFN-gamma binding to receptor that involves both the N- and C-terminal domains. The N-terminus binds in the classical "lock-and-key" manner characterized by specific ligand-receptor binding. The hydrophilic C-terminus binds to a region of the receptor distinct from the N-terminus likely through the polycationic region, which is conserved across species barriers. Binding of this type would exhibit high affinity and low specificity similar to a piece of velcro. This interaction becomes specific when the C-terminus is in the context of the whole IFN-gamma molecule and may act to increase the affinity of receptor binding and/or facilitate signal transduction.  相似文献

Mapping the long arm of Chromosome (Chr) 4 has assumed medical relevance with the establishment of linkage of facioscapulohumeral muscular dystrophy (FSHD) to distal 4q markers. We have constructed a multipoint linkage map using DNA markers that map to the long arm of Chr 4. Segregation data were collected for 17 DNA markers on the multigenerational CEPH mapping families, and data for one marker were taken from the published CEPH database. Genotypic information for six of these markers was also collected from a set of 24 families that exhibited inheritance of FSHD. Multipoint analyses allowed us to construct a map of 12 loci, connecting two previously separate linkage groups. Significant sexspecific differences in recombination were found for some genetic intervals. Four loci from the distal region of this map showed linkage with FSHD. A map using these terminal markers gave the strongest support for FSHD in the most distal position over all other possible positions. Offprint requests to: B. Weiffenbach  相似文献

Measurements of protein and amino acid metabolism in man using stable isotopes and selected ion monitoring gas chromatographic/mass spectrometric techniques are limited by the requirement of relatively high levels of labelling for adequate precision (greater than 0.05 at % excess). We describe here a means of extending the scope of such studies by measurement of lower levels of enrichment achieved in gaseous CO2 derived from whole blood or protein-bound amino acids following the administration of tracer amounts of appropriately labelled substrates. Construction and operation of a novel glass vacuum line required for this work are described in detail and specific applications relevant to clinical investigations are outlined. Measurements of both the total amount of CO2 and its 13C enrichment are performed in an isotope-ratio mass spectrometer which provides acceptable levels of accuracy and reproducibility for both measurements (+/- 0.1% and +/- 0.0001 at % excess respectively).  相似文献
