Background
Bone marrow stromal cell antigen 2 (BST-2) is a cellular factor that restricts the egress of viruses such as human immunodeficiency virus (HIV-1) from the surface of infected cells, preventing infection of new cells. BST-2 is variably expressed in most cell types, and its expression is enhanced by cytokines such as type I interferon alpha (IFN-??). In this present study, we used the beta-retrovirus, mouse mammary tumor virus (MMTV) as a model to examine the role of mouse BST-2 in host infection in vivo.Results
By using RNA interference, we show that loss of BST-2 enhances MMTV replication in cultured mammary tumor cells and in vivo. In cultured cells, BST-2 inhibits virus accumulation in the culture medium, and co-localizes at the cell surface with virus structural proteins. Furthermore, both scanning electron micrograph (SEM) and transmission electron micrograph (TEM) show that MMTV accumulates on the surface of IFN??-stimulated cells.Conclusions
Our data provide evidence that BST-2 restricts MMTV release from naturally infected cells and that BST-2 is an antiviral factor in vivo. 相似文献92.

Populations of feral swine (Sus scrofa) are estimated to include >2 million animals in the state of Texas, USA, alone. Feral swine damage to property, crops, and livestock exceeds $50 million annually. These figures do not include the increased risks and costs associated with the potential for feral swine to spread disease to domestic livestock. Thus, effective bio-security measures will be needed to quickly isolate affected feral swine populations during disease outbreaks. We evaluated enclosures built of 0.86-m-tall traditional hog panels for containing feral swine during 35 trials, each involving 6 recently caught animals exposed to increasing levels of motivation. During trials, fences were 97% successful when enclosures were entered by humans for maintenance purposes; 83% effective when pursued by walking humans discharging paintball projectors; and in limited testing, 100% successful when pursued and removed by gunners in a helicopter. In addition to being effective in containing feral swine, enclosures constructed of hog panels required simple hand tools, took <5 min/m to erect, and were inexpensive ($5.73/m excluding labor) relative to other fencing options. As such, hog-panel fences are suitable for use by state and federal agencies for rapid deployment in disease response situations, but also exhibit utility for general control of other types of damage associated with feral swine. © 2011 The Wildlife Society. 相似文献

We report on the construction of sex-specific high-density linkage maps and identification of sex-linked markers for the black tiger shrimp (Penaeus monodon). Overall, we identified 44 male and 43 female linkage groups (2n = 88) from the analysis of 2,306 AFLP markers segregating in three full-sib families, covering 2,378 and 2,362 cM, respectively. Twenty-one putatively homologous linkage groups, including the sex-linkage groups, were identified between the female and male linkage maps. Six sex-linked AFLP marker alleles were inherited from female parents in the three families, suggesting that the P. monodon adopts a WZ-ZZ sex-determining system. Two sex-linked AFLP markers, one of which we converted into an allele-specific assay, confirmed their association with sex in a panel of 52 genetically unrelated animals. 相似文献

We present a new convenient method for quantitative three-dimensionally resolved diffusion measurements based on the photobleaching (FRAP) or photoactivation (FRAPa) of a disk-shaped area by the scanning laser beam of a multiphoton microscope. Contrary to previously reported spot-photobleaching protocols, this method has the advantage of full scalability of the size of the photobleached area and thus the range of diffusion coefficients, which can be measured conveniently. The method is compatible with low as well as high numerical aperture objective lenses, allowing us to perform quantitative diffusion measurements in three-dimensional extended samples as well as in very small volumes, such as cell nuclei. Furthermore, by photobleaching/photoactivating a large area, diffusion along the optical axis can be measured separately, which is convenient when studying anisotropic diffusion. First, we show the rigorous mathematical derivation of the model, leading to a closed-form formula describing the fluorescence recovery/redistribution phase. Next, the ability of the multiphoton FRAP method to correctly measure absolute diffusion coefficients is tested thoroughly on many test solutions of FITC-dextrans covering a wide range of diffusion coefficients. The same is done for the FRAPa method on a series of photoactivatable green fluorescent protein solutions with different viscosities. Finally, we apply the method to photoactivatable green fluorescent protein diffusing freely in the nucleus of living NIH-3T3 mouse embryo fibroblasts. 相似文献

Intraguild predation (IGP) betweenthe pentatomid Podisus maculiventris(Say) and the coccinellid Harmoniaaxyridis (Pallas) in the absence or presenceof the extraguild prey Spodopteralittoralis (Boisduval) and Myzuspersicae (Sulzer) was studied in thelaboratory. Interactions were asymmetric infavor of the pentatomid. Podisusmaculiventris readily fed upon eggs and larvaeof H. axyridis, but adult beetles wererarely attacked. Success of attacks by P.maculiventris was stage dependent, fourthinstars and adults being more successful inkilling ladybeetle larvae than second instars.Attacks by H. axyridis on the pentatomidwere rare and none of them were successful. Theeffect of introducing extraguild prey on thelevel of IGP was tested both in petri dishesand on caged sweet pepper plants. Whensufficient numbers of S. littoralislarvae were present to satiate the pentatomid,predation on H. axyridis larvae decreasedsignificantly, indicating that the coccinellidis a less preferred or less vulnerable prey.When the aphid M. persicae was presentedas extraguild prey, levels of IGP were notaltered. Nymphs of P. maculiventrissuccessfully completed development whenexclusively fed on larvae of H. axyridis,but developmental time was longer than onlepidopteran prey. No pentatomid nymphs reachedadulthood on aphids alone. IGP by P.maculiventris on H. axyridis may be ofsome importance in greenhouse crops, where bothpredators are being used increasingly inaugmentative biological control programs.Nonetheless, it is expected that in practicelarger larvae and adults of H. axyridiswill escape most attacks by the pentatomid. 相似文献
